Opis:
Mycological analyses of infected mountain pine needles were conducted for years 2011−2012. Yellow spots on needles were found in subalpine zone of the Karkonosze National Park. The isolation of fungi was conducted at the end of July in both years when the intensity of disease symptoms on needles was the highest. Infected needles were sampled from Kopa, Śląskie Kamienie, Kocioł Smogorni, Sokolnik, Łabski Szczyt, Szrenica, Kocioł Małego i Wielkiego Stawu study sites. The dominant species isolated from the infected needles was a probably new species of the genus Lophodermium. Except for this pathogen, Lophodermium pinastri, Botrytis cinerea and Epicoccum niger were identified together with several saprotrophic species, e.g. Aspergillus niger.

Carbon materials, bamboo (Bambusa vulagris) and yucca (Yucca flaccida) pyrolysed at 550°C, 750°C and 950°C, were tested as oximetric probes by electron paramagnetic resonance spectroscopy at X-band (9.3 GHz). The following parameters of the spectra: amplitude, linewidth and g-factor, were determined. Influence of oxygen molecules 02 on EPR spectra of the individual carbon materials was compared. Strong EPR spectra were recorded for samples carbonized at 550°C and weak signals were obtained for plants carbonized at higher temperatures: 750°C and 950°C. It was stated that amplitudes of EPR lines of all the carbonized plants decrease in the air environment compared to amplitudes of spectra measured in vacuum. This effect increases with degree of vacuum. Changes in the EPR spectra of samples studied in the air environment may be applied in medicine to determination of oxygen content in different cells. Because of strong resonance signals as oximetric probes we proposed bamboo and yucca carbonized at 550°C, and we rejected plants carbonized at 750°C and 950°C with Iow EPR signals.

Electron paramagnetic resonance spectroscopy (EPR) was used to examination of spin-lattice relaxation in vascular plants carbonized at 650°C. Application of EPR method of continuous microwave signal saturation in medicine was proposed. The first derivative EPR spectra of pyrolysed bamboo and yucca were measured for samples in air and in vacuum. Influence of microwave power in the range 0.7-100 mW on amplitudes of EPR lines were evaluated. It was stated that amplitudes of the studied carbon materials increase with increasing of microwave power in the studied range. Saturation of EPR lines was not observed, so fast spin-lattice relaxation processes exist in the analyzed materials. Changes of EPR amplitudes for samples in air were slower than for evacuated samples. This effect may be used for determination of oxygen contents in the biological cells cultures and for analysis of optimal parameters of photodynamic therapy of cancer.

The RP-TLC method was used to determine the dissociation constant of chrysin (5,7-dihydroxyflavone) in methanol-aqueous (1:1 v/v) solutions. In this method the pK value was quantified on the basis of retention data and a retention model. The accuracy of determining the model parameters was analysed using the following statistical criteria: the sum of the squared differences between the experimental and theoretical data, approximation of standard deviation, and the Fisher test. Besides, in this work the potentiometric method was used. Investigations were carried out at ionic strength I=0.1 at T= 298 K. The dissociation constant were determined using the Rossotti method as well as the numerical method based on the procedures of non-linear curve fitting using Microsoft Excel Solver and the user-defined function. It has been found that the differences in the evaluated pK values were relatively small and did not exceed 1.2%.

During cold periods in the Pleistocene Epoch, many plants known as the “relict species” migrated and inhabited new areas. Together with plants, some microfungi also migrated, remaining present on plants and in plant communities. However, the relationship between fungi and the migrating plants (especially host plants) is not well understood. Therefore, we examined the diversity and distribution of microfungi associated with two migratory relict plants in the Sudetes and Carpathian Mountains: Salix herbacea L. and Juncus trifidus L. In total, we found 17 taxa of fungi that were collected from nine different locations. Nine fungal taxa were collected on S. herbacea, and eight taxa on J. trifidus. Localities richest of fungi on S. herbacea were Mały Śnieżny Kocioł (Karkonosze Mts, Sudetes) and on J. trifidus, the Tatra Mts (Carpathian Mts). This work provides new insights into the distribution of fungi inhabiting S. herbacea and J. trifidus in Poland.

