Prostate cancer is one of the most commonly diagnosed cancers in men throughout the world. It is believed that changes to the structure of protein binding sites, altering its metabolism, may play an important role in carcinogenesis. Sulphur, often present in binding sites, can influence such changes through its chemical speciation. Hence there is a need for precise investigation of coordination environment of sulphur. X-ray absorption near edge structure spectroscopy offers such possibility. Cell culture samples offer histologically well defined areas of good homogeneity, suitable for successful and reliable X-ray absorption near edge structure analysis. This paper presents sulphur speciation data collected from three different human prostate cancer cell lines (PC-3, LNCaP and DU-145). Sulphur X-ray absorption near edge structure analysis was performed on K-edge structure. The spectra of cells were compared with those of cancerous tissue and with organic substances as well as inorganic compounds.

Expression of GA733-1 and GA733-2 genes in gastrointestinal tumours was investigated. Twelve cases of colorectal and stomach cancers were analysed. The Northern blot and RT-PCR methods were used. The second method appeared to be much more sensitive and useful in these investigations. No expression of GA733-1 gene was found in the investigated cases. GA733-2 expression was observed in 83% of colon cancers (five to six analysed cases) and in 33% of stomach cancers (two to six analysed cases). No correlation was found between the expression and clinical properties of tumours. Moreover, in two cases the presence of GA733-2 mRNA was observed in the surrounding morphologically normal tissue. These results indicate that GA733-2 gene can be a good marker of colon but not stomach cancers.

In order to assess the effect of alternating electromagnetic and direct magnetic field on adult Helix aspersa O. F. Müller and their eggs, adults and eggs of H. aspersa maxima Taylor and H. aspersa aspersa O. F. Müller were exposed to such fields with different parameters, and the following life cycle traits were observed: condition of hibernating adults, their survival rate during hibernation and reproduction, egg mass, hatching success, growth rate of hatchlings and body mass of next generation adults. Adults were exposed to sinusoidal, alternating electromagnetic fields of 50 Hz, 100 Hz and 200 μT, and magnetic induction of 50 Hz, as well as a field of 175 μT and 10–1000 Hz (repetition period 80 minutes). Egg batches were exposed to direct magnetic fields of magnetic induction 5 μT and 10 μT and alternating electromagnetic fields of analogous induction and frequency of 50 Hz. The effects varied depending on the parameters of the fields, subspecies and life cycle stage. Adult H. aspersa maxima was more sensitive to the changes in characteristics of electromagnetic field than H. aspersa aspersa, but the two subspecies showed a similar reaction to exposing their eggs to direct or alternating magnetic field. Probably direct field had a greater selection effect compared to alternating field, removing weaker genotypes at embryonic stage; this resulted in a greater mean body mass of the next generation adults. The results are preliminary; further studies, with further modifications of parameters of the fields applied are necessary.

The main challenge of this research was to functionalize the surface of poly(L-lactide-co-glycolide) (PLGA) membranes with amphiphilic poly(2-oxazoline) (POx) in order to change PLGA chemical state and properties. Poly(2-oxazolines) are very powerful polymers, which thanks to active pendant groups can be easily functionalized with biologically active molecules or peptides. The membranes were prepared by dissolving PLGA, POx, and poly(ethylene glycol) (PEG, 1000 Da) in methylene chloride (DCM), followed by PEG leaching. POx molecules were preferentially adsorbed at the interface PLGA-POx-PEG thanks to affinity to both hydrophilic (PEG) and hydrophobic (PLGA) chains. The properties of the membranes were characterized with Fourier transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM) and wettability tests. Cytocompatibility of the materials in contact with osteoblast-like MG-63 cells was studied by evaluation of cell viability (Alamar-Blue test), live/dead and phalloidin/DAPI staining. The results show that the presence of POx influenced topography of the PLGA membranes, but did not have an impact on their wettability. All membranes were fo-und cytocompatible with model osteoblasts. Presence of POx resulted in better cell adhesion as shown by microscopic studies after fluorescence staining for nuclei and cytoskeleton actin filaments. In summary, one-step phase separation process between PLGA, PEG, and POx, dissolved in DCM followed by drying and PEG leaching resulted in cytocompatible PLGA membranes with immobilised POx, which might be considered for guided tissue regeneration technique in periodontology and in bone tissue engineering.

