Opis:
Successful in vitro propagation through ex vitro rooting mechanism has been achieved in Piper longum by nodal shoot segment cultures. Shoot tip explants were less proliferative compared to the nodal meristems. The explants were sterilized using 0.1% HgCl2 and cultured on Murashige and Skoog (MS) medium with various concentrations of growth regulators. MS medium supplemented with 1.0 mg/L 6-benzylaminopurine (BAP) was found suitable for bud breaking within four weeks. Kinetin (Kin) was not reported impressive compared to BAP in culture induction response. The shoots were multiplied and elongated on MS medium supplemented with 0.5 mg/L each of BAP and Kin and 0.1 mg/L indole-3 acetic acid (IAA), where the shoots were elongated up to 5.8 cm length. Rooting and acclimatization was achieved by ex vitro rooting methods using 300 mg/L indole-3 butyric acid (IBA) for 5 min, and 5.3 roots with 3.2 cm average length were observed. The rooted shoots were transferred to the greenhouse for acclimatization. The hardened plantlets were transferred to earthen pots and maintained in the greenhouse. Normal flowering was observed in micropropagated plants of P. longum.

Opis:
In vitro flowering was successfully induced from the shoots regenerated in cultures of Aerva lanata. In vitro flowering method can be monitored effectively in studying floral transition and flower development from vegetative to reproductive phase. The nodal shoot segments were used as explants in the present study. The explants were surface sterilized using HgCl2 and inoculated on Murashige and Skoog’s (MS) medium supplemented with 6-benzylaminopurine (BAP) for bud break. Maximum numbers of shoots were induced on MS medium supplemented with 1.0 mg l-1 BAP. The cultures were multiplied by continuous subculturing of shoots with mother explants on fresh MS medium with BAP and Kinetin (Kin). Rooting was achieved on one fourth MS medium supplemented with 2.0 mg l-1 indole-3-butyric acid (IBA). The multiplied shoots were subjected to various concentrations of different plant growth regulators and photoperiod regimes to induce flowering in vitro. The maximum numbers of in vitro flowers were recorded from the shoots on full strength MS medium with 0.5 mg l-1 BAP and 0.1 mg l-1 indole-3-acetic acid (IAA) on 12/12 h/d photoperiod.

Opis:
In vitro propagation methods using nodal segments of young tissues of Hibiscus subdariffa plant have been established. The nodal shoot segments responded better than apical shoot tip explants. Murashige and Skoog (MS) semi-solid medium supplemented with 1.0 mg/L 6-benzylaminopurine (BAP) induced 4.2 ±0.24 shoots per node with 2.40 ±0.18 cm in length after four weeks of inoculation. About 92% explants responded on this medium combination. The shoots were further multiplied (8.2 ±1.37 shoots with 5.9 ±1.05 cm length) by repeated subculture of mother explants and freshly induced shoot clumps on MS medium with the reduced concentrations of BAP and Kinetin (0.5 mg/L each) with additives within 4 weeks. Half strength MS medium augmented with 2.0 mg/L indole-3 butyric acid (IBA) was found most favorable for root induction with 98% response. On this medium 7.5 ±0.61 roots with 4.8 ±0.23 cm length of roots were recorded. The regenerated plantlets were successfully hardened and field transferred.

Opis:
The synthesis of Zinc oxide (ZnO) nanoparticles using plant parts is always an attractive and eco-friendly method. The present study focuses on the production of ZnO nanoparticles from Zinc Nitrate hexahydrate (Zn (NO3)2•6H2O) using the aqueous extracts of a medicinal plant Micrococca mercurialis. The plant is found to be rich in primary and secondary metabolites such as proteins, steroids and alkaloids, which are responsible and enhancing the biogenic synthesis of ZnO nanoparticles. Leaves, stems, roots and fruits of M. mercurialis were used to prepare the aqueous extracts. The characterization and confirmation of synthesized ZnO nanoparticles was performed by UV-Visible spectrophotometer. The reaction mixtures showed significant and sharp UV absorbance peaks at 305 nm, 299 nm, 311 nm and 302 nm corresponding to the aqueous extracts of leaf, stem, root and fruits reacted with zinc nitrate respectively. It has been proved that the medicinal plant M. mercurialis can be used to synthesize ZnO nanoparticles and the biological method of synthesis of ZnO nanoparticles are safe and good alternative to the chemical and physical methods.

The present communication reports the biosynthesis of zinc oxide nanoparticles using aqueous extracts of leaves, stem, root, flowers and fruits of Duranta erecta L. This plant gains horticultural and medicinal importance due to its various biological activities. Zinc Nitrate Hexahydrate [Zn (NO3)2•6H2O] solution was used as a precursor to synthesize Zinc oxide nanoparticles. The various plant (leaves, stem, roots, flowers, fruits) extracts played as reducing agents. The leaf extract showed strong absorbance peak at 302 nm, stem and flower peaks were located at 299 nm, roots at 293 nm and fruit extract solution peak observed at 317 nm.

Opis:
A method for vegetative propagation of Moringa concanensis was developed in the present study. Stem cuttings were collected from wild populations in Tamil Nadu. The cuttings with 2-4 nodes were treated with various concentrations of Indole Butyric acid (IBA), Indole Acetic Acid (IAA) and α-Naphthalene Acetic Acid (NAA) for 5 min. The effectiveness of auxins on shoot bud induction and rooting response of stem cuttings was in the order of IBA>NAA>IAA. The auxin treated cuttings were planted on planting media and placed in the greenhouse. The survival rate and relative growth rate of stem cuttings were analyzed at every 4 week interval for 24 week. Stem cuttings treated with 250 mg/L IBA was responded better and survived for two years. Indole-3 acetic acid treated stem cuttings initiated branches within 7 weeks but very few branches arose. The cuttings treated with NAA responded with less relative growth than IBA. This method proves the rooting ability of IBA and growth performance of stem cuttings, which can be used for the mass propagation and conservation of this rare medicinal plant to prevent the exploitation from the wild.