Mountain pine (Pinus mugo) is the most important component of the sub−alpine and alpine zone in the Tatras, and other ranges of central Europe. In Poland, only population from the Karkonosze Mts. has been studied in terms of phytopathological threats so far. There is not much information on the diseases caused by fungi in the mountain pine from other regions either. The mycological analyses of infected mountain pine needles were conducted for years 2013 and 2014 in subalpine zone of the Tatra Mts (Morskie Oko, Hala Gąsienicowa, Hala Kondratowa). Pine needles rust Coleosporium tussilaginis and yellow spots Lophodermium conigenum were found. To a lesser extent the needles were spotted with symptoms caused by Mycosphaerella dearnessi. Infection symptoms and fraction of the infected needles on one shoot were determined. The isolation of fungi was conducted in end of July, when the intensity of disease symptoms on needles was the highest. The main species isolated from the infected needles was Lophodermium spp., while the dominant one was L. conigenum. Except for this pathogen, L. pinastri, Botrytis cinerea and Epicoccum niger were identified together with several saprotrophic species, e.g. Aspergillus niger. The most frequent disease of mountain pine needles in the Tatra Mts. is pine needles rust. Observations in the Karokonosze and the Tatra Mts. indicate that both populations suffer from similar diseases. The differences show that pathogens adapt to different environmental conditions. Therefore constant monitoring of the health status of mountain pine growing in nature seems to be necessary.

Pinus × rhaetica is holoarctic species, which represents mixed morphological features from both P. uncinata and P. mugo. It is a typical species living in peat−bogs in highlands and it is endangered by drainage and the decline of its natural area. By now, the greatest population of peat−bog pine is living in the Wielkie Torfowisko Batorowskie peat−bog, but its health condition is alarming. In Poland P. × rhaetica is placed on the red list of plants and fungi as an endangered species, with a high risk of extinction in the wild. In order to preserve the genetic potential of the population, the national park has taken active protective measures, establishing a conservative nursery. The aim of this study was to determine the potential role of fungi growing on needles of peat−bog pine in health condition. The study was carried out in three conservative pine nurseries in the Góry Stołowe National Park (SW Poland). Needles and dead wood of peat−bog pine were collected in July 2016. Material was decontaminated in 1% surfactant solution of sodium hypochlorite. The next step was the liner of about 0.5 cm pieces of needles of 6 Petri dishes with solidified, acidified PDA medium (agar−potato glucose). The dominant microscopic fungi inhabiting needles are Lophodermium pinastri, isolated from all of the plots, and Truncatella hartigii, which was observed only on Wielkie Torfowisko Batorowskie peat−bog (tab. 1). A much greater diversity of species of fungi characterized that peat−bog (tab. 2).

Ustilago trichophora affects Echinochloa crus-galli and other species of Echinochloa genus. U. trichophora is a member of the order Ustilaginales, of which the germinating teliospores form a promycelium with sporidia. Investigations by Ingold (1996) proved that spores coming from various parts of the world germinate in a different way. The aim of the investigation was to determine the mode of germination of U. trichophora spores found in Poland.

Pojęcie „speleomykologia” zostało użyte przez autorów w literaturze światowej po raz pierwszy w roku 2014. Oznacza ono całość badań naukowych mających na celu poznanie mykocenozy jaskiń i innych obiektów podziemnych (sztolnie, kopalnie, bunkry, tunele itp.). Speleomykologia obejmuje badania składu gatunkowego oraz liczebności zarodników i innych organów tworzących kolonie grzybów zasiedlających skały, wodę, powietrze, a także obecną w jaskiniach materię organiczną (Pusz i in., 2014). Celem wstępnych badań speleomykologicznych prowadzonych w odkrytej kopalni w Srebrnej Górze była ocena zanieczyszczenia mikrobiologicznego powietrza – zarówno przez grzyby i ich zarodniki oraz inne mikroorganizmy.
Speleomycology is a new scientific branch of mycology concentrating on research of fungi present in caves and other underground objects such as adits, mines, bunkers, tunnels, etc. The aim of this preliminary study was to examine a newly discovered mine in Srebrna Góra. Initial aeromycological analysis has been conducted. Also, presence of entomopathogenic fungi was surveyed in terms of colonies count and their biodiversity. Both aeromycological analysis and survey concerning entomopathogenic fungi showed that cosmopolitan and non-specific fungi species were present in the underground object. It is worth noting that many of the found fungi might be present in the hypogean environment due to being brought there by the explorators.

