The coliform group has been used extensively as an indicator of water quality and has historically led to the public health protection concept. Total coliforms are a group of bacteria commonly found in the environment, for example in soil or vegetation, as well as the intestines of mammals, including humans. Total coliform bacteria are not likely to cause illness, but their presence indicates that the water supply may be vulnerable to contamination by more harmful microorganisms. Escherichia coli (E. coli) is the only member of the total coliform group of bacteria that is found only in the intestines of mammals, including humans. The presence of E. coli in water indicates recent fecal contamination and may indicate the possible presence of disease-causing pathogens, such as bacteria, viruses, and parasites. Although most strains of E. coli bacteria are harmless, certain strains, such as E.coli 0157:H7, may cause illness. About 80 % of communicable diseases in the world are waterborne. According to WHO estimate about 80 % of water pollution in developing country, like India is carried by domestic waste. In India 70 % of the water is seriously polluted and 75 % of illness and 80 % of the child mortality is attributed to water pollution. The improper management of water systems may cause serious problems in availability and quality of water. The major pathogenic bacteria responsible for water borne disease are spread by the faeco-oral route, in which water may play an intermediate role. The aim of this review is to examine methods currently in use for the detection of coliforms in drinking water and also to evaluate the possible health hazards associated with drinking water contaminated with coliforms.

Air pollution is a matter of great concern in the globe. Generally air pollutant generates from industries, automobiles, etc. and the primary pollutants may easily convert to secondary pollutants. Both of these pose serious threat to the plant community viz. crops, vegetables and avenue plant species are depending on the emission pattern, atmospheric transport and leaf uptake and on the plant’s biochemical defense capacity. An impact caused by air pollutants depends not only upon its concentration, but also on the duration (acute and chronic exposure) and combination of air pollutants. Biomonitoring on plant species is an easy tool to know bioindicator species in which exposure of air pollutants can easily be identified. The present review deals with past and present research works of major gaseous pollutants emissions and their impact on crop, vegetables and tree species performance from available literatures.

Vibrio cholerae, the causative agent of Asiatic cholera, is a gram-negative motile bacterial species acquired via oral ingestion of contaminated food or water sources. Cholera has spread from the Indian subcontinent where it is endemic to involve nearly the whole world seven times during the past 185 years. V. cholerae serogroup O1, biotype El Tor, has moved from Asia to cause pandemic disease in Africa and South America during the past 35 years. A new serogroup, O139, appeared in south Asia in 1992, has become endemic there, and threatens to start the next pandemic. The facultative human pathogen V. cholerae represents a paradigm that evolved from environmental non-pathogenic strains by acquisition of virulence genes. The major virulence factors of V. cholerae, cholera toxin (CTX) encoded by the ctxAB genes residing in the genome of filamentous lysogenic bacteriophage (CTXɸ) and toxin coregulated pilus (TCP) encoded by vibrio pathogenicity island (VPI). CTX, a potent stimulator of adenylate cyclase, causes the intestine to secrete watery fluid rich in sodium, bicarbonate, and potassium, in volumes far exceeding the intestinal absorptive capacity, by ADP-ribosylation of the alpha subunit of the GTP-binding protein. Thus intestinal infection with V. cholerae results in the loss of large volumes of watery stool, leading to severe and rapidly progressing dehydration and shock. Without adequate and appropriate rehydration therapy, severe cholera kills about half of affected individuals. Today, cholera still remains a burden mainly for underdeveloped countries, which cannot afford to establish or to maintain necessary hygienic and medical facilities. During the last three decades, intensive research has been undertaken to unravel the virulence properties and to study the epidemiology of this significant human pathogen. This review provides an overview of the role of CTX in the occurrence of this disease in humans.

Opis:
The evolutionary transition from phenotypic to molecular analysis of infectious disease in bacterial epidemiology led to the search for suitable approaches to ascertain genomic relatedness or heterogeneity between bacterial clinical isolates. Pulsed-field gel electrophoresis (PFGE) technique was developed for separating and analyzing long DNA fragments of several megabases in alternating electric field. Comparison of electrophoresis profiles of restriction enzyme-digested genomic DNA from bacterial isolates has proved to be a useful epidemiological tool for genetic discrimination of bacterial strains, detection of genetic relatedness, to locate the source of outbreak and to monitor the spread of the microorganisms in endemic zones. PFGE is considered as a gold standard method for typing of bacterial isolates because of the remarkable endurance of this technique as a typing method for the last 20 years in molecular epidemiology. In this current review the pros and cons of PFGE use in current molecular microbiological research are explored in the context of determination of genome organization of certain food-borne bacterial isolates causing infectious diseases in human beings.

