Opis:
For breeding purposes, the number of neo-tetraploids of apple cultivars have been derived by in vitro technique. The first attempts at rooting and acclimatization of tetraploid shoots failed. The aim of the study was to develop an effective method for rooting microcuttings of apple neo-tetraploids. In the first stage of the study, in vitro rooting method was optimized for the shoots of diploid donor cultivars. Shoots were rooted on Murashige and Skoog [1962] (MS) medium with a reduced content of nitrogen, in the presence of auxins alone or in combination (indole-3-butyric acid – IBA, 1-naphthaleneacetic acid – NAA and indole- -3-acetic acid – IAA) with addition of putrescine, arginine or ornithine. The compounds were applied continuously for 25 days (one-step rooting system) or for seven days with subsequent transplanting shoots onto a medium without these compounds (two-step rooting method). Tetraploid microcuttings of the cultivars ‘Free Redstar’, ‘Gala Must’, ‘Pinova’ and ‘Redchief’ were evaluated for rooting on the selected medium considered optimal for their diploid counterparts. The shoots of all diploid apple scion cultivars had low rooting capacity. IBA alone poorly stimulated root formation. Significant improvement of rooting to 60–80% was achieved through the application of auxins, 2.5 µM IBA or 1.3 µM NAA combined with 5 µM IAA and 50 µM putrescine in the two-step rooting system with darkness and increased temperature of 26°C during seven-day induction phase. The replacement of benzyladenine (BA) by meta-Topolin (m-T) in the last multiplication subculture influenced positively shoot acclimatization. Tetraploids had comparable or slightly lower rooting and acclimatization ability compared to their diploid counterparts.

Opis:
The effects of TDZ and paclobutrazol on the primary regeneration on tulip flower stalk explants of six cultivars and subsequent shoot multiplication were examined. Explants, flower stalk slices, were excised from cooled and subsequently forced bulbs. The explants were incubated for two months in darkness on medium containing NAA and cytokinins, 2iP and BAP, as control, or TDZ (0.5-4 mg l-1) and paclobutrazol (0.05-0.4 mg l-1). Then, the regenerating explants were subcultured on medium with TDZ and NAA applied at low concentrations. Different regeneration capabilities were found depending on cultivar and growth regulators. The percentage of explants forming leaf-like structures ranged, on the control medium, from 80% in 'Blue Parrot' and 'Prominence' to below 30% in 'Apeldoorn' and 'Mirjoran'. TDZ, applied at optimum for each cultivar concentration, greatly increased the regeneration potential up to 70-100%. Paclobutrazol, added to the TDZ-containing medium, significantly enhanced the response of explants, resulting in high numbers of leaf-like structures formed per explant (13.7-22.8). The structures developed gradually into characteristic forms: the growing up cotyledonary leaf, the probable root primordium formed at its base, the growing downwards stolon and the shoot meristem developed finely on its tip. It is suggested that such primary regeneration may have a nature of somatic embryogenesis. Then, the adventitious shoots developed and formed clusters, which were divided into 2-3 smaller ones every two months. The growth regulators, used at initial stage, markedly influenced subsequent shoot multiplication. Thus, the most intensive shoot formation was noted with TDZ at concentrations of 0.5-2 mg l-1 and paclobutrazol of 0.05-0.1 mg l-1.

The results of PCR examination indicated that during two years of tissue culture at standard conditions, on the medium with BAP 1 mg l-1 and continuous temperature of 20oC, phytoplasma could be detected in diseased plants of rose cv Sacha and Jazz. In the second year of micropropagation phytoplasma detection rate in tissues of infected roses increased and was relatively higher than in the first one. To test whether phytoplasmas are sensitive to temperature and light intensity, phytoplasma-affected micropropagated rose plants were grown on medium with BAP 1.0 or 0.5 mg l-1 and at the temperature of 4, 15, 20 or 25oC in darkness or in the light. PCR analysis indicated that phytoplasma detection was not effected by these conditions during 4 weeks of culturing. However, phytoplasma was not detectable in rose plants after 8 weeks culturing on the same medium without transplanting. Micropropagated rose shoots maintained on medium with Gentamycin or Baytril at the concentration of 25.0 or 50.0 mg l-1 had reduced growth and were chlorotic. However, no direct effect of applied antibiotics on phytoplasma detection was evidenced.

Apple species and cultivars differ in nuclear (2C) DNA content and ploidy level. The majority of these genotypes are diploids, but there are some triploids and a few tetraploids. Nuclear DNA content is a specific feature and its flow cytometric evaluation can be helpful in differentiating taxa. For many apple genotypes – including all the Polish ones, these characteristics are not known. 2C DNA was evaluated in relation to leaf, flower, fruit, pollen grain and stomata sizes as well as to the flowering time for seventy genotypes (including 46 Polish cultivars) gathered in the gene bank of the Research Institute of Horticulture, Skierniewice, Poland. For standard cultivars with the known chromosome number, 2C value was 1.71 pg for diploid cultivar ‘Alwa’ (2n=2x=34), 2.55 pg for triploid ‘Boskoop’ (3x=51), and 3.37 pg for tetraploid genome (4x=68) of mixoploid ‘McIntosh 2x+4x’. In 61 cultivars (including 41 Polish ones), the nuclear DNA content ranged from 1.58 to 1.78 pg indicating their diploid chromosome number. Five cultivars were identified as triploids (‘Bursztówka Polska’, ‘Pagacz’, ‘Rapa Zielona’, ‘Rarytas Śląski’ and ‘Witos’) owing to their nuclear DNA amount ranging between 2.42 and 2.58 pg. Leaf, flower, fruit, stomata and pollen grain sizes were on average significantly larger in triploids. Thus, in 3x plants the mean leaf surface was 49.1 cm², flower diameter – 52.4 mm, fruit weight – 204.7 g, stomata length – 32.1 μm and pollen grain diameter – 33.7 μm, whereas in diploids – 36.0 cm², 46.1 mm, 162.7 g, 28.4 μm and 30.7 μm, respectively. Pollen grain viability was on average significantly higher in diploids (75.6%), compared to triploids (22%). These results confirm that in apple, as in many other plant species, the higher ploidy level of triploids is generally associated with increased sizes of pollen grains, stomata, flowers, fruits and leaves but decreased pollen viability. No clear correlation between ploidy level and flowering time was found. In the case of mixoploid apple genotypes possessing diploid and tetraploid genomes, some phenotype observation is helpful in describing the ploidy level of the histogenic layers, L1 and L2. Small stomata sizes (similar to diploid) indicate diploid L1 and larger leaf sizes, compared to diploid counterparts, show tetraploid L2. The results will be used for breeding, in which it is important to determine maternal and paternal genotypes as well as the direction of the crossing that is of great importance in obtaining seeds and materials for further selection.