Cryphonectria parasitica is a pathogen that causes chestnut blight – a disease that has decimated chestnut trees Castanea dentata in North America and C. sativa in Europe. C. parasitica also infects other tree species, including oaks. Although the disease on oaks progresses and develops in a milder way, it can pose a threat to them. The main purpose of the work is to investigate the risk of C. parasitica occurring in oak stands. The work is based on a review of the available literature taking into account an analysis of pathogen detection and identification methods, current distribution of disease in Europe, distribution of potential host plants, favourable climatic conditions, and ways of spreading (pathways).

The aim of this research was to determine the composition of microscopic fungi colonizing Norway spruce dead wood depending on the degree of decomposition of wood and the location in the Tatra National Park (TPN; southern Poland). The work included lying and standing dead wood of Norway spruce as well as tree stumps. The sampling plots were located in the Białka Valley (eastern part of the TPN) where the forests are unmanaged and area is mostly under strict pro− tection, and in the western part of the Park, in the Chochołowska Valley, where the majority of forests is privately owned and managed (landscape protection). Samples were collected three times in the growing season, in spring, summer and autumn 2016. The sampling plots were located in the forests covered by different forms of protection: strict, active and landscape. The dominant microscopic fungi inhabiting analysed Norway spruce dead wood in selected regions of the Tatras are fungi belonging to Trichoderma genus: T. harzianum: T. polysporum, and T. hamatum. A much greater diversity of fungi species isolated from the dead wood was found for the Białka Valley, subjected to strict and active protection, than Chochołowska Valley, where forests are subject of the economic utilisation. More species and colonies were obtained from lying and older dead wood than from the stumps in the higher classes of distribution.

Opis:
Swedish whitebeam (Sorbus intermedia (Ehrh.) Pers.) is a species with a very limited range. It occurs only in southern Scandinavia and along the Baltic coast, without the Gulf of Finland. Its southernmost natural localities are situated in northern Poland. They are few in number, grouped in two regions: near Kołobrzeg and the Gulf of Gdańsk. Currently, only three of them survive: vicinity of Sarbinowo as well as Kępa Redłowska and Przylądek Rożewski. In the Podtatrze region, S. intermedia has long been planted along roads, on green areas, in gardens and parks, among others in Zakopane. In the Tatras it did not appear, so it is a foreign species for this region. Currently, in the Tatra National Park there are several hundred fruiting specimens of this species growing. Most of them are part of a forty−year−old stand planted on the back of the Grzybowiec ridge. The aim of this research was to recognise the health status of S. intermedia. Phytopathological research was conducted in 2018 in the Tatra National Park. Leaves that were subject to analysis were decontaminated in 1% surfactant solution of sodium hypochlorite. Their fragments of about 0.5 cm were placed on Petri plates with PDA (Potato Dextrose Agar). The growing fungal colonies were consequently re−planted onto PDA slants for further culturing and taxonomic identification. The pathogens were identified basing on morphological features and with molecular tools. All S. intermedia in the Tatra National Park showed disease symptoms. Infected S. intermedia leaves were colonized by A. alternata, which fraction amounted to over 40%. The following dominant was B. exigua with over 30% share. F. avenaceum (close to 10%) and E. nigrum and D. macrostoma were also found. The S. intermedia population growing in the Tatra National Park is characterized by a poor condition, which may in the future affect the population of this plant in the Tatra Mts.
