Opis:
Biotransformation of deuterated-4'-O-methylnorbelladine into alkaloids galanthamine and lycorine in tissue cultures of Leucojum aestivum was demonstrated using HPLC coupled to mass spectrometry. GC-MS screening was also carried to investigate other native and deuterated alkaloids. A total of six labeled alkaloids were identified indicating that 4'-O-methyl-d3-norbelladine is incorporated into three different groups of Amaryllidaceae alkaloids that are biosynthesized by three modes of intramolecular oxidative phenol coupling.

Selenium compounds have been implicated as anticancer agents; however, the mechanism of their inhibitory action against cancer development has not been extensively investigated. A constitutive activation of the Wnt/β-catenin pathway is a central event in colorectal carcinogenesis. In this pathway, excessive cell proliferation is initiated by generation of β-catenin followed by overexpression of proto-oncogenes, such as c-Myc. It is believed that under physiological conditions the level of c-Myc is efficiently controlled by accessibility of the β-catenin protein through the process of phosphorylation by glycogen synthase kinase 3β (GSK-3β). Here, we determined whether selenomethionine (SeMet) can inhibit cell growth and affect the Wnt/β-catenin pathway in the HT-29 human colorectal cancer cells in vitro. The effective cytotoxic doses of SeMet have been selected after 48 h of incubation of this compound with colorectal cancer HT-29 cell line. MTT assay was used to assess cell viability and the protein and mRNA levels of β-catenin and c-Myc were determined by Western blotting and qPCR, respectively. SeMet potently inhibited growth of HT-29 cells, significantly decreased level of the β-catenin protein and mRNA concentration, down-regulated the c-Myc gene expression and up-regulated the pro-apoptotic Bax protein level. Moreover, SeMet increased the level of GSK-3β phosphorylated at serine 9 (S9) and significantly increased the level of β-catenin phosphorylated at S33 and S37. We conclude that SeMet suppresses growth of HT-29 colorectal cancer cells by a mechanism linked to the Wnt/β-catenin pathway, however, degradation of β-catenin may occur independently of GSK-3β catalytic activity and its phosphorylation status.

The aim of this paper is to present the development of general government debt in two Eurozone countries: Ireland and Spain that suffered from serious imbalance in public finance during the last crisis. Prior to the crisis, both economies were developing well against the background of the whole Eurozone and had a relatively good situation in public finance. The genesis of the crisis was also quite similar in these two countries. The similarity of factors influencing the crisis and the pre-crisis high development of both economies were among the reasons for selection of these two countries to be compared. Thus, the article focuses on the outbreak of the crisis and the fiscal consolidation period of 2008-2015, however the pre-crisis analysis is also provided. The debt sustainability analysis carried out in the article shows the possibility of growing out of debt in both countries depending on the macroeconomic circumstances. Both Ireland and Spain have been aiming to achieve a primary surplus. Besides the similarity of pre-crisis conditions, in this respect, the progress was highly noticeable, especially in Ireland, where it resulted from a fiscal consolidation but also a high real GDP dynamics that supported the process. Due to this, Ireland has already managed to lower the debt-to- -GDP ratio and put it on a downward path. Spain, on the contrary, has recorded a high debt-to-GDP ratio which is still on the upward path and is forecasted to continue until 2016.

Przedmiotem analiz są wymiary patriotyzmu praktykowane w życiu przez Halinę Semenowicz (1910-2004) – prekursorkę ruchu freinetowskiego w Polsce – w świetle jej dzienników z lat: 1979, 1982, 1989, 1992, 1999, 2001. W pierwszej kolejności zastosowano analizę frekwencyjną w poszukiwaniu pojęć będących rozwinięciem pojęcia obywatela, by na tej podstawie ocenić nasycenie narracji pojęciami związanymi z patriotyzmem. Następnie dokonano analizy danych ze względu na poznawcze, afektywne i behawioralne komponenty patriotyzmu. Na tej podstawie można stwierdzić, że Halina Semenowicz reprezentuje dojrzały patriotyzm z nutą romantyzmu, który wyrastał z dojrzałej i zintegrowanej osobowości, postawy otwartości na uczenie się przez całe życie. Halina Semenowicz może być w szczególności przykładem dla innych praktykowania patriotyzmu opartego na pluralizmie wartości i różnorodności dróg bycia Polakiem.
The subject of the analyses are the dimensions of the patriotism practiced during her lifetime by Halina Semenowicz (1910-2004) – the precursor of the Freinet movement in Poland – in the light of her journals from years 1979, 1982, 1989, 1992, 1999, 2001. First, the turnout analysis was conducted to search for the terms which would be an expansion of the word citizen and on the basis of that, to assess the saturation of the narration with the words connected to patriotism. Then the data was analyzed when it comes to the cognitive, affective, and behavioural components of patriotism. On the basis of the analyses it can be stated that Halina Semenowicz represents mature patriotism with a bit of romanticism which grew from the mature and integrated personality, the attitude of openness to lifelong learning. Halina Semenowicz can be in particular an example for others of practicing patriotism based on the pluralism of values and the variety of the ways of being
