Introduction. Reactivation of BKV infection caused by decrease of immunity associated with organ transplantation can lead to nephropathy and organ loss. Antiviral drugs such as ganciclovir or foscarnet did not display any activity against BK virus. A source of hope can be found in chemotherapeutics usually used in bacterial infections that inhibit the replication of viral genetic material in cells. One of the aforementioned chemotherapeutics is ciprofloxacin. The aim of the study is to evaluate the antiviral activity of ciprofloxacin against BK virus in vitro culture. Materials and method. Initially, the effects of ciprofloxacin on Vero cell viability were assessed with MTT. After determining the 3 largest non-cytotoxic concentrations of antibiotics, their effect on viral replication in cells was determined. 24-h cells culture in DMEM medium supplemented with 10% serum after 2h incubation with a 6.3 x 105 copies/ml infection virus were used to evaluate the effects of different concentrations of ciprofloxacin on the number of BKV copies. Quantification of BKV replication in the culture was performed by real-time PCR. Results. By means of the MTT test, the 3 largest non-cytotoxic ciprofloxacin concentrations were determined as follows: 31.25; 62.5; 125 μg/ml. Cells treated with ciprofloxacin at a concentration of 125 μg/ml display the greatest decrease in BKV replication that equaly 1.8 – 3 log/5.8 – 9.2 Ct, compared to the control of the virus without drugs. A decline in the number of copies of the virus was observed during the lower concentration of the antibiotic (62.5, 31.25 μg/ml) by 1.3 – 2 log/4 – 6.4 Ct and 0 – 0.8 log/0 – 2, 4 Ct, respectively. Conclusions. Ciprofloxacin exhibits antiviral activity against the BK virus by inhibiting its replication in the in vitro culture in Vero cell line.

Introduction and objective. Matrix metalloproteinases (MMPs) are proteolytic enzymes responsible for the decomposition of extracellular matrix elements. They play an important role during embryogenesis, wound healing, endometrial epithelial exfoliation, the formation of new blood vessels, and also during cancer development. Throat and larynx tumours are included in a large group of head and neck cancers. These tumours are characterized by a poor prognosis. Despite advances in medical science, cancer treatment is difficult and often ineffective. The aim of the study was to evaluate the level of MMP-3 and MMP-9 in the serum and saliva of patients with pharynx and larynx tumours. Materials and method. Samples of saliva and serum were collected from 60 patients with larynx or throat cancer. Twenty patients without cancer comprised the control group. MMPs in saliva and serum were determined by the ELISA method. Results. In the study group, concentrations of MMP-3 in saliva were from 0.2 – 77.6 ng/ml. Patients with malignant tumours had higher saliva MMP-3 levels than healthy subjects. The concentration of MMP-3 in the serum of the study group ranged from 10.9 – 200.00 ng/ml, which was also higher than in the control group. There were no statistically significant difference in the MMP-9 level between the study and control groups (both in serum and saliva). Conclusions. This study is another element that shows the phenomena taking place at the cellular level during oncological disease. In serum and saliva samples, higher values of MMP3 were found in patients with cancer. The increase in the concentration of this enzyme in the risk group may be used for early detection of tumour transformation and evaluation of treatment.

The Epstein-Barr virus (EBV) was discovered in 1964 by Michael Anthony Epstein and Yvonne Barr, who discovered a herpesvirus-like infectious agent in a biopsy specimen from a patient with Burkitt’s lymphoma. This virus belongs to the Herpesviridae family (subfamily Gammaherpesvirinae, genus Lymphocryptovirus). EBV is a ubiquitous herpesvirus that is causally associated with various malignant tumours. According to the current nomenclature, it was named human herpesvirus type 4 (human herpesvirus 4 – HHV-4). Primary infection usually occurs in childhood. In developing countries, the infection rate among young children is higher than in developed countries. It was the first human tumour virus and it is currently categorized as a group-1 carcinogen due to its association with various cancers. It is estimated that over 90% of the adult population has been infected with this pathogen, but only a minority will develop the disease. EBV establishes latent infection characterized by the expression of a limited number of viral genes called latent genes. Moreover, during its life cycle, EBV periodically reactivates and can be transmitted to other susceptible hosts. The oral cavity is the main site of EBV occurrence and the most common source of infection. This study discusses EBV frequency and its association with the occurrence of malignant tumours and the pathways of tumour progression.

Introduction and objective. Colorectal cancer is one of the most common cancers worldwide. In Poland, it is the second most common cancer, regardless of gender. The aim of study was to analyze the incidence of HPV and BKV in the tissue of colorectal cancer and to determine the relationship between the presence of these viruses and the development of this cancer. Materials and method. The experiments were conducted using 50 colorectal cancer tissues collected from histological sections. The clinical material was embedded in paraffin blocks. Next, DNA extraction was performed. Isolates of colorectal cancer tissue were tested for the presence of HPV DNA. BKV DNA was detected by PCR using specific primers and then differentiated from JCV by digestion with BamHI enzyme. Results. In clinical specimens taken from patients with colorectal cancer, HPV DNA was detected in 20% of cases. In 10% of cases the presence of HPV type 18 was confirmed, in the other 90% of the samples HPV type 16 was detected, while the presence of BKV was confirmed in 30% of cases. Coinfection with HPV and BKV was shown in 12% of patients. In one case, BK virus coexisted with HPV type 18, in the remaining 5 cases with HPV type 16. Conclusions. Developing colorectal cancer can show no symptoms, even for many years. This is why it is so important to become familiar with as many etiological factors as possible. The development of many human neoplasms is often initiated by exposure to infectious agents – such as bacterial or viral infections. Similar to the human papillomavirus, the BK virus was detected in clinical specimens. It seems that HPV and BKV infections can contribute to the neoplastic process, which requires detailed studies on a larger group of patients.