Shigella species are intracytosolic Gram-negative invasive enteropathogenic bacteria, causing the rupture, invasion and inflammatory destruction of the human colonic epithelium. They utilize the host cytoskeletal components to form propulsive actin tails. The so-called invasive phenotype of Shigella is linked to expression of a type III secretory system (TTSS) injecting effector proteins into the epithelial cell membrane and cytoplasm, thereby inducing local but massive changes in the cell cytoskeleton that lead to bacterial internalization into non-phagocytic intestinal epithelial cells. The molecular and cellular bases of this invasive phenotype essentially encompass crossing of the epithelial lining, apoptotic killing of macrophages, entry into epithelial cells, and escape into the cytoplasm, followed by cell-to-cell spread. Intracellular colonization is likely to protect the micro-organisms from killing by humoral and cellular effectors of the innate immune response. Concurrently, the capacity of Shigella to reprogram invaded epithelial cells to produce proinflammatory mediators plays a major role in the strong inflammatory profile of the disease. This profile is likely to impact on the nature and quality of the adaptive response, which is dominated by humoral protection at the mucosal level. In recent years, a large amount of information has been generated regarding the host, pathogen and environmental factors that impact the pathogenesis of shigellosis at the cellular and molecular level. This review summarizes what is currently known about Shigella, detailing those factors that contribute to pathogenesis and examining the current progress in the development of a vaccine.

The application of random amplified polymorphic DNA- polymerase chain reaction (RAPDPCR) was found to be a simple, cheap and rapid tool to discriminate human pathogenic bacterial isolates especially at intraspecific level. This molecular biological technique relies on the use of random oligonucleotide primers that arbitrarily amplifies specific regions of the genome which gives rise to a unique genomic fingerprint of the strains under investigations. With continued development of novel molecular-based technologies for rapid, high-throughput detection of food borne pathogenic bacteria, the future of conventional microbiological methods such as viable cell enumeration, selective isolation of bacteria on commercial media, and immunoassays seems tenuous. Approaches that enhance recovery of sub lethally injured bacteria, differentiation among species, differentiation among bacteria of interest using biochemical profiling, enumeration using impedance technology, techniques to confirm the presence of target pathogens using immunological methods, and bioluminescence applications for hygiene monitoring are of utmost need in identifying and combating the human pathogenic isolates. The aim of this study is to estimate the efficiency of RAPD-PCR technique in assessing the genetic diversity of diseases causing bacterial isolates. The use of RAPD-PCR in evaluating the genomic variability among the pathogenic strains belonging to different genus are also been discussed in the present report.

Diarrhoea in developing countries is caused by an increasingly long list of bacterial, viral, and parasitic pathogens with rotavirus, Enterotoxigenic Escherichia coli, Campylobacter, Shigella, and Salmonella heading the list. Using methods to detect most of the known enteropathogens, one or more enteropathogen(s) is isolated in two-thirds of diarrhoeal illnesses in the developing world. Deoxyribonucleic acid probes have proved very useful in detecting pathogens such as enterotoxigenic (ETEC), enteroinvasive (EIEC), enteropathogenic E. coli (EPEC), and Shigella but have not yet proved to be particularly rapid or less expensive. Molecular biology has proved useful in epidemiological studies as a means of strain identification and detection of genome diversity. Since the introduction of ribonucleic acid gene restriction patterns as taxonomic tools in 1986, ribotyping has become an established method for systematics, epidemiological, ecological population and genome diversity studies of microorganisms including Shigella. The technological development culminated in the automation of ribotyping which allowed for high-throughput applications. PCR ribotyping has proved being a highly discriminatory, flexible, robust and cost-efficient routine technique which makes inter-laboratory comparison and build of ribotype databases possible, too. The aim of the present review is to determine the present status of ribotyping technique in detecting the diversity in Shigella isolates.

Shiga toxins belonging to a family of structurally and functionally related protein toxins serve as the key virulence factors for pathogenecity of the virulent enteric bacterial strains namely Shigella dysenteriae serotype 1 including Shiga toxin-producing Escherichia coli (STEC). S. dysenteriae type 1 isolates remain major public health concerns due to their widespread outbreaks and the severity of extra-intestinal diseases, including acute renal failure and also affects the central nervous system. Despite practicing improved hygienic conditions and regulating food and drinking water safety, the enteric pathogens are imposing a major threat in the well being of the human society. Shiga toxin on entry into host cells’ endoplasmic reticulum (ER) activates the stress response in ER and inhibits protein synthesis by catalytic inactivation of eukaryotic ribosomes. In many cell types shiga toxins trigger apoptosis. Recent studies have shown that shiga toxins induce autophagy which activates different signaling pathways in toxin-sensitive and toxin-resistant human cells. In this review the molecular basis of Shiga toxins’ effect on host cell leading to manifestation of the infection in affected individuals are discussed with an emphasis on recent findings.