Vibrational spectroscopy has been extensively used for in vitro and in vivo investigations of degradation mechanism and kinetics of different biomedical materials as well as it has been used to characterize the crystalline and amorphous domains in bio-mineralization process. Infrared and Raman spectroscopy methods are valuable tools in the biomaterials engineering allowing to study processes occurring during their preparation. In vitro tests, where the materials are immersed in simulated body fluids and/or artificial saliva, were used to evaluate the biocompatibility of biomaterials. This kind of tests are a wide range of repeatable and reproducible methods, which are regulated by international standards for commercial use and scientific development of new materials and products. The aim of this work was to examine phase composition of materials applied in dentistry. The bioactivity of such biomaterials was studied by immersing the samples in synthetic body fluid and artificial saliva. The changes were determined by the Fourier transform infrared and Raman microspectroscopy as well as scanning electron microscopy. It was found that results obtained by vibrational spectroscopy show the differences between the studied samples. Chemical reactions occurring during incubation of cements in artificial saliva as well as in synthetic body fluid result in formation of phosphates which deposit on the cement surface.

Carbon nanoforms due to their unique properties can be applied in many areas also in medicine. This article presents preliminary genotoxicity studies of electrospun carbon nanofibers (ECNF). This material, apart from its numerous applications, may also be a candidate for use in medical therapy and diagnostics. Polyacrylonitrile (PAN) nanofibers received in the electrospinning process were carbonized and thereafter subjected to oxidation treatment (ECNF-F). Both types of fibres were analyzed with regard to genotoxic influence on the fibroblast line cells using comet assay. Additionally, comet assay experiments were conducted on biocompatible carbon nanotubes with a hydrophilic surface. The results indicate the key role of the oxidation process in the functionalization of carbon nanoparticles intended for medical purposes.

The infrared spectromicroscopy is a quite recent technique rapidly developing thanks to the availability of new instruments and new brilliant synchrotron radiation sources in different areas and in particular to biomedical researches. In order to achieve a diffraction limited spatial resolution in tissue samples, we performed experiments at SINBAD, the synchrotron infrared beamline of the Laboratori Nazionali di Frascati. We characterized the chemical composition of prostate tissue samples taken from patients affected by prostate cancer disease. Different sizes of the pinholes were considered for the measurements. In the case of prostate tissue sections the results show the possibility to determine the intensity ratio of the $CH_2$ and $CH_3$ bands set at 2930 $cm^{-1}$ and 2960 $cm^{-1}$, respectively. Experiments were also performed with a pinhole of 5 μm of diameter and the differences in both histological and chemical compositions of such samples were determined.

The single cell gel electrophoresis method, known as comet assay, is a rapid and sensitive technique for testing novel chemicals and nanoparticles for genotoxicity, monitoring environmental contamination with genotoxins and human biomonitoring. In our studies we check the applicability of this method for the evaluation of biocompatibility of modified (MWNF) and non-modified multi-walled carbon nanotubes (MWNT) as well as potential genotoxicity of mercury(II) nitrate. The obtained results enabled us to conclude that the presence of Hg(NO₃)₂ (p<0.001) and MWNT (p<0.04) cause a significantly higher level of DNA damage in comparison to functionalised nanomaterials MWNF. It was implied that for the three investigated agents only mercury significantly enhanced genotoxic effect of X-ray exposure (p<0.001) and inhibition of radio-induced DNA damage repair. On the contrary, the presence of MWNF have no influence on cellular repair efficiencies, while incubation with MWNT causes apoptosis and consequently results in lack of attached cells. In conclusion, our results confirmed the genotoxicity of mercury and non-modified carbon nanotubes as well as the biocompatibility of modified nanotubes. Additionally, we proved the usefulness of comet method for the evaluation of genotoxicity and DNA repair under the influence of different compounds and nanomaterials.
