Opis:
The present work has the objective of studying the effect of shot peening with glass microspheres on SAE 1020 steel in its resistance to fatigue. Fatigue tests were carried out by rotary bending with load control and loading on balance in specimens with and without shot peening. A rotation speed of approximately 750 rpm (12.5 Hz) was employed in the fatigue tests. Vickers microhardness tests were performed in order to verify the surface hardening produced by shot peening with glass microspheres. Analysis of the steel surface and fatigue fractures was performed using scanning electron microscopy (SEM). Fatigue tests were performed in order to obtain S-N curves (Wöhler curves). It was observed that shot peening with glass microspheres improved the fatigue strength of the steel at high cycle.

Opis:
In this paper we investigate different processes which lead to charge carrier generation in ITO/PEDOT:PSS/MEH-PPV/Al photodetector. Primary photogenerated singlet excitons contribute the photocurrent directly through dissociation on the electrodes. Singlet exciton distribution in the active area of the photodetector is calculated on the base of continuity equation including exciton generation, diffusion, and recombination. Depending on the applied electric field and incident photon flux secondary charge carrier generation processes appear in the active MEH-PPV film. Two bulk charge generation mechanisms are considered. Nonlinear recombination of excitons through exciton-exciton annihilation leads to electron and hole polaron generation. Exciton states are also depopulated by exciton dissociation in the presence of high electric field. We assume hole polarons to be majority charge carriers. Polaron contribution to photocurrent is calculated by solving the continuity and drift-diffusion equations for these carriers. Photocurrent spectra are measured for different bias voltages and different incident photon flux intensities. A good agreement between experimental and simulated data confirms our theoretical approach.

Opis:
The wide hybridisation has been applied to cereals breeding allowing to the production of new amphiploids. Triticale (X Triticosecale Wittmack) was the first synthetic amphiploid of the Triticeae tribe and performed well in comparison with its parent’s wheat and rye. In this work we aimed to analyse the meiotic behaviour and to identify the chromosomes present in both mitotic and meiotic metaphase cells of the portuguese cultivar ‘Douro’ (2n=42, AABBRR) using in situ hybridisation technique with genomic DNA from rye and repetitive DNA sequences as probes. Triticale ‘Douro’ presented a high level of homologous pairing. The mean number of bivalents was 20.14 and the average of ring bivalents was higher than rod bivalents. The total frequency of univalents was reduced (1.72), presenting the wheat univalents a higher frequency comparatively to the rye univalents. No multivalent configurations were seen, confirming the high meiotic regularity and stability of triticale ‘Douro’. The simultaneous use of pSc119.2 and pSc250 repetitive DNA probes on mitotic and meiotic chromosomes allowed the identification of all rye chromosomes. The pSc119.2 probe identified some wheat chromosomes, namely from B genome, and the chromosome pairs 4A and 5A.

Opis:
Introduction and objective. Recently, investigations in a swine herd identified evidence of the existence of a novel member of the Hepadnavirus family endemic in swine. The aim of this study was to investigate the serological and molecular markers of Hepadnavirus circulation in Brazilian domestic swine and wild boar herds, and to evaluate the identity with HBV and other Hepadnaviruses reported previously. Materials and methods. For the study, 376 swine were screened for hepatitis B virus serological markers. Analyses were performed in serum samples using commercial enzyme-linked immunosorbent assay (ELISA) kits (DiaSorin®) for anti-HBc, HBsAg and anti-HBs. Reactive and undetermined swine serum samples were selected to perform DNA viral extraction (QIAamp DNA Mini Kit, Qiagen®), partial genome amplification and genome sequencing. Results. From 376 swine samples analysed, 28 (7.45%) were reactive to anti-HBc, 3 (0.80%) to HBsAg and 6 (1.6%) to anti-HBs. Besides, more 17 (4.52%) swine samples analyzed were classified in the grey zone of the EIA test to anti-HBc and 2 (0.53%) to HBsAg. From 49 samples molecularly analyzed after serological trial, 4 samples showed a positive result for the qualitative PCR for Hepadnavirus. Phylogenetic reconstruction using partial genome sequencing (360 bp) of 3 samples showed similarity with HBV with 90.8–96.3% of identity. Conclusions. Serological and molecular data showed evidence of the circulation of a virus similar to hepatitis B virus in swine.