The practice of eating insects is known as entomophagy. Many animals, such as spiders, lizards and birds, are entomophagous, as are many insects. People throughout the world have been eating insects as a regular part of their diets for millennia. As people in rural areas suffer from under nutrition, especially protein-energy malnutrition (PEM) in Africa, Latin America and Asia, alternative nutritional food sources are needed. From ants to beetle larvae – eaten by tribes in Africa and Australia as part of their subsistence diets – to the popular, crispy-fried locusts and beetles enjoyed in Thailand, it is estimated that insect-eating is practised regularly by at least 2 billion people worldwide. More than 1900 insect species have been documented in literature as edible, most of them in tropical countries. The most commonly eaten insect groups are beetles, caterpillars, bees, wasps, ants, grasshoppers, locusts, crickets, cicadas, leaf and plant hoppers, scale insects and true bugs, termites, dragonflies and flies. The purpose of the present review is to determine the status of present research in the context of the potentiality of insects as alternative food source to cope up with the emerging problem of global food crisis.

Over the years laser welding has evolved as a fabrication process capable of overcoming the limitations of conventional joining methodologies. It facilitates the welding of diverse range of materials like metals, non-metals, polymers etc. Laser transmission welding is a technique employed for fabricating intricate shapes/contours in polymers with better precision compared to the other conventional processes. Nylon6, a synthetic semi-crystalline polymer is utilized as an engineering thermoplastic due to its high strength and temperature resistant properties. In the earlier researches, various welding techniques were employed for the fabrication of polymers and metals keeping the laser beam stagnant, and much emphasis was given only to temperature distribution along the different axes and limited attention was given to residual stress analysis. Therefore, in this research work, a three-dimensional time-dependent model using a moving laser beam is used to fabricate unreinforced Nylon6 specimens.

The data of individual sector on environmental science is compiled in information technology (IT) through database software known as Environment Information (EI). Environment information is an easy accessible database in individual sector, where people can know both new research scope and impact on environment. The present study deals to inform that EI is an area, in which the knowledge of environmental science and IT is combined through database software. In this paper, individual research area, data format and beneficial aspects on environmental science is tabulated for EI. There are several research areas in which the data viz. ecology and ecosystem, biodiversity and conservation of important species, health hazards by diseases, health care facilities, toxicological aspects, wastes types, source generation and management, alternative energy generation facilities, environment education and awareness etc. It is a conceptual approach to gather knowledge of environment related problems and prospects and can easily make a database for the intellectuals, academicians, scientists, regulatory authorities, policy makers, researchers, students etc. These help to know benefits in research area, regulatory process, decision making and proper environment management. People can easily access compiled database in an individual sector of environment science.

An experiment was conducted to evaluate the effect of different concentration and combination of potassium [1.0 g/l (K1), 2.5 g/l (K2) and 5.0 g/l (K3)] and gibberellic acid [0.005 g/l (G1), 0.01 g/l (G2) and 0.015 g/l (G3)] foliar application on growth characteristics, quality and yield of F1hybrid cucumber (cv. KUK-9).The results showed that the highest amounts for growth, biochemical attributes, fruit maturity, quality belonged to K2 (2.5 g/l) plus G2 (0.01 g/l) combination treatment. Plant height as well as chlorophyll content attained their greatest quantities under G2K2 treatment. Dry matter, TSS % and antioxidant activity showed significant differences compared with control, G1K1 and G2K2 treatment showed the greatest sum of data. Fruit mineral content were also significantly affected by different treatments. Higher fruit potassium was recorded with G3K3 treatment.

This paper presents a mathematical model of a power controller for a high-frequency induction heating system based on a modified half-bridge series resonant inverter. The output real power is precise over the heating coil, and this real power is processed as a feedback signal that contends a closed-loop topology with a proportional-integral-derivative controller. This technique enables both control of the closed-loop power and determination of the stability of the high-frequency inverter. Unlike the topologies of existing power controllers, the proposed topology enables direct control of the real power of the high-frequency inverter.