Introduction and objective. Lithium is used in medicine but its application may cause diverse side effects. Selenium has been found to show protective properties against negative influence of different harmful factors. This study was aimed at evaluating the influence of non-toxic dose of lithium on antioxidant parameters in FaDu (ATCC HTB-43) and Vero (ECACC No. 84113001) cell lines as well as the possible protective effect of non-toxic concentration of sodium selenite. Materials and method.The cells were subjected to 0.17 mmol/L of Li2CO3 and/or 2.9 μmol/L of Na2SeO3 · 5H2O for Vero as well as 0.47 mmol/L of Li2CO3 and/or 3.0 μmol/L of Na2SeO3 · 5H2O for FaDu cells. The incubation was continued for the subsequent 72 h. In the cells total antioxidant status (TAS) values, activities of antioxidant enzymes – superoxide dismutase (SOD) and glutathione peroxidase (GPx) as well as the reduced glutathione concentration (GSH) were determined. Results and conclusion. In Vero cells lithium decreased all studied parameters, particularly GPx. Selenium co-treatment showed a distinct protective effect. In FaDu cells the similar effect was observed only in case of GSH. The results point to differences in action of lithium and selenium in physiological and pathological state. As long-term lithium therapy is applied in psychiatric patients the results regarding Vero line let suggest that selenium might be considered as an adjuvant alleviating side effects of Li-treatment.

Wprowadzenie. W czasie ciąży kobiety są szczególnie narażone na zakażenia drobnoustrojami, które potencjalnie nie zagrażają życiu, ale mogą być bardzo niebezpieczne dla rozwijającego się płodu. Kobieta może uniknąć zakażenia, jeżeli miała kontakt z patogenem w dzieciństwie lub gdy nabyła odporność w wyniku szczepień ochronnych. Do wirusów, które zagrażają ciężarnej, należą m.in. wirus cytomegalii oraz wirus różyczki. Patogeny te mogą powodować zakażenia wrodzone, które są powodem szeregu wad wrodzonych lub nawet zatrzymują rozwój płodu. Dlatego tak ważna jest wczesna diagnostyka zakażeń drobnoustrojami u kobiet ciężarnych i zapobieganie im. Cel pracy. Celem pracy była ocena miana przeciwciał przeciwko wirusowi cytomegalii i różyczki u kobiet w wieku rozrodczym. Materiał i metody. W celu oceny miana przeciwciał klasy IgG przeciwko wirusowi cytomegalii oraz wirusowi różyczki u kobiet będących w wieku rozrodczym przebadano 42 próbki surowic krwi pobranych od kobiet w wieku 20–25 lat. Oznaczenie obecności oraz liczby specyficznych przeciwciał dokonano za pomocą metody ELISA, przy użyciu zestawów NovaLisaTM. Wyniki. Przeprowadzone badania wykazały obecność przeciwciał klasy IgG przeciwko wirusowi cytomegalii u 61,9% oraz przeciwciał klasy IgG przeciwko wirusowi różyczki u 97,6% badanych kobiet. Wnioski. 1. U ok. 40% badanych nie wykryto przeciwciał przeciwko wirusowi cytomegalii, co wskazuje na brak dotychczasowego kontaktu z CMV. 2. Wykazano wysoki stopień uodpornienia przeciwko wirusowi różyczki wśród kobiet w wieku rozrodczym.
Introduction. During the course of pregnancy, women are particularly prone to microbial infections that are not potentially life-threatening; however, they may pose a serious threat to developing foetus. A woman can prevent infection either by being in contact with the pathogen during childhood or by acquiring immunity through vaccination. The viruses that are threatening to pregnant women include, among others, cytomegalovirus and rubella virus. These pathogens may cause congenital infections that can lead to a number of birth defects or even stop the development of the foetus. Therefore, an early diagnosis and prevention is crucial as far as pregnant women are concerned. Objectives. The aim of the study was to evaluate cytomegalovirus and rubella antibody titers in women of childbearing potential. Material and methods. In order to assess the titer of IgG class antibodies to cytomegalovirus and rubella in women of childbearing age, 42 blood serum samples collected from women aged between 20–25 years were examined. Determination of the presence and quantity of the specific antibodies was performed by utilising the NovaLisaTM sets and ELISA method. Results. The study indicated the presence of IgG antibodies to cytomegalovirus in 61.9% of the examined women and IgG antibodies to rubella virus in 97.6% of those examined. Conclusions. 1. Approximately 40% of the examined had no antibody against cytomegalovirus, which indicates the absence of a previous contact with CMV. 2. A high degree of immunization against rubella was observed among women at child-bearing age.
