Opis:
Antazoline is an antihistaminic, immunosuppressive, antiarrhythmic agent. Antazoline can be administered orally, intravenously, intramuscularly or via the ophthalmic route. Antazoline has a limited application as an antiarrhythmic drug. This review was undertaken with the aim to combine the old and the new results of different types of studies (clinical, retrospective, or pharmacokinetic) and sum up the positive and negative effects of antazoline in cardiology and emergency medicine. A literature queries were performed in the following databases: PubMed, Embase, Google scholar, and Web of Science (all from their inception date till June 2019). The queries covered antazoline in combinations with such terms as antiarrhythmic activity, arrhythmia, electrocardiography, emergency medicine, and cardiology. Additional publications were found by checking all the reference lists. The newest research shows that antazoline may have the highest success rate of pharmacological cardioversion among all drugs used in the contemporary treatment of cardiac arrhythmia (up to 85.3%). The rate of cardioversion with antazoline alone was higher than combined amiodarone and/or propafenone. Most of the studies which were reviewed indicated that paroxysmal atrial fibrillation, compared to chronic atrial fibrillation, responded more satisfactorily to antazoline treatment. Most patients tolerated antazoline well and conversion was safe, effective, and smooth. Some research proves that the antazoline use may also reduce the rate of hospitalization due to the adverse effects. Antazoline has a definite place in the therapeutic repertoire for several cardiac arrhythmias. It should not be neglected as an old drug, and ought to be reconsidered in emergency medicine treatment recommendations

Opis:
The purpose of this study was to analyze the indolic, phenolic, and fatty acid content and antioxidant activity of garlic sprouts growing in the dark and in the daylight. The pro- or anti-inflammatory properties of the garlic sprout extract were investigated by evaluating the cyclooxygenase-2 (COX-2), prostaglandin E synthase (cPGES), glutathione S transferase (GSTM1), nuclear factor NF-κB, peroxisome proliferator-activated receptors (PPARs), and aryl hydrocarbon receptor (AhR) protein levels in the RAW 264.7 cells activated with lipopolysaccharide (LPS). The highest amount of total indolic (73.56 mg/100 g f.w.) and phenolic compounds (36.23 mg/100 g f.w.) was detected in garlic sprouts grown in the daylight. Studies on antioxidant activity (the FRAP and DPPH method) of garlic sprouts showed that this activity is significantly higher for sprouts grown in full access to light when compared to those grown in the dark. In garlic sprout extracts, α-linolenic acid (ALA) was found to be in greater amount. COX-2 and cPGES level was lower when compared to LPS alone activated cells. After garlic extract treatment, higher level of GSTM1, PPARΥ, cytosolic p50 and p65 protein, as well as a lower NF-ĸB p50/p65 activity was noted in the RAW 264.7 cells which suggested PPARs and AhR transrepression mechanism of NF-ĸB signalling. The obtained results indicate Allium sativum sprouts are a rich source of n-3 fatty acids, indolic and phenolic compounds characterized by anti-inflammatory and antioxidative activity, which may support their high therapeutic and dietary potential.

Alterations in membrane lipid composition, particularly fatty acids content is known to result in functional and structural changes. Dietary lipids and physical activity or stress play an important role in this change. It was of interest to study the influence of swimming in different temperatures on the content of fatty acids in the phospholipids of the erythrocyte membranes of rats. Our research demonstrated a significant influence of swimming on the content of fatty acids in the phospholipids of the erythrocyte membranes of rats. These changes of fatty acids, particularly the decrease of arachidonic acid and significant increase of saturated fatty acids in the phospholipids of the erythrocyte membranes in swimming rats may be induced by oxygen radicals generated in different kinds of stress.

It was reported that dietary fructose imposes a number of effects on lipid metabolism including hypertriglyceridemia. The daily intake of fructose in humans is mainly due to sucrose. It was reported that the consumption is still increasing, making a background for health implications. The mechanism of metabolic disorders is poorly understood, but a lot of studies indicate that the liver lipid homeostasis deregulation is essential for a fructose effect on metabolism. The aim of the study is to estimate if fructose affects the profile of fatty acids in in vitro and in vivo models. In this study in vitro and in vivo experiments were conducted to assess the effect of dietary fructose on the fatty acid profile in the cell culture or in the liver of rats. The results showed that in the fructose experimental groups, both in the cell and liver homogenates, the content of the saturated fatty acids were significantly higher than in control groups. According to the obtained data fructose in the medium and in the diet affects saturation of fatty acids in the cell cultures and in the livers of rats. The findings obtained in the experiments support the thesis that fructose influences the homeostasis of lipid metabolism in the liver and may give an opportunity to discuss the limitation of the content of this kind of sugar in food.

Opis:
Metabolism of polyunsaturated fatty acids results in biosynthesis of mediators with different physiological effects. These metabolites include prostaglandins, prostacyclins, isoprostanes and others that are important signalling molecules and regulate a variety of physiological and pathophysiological processes including inflammation. Prostaglandins and isoprostanes are produced by either non-enzymatic lipid peroxidation or by enzyme-induced peroxidation (cyclooxygenases and lipoxygenases). They are used as biomarkers of oxidative stress. The aim of our study was to assess the effect of eicosapentaenoic acid (EPA) supplementation with added benzo(a)pyrene (BaP) on HepG2 cells by using a UHPLC/MS-TOF method. This rapid and simple method was developed for the identification, separation and quantification of 8-iPGF3α, PGF3α, 8-isoPGF2α and 5-iPF2α in cultured cells. The UHPLC/MS-TOF method was validated. The calculated limit of detection was in the range of 0.16-0.50 ng/mL, precision (% RSD): 1.2-2.1% and recoveries better than 88%. This method empowered qualitative and quantitative analysis of the selected individual prostaglandins derived from arachidonic acid and eicosapentaenoic acid from cell extracts.

The aim of the study was to investigate the influence of kohlrabi and broccoli sprouts added to diet on the concentrations of short-chain fatty acids in faeces samples of rats with thyroid dysfunction. Two models of hypothyroidism were used. First one was based on the diet with iodine deficiency and the second by sulfadimethoxine addition to the animal drinking water. In most cases the concentrations of acetic acid, propanoic acid and butanoic acid in stool samples obtained from rats with thyroid dysfunction were insignificantly lower than found in control group. Moreover, almost in all cases the diet enriched with brassica sprouts insignificantly increased the concentration of fatty acids in investigated material. Significant positive correlation between concentrations of butanoic and acetic acids was found in different groups of rats. In conclusion, the addition of brassica or kohlrabi sprouts in rat’s diet may slightly alleviate the decline of concentrations of some fatty acids in stool of animals with thyroid dysfunction. Long term effects of this phenomenon should be evaluated in future studies.

Free radicals containing one or more unpaired electrons have considerable reactivity. Those radicals derived from oxygen represent the most important class of such species. Free radicals generated in organism are reactive oxygen species (ROS) and reactive nitrogen species (RNS) well recognised for playing a dual role as both deleterious and beneficial species. The cumulative production of ROS/RNS is termed oxidative stress and were discovered in pathomechanism of many of diseases. It was observed that in medium concentration ROS and RNS are play a role in inflammation, apoptosis and as secondary messengers in intracellular signalling cascades.
