Fatty acid amide hydrolase (FAAH) inhibitor PF-3845 reduces viability, migration and invasiveness of human colon adenocarcinoma Colo-205 cell line: an in vitro study

Earlier reports suggest that the endocannabinoids may play a role of endogenous tumor growth modulators. In this study, we investigated whether inhibition of the enzymes involved in the synthesis and degradation of endocannabinoids may reduce colorectal cancer cell invasion and migration. The human colon adenocarcinoma Colo-205 cells were incubated with PF-3845, JZL-184 and RHC-80267 (fatty acid amide hydrolase (FAAH), mono- (MAGL) and diacylglycerol lipase (DAGL) inhibitors, respectively) for 48 h. The MTT colorimetric assay was performed to quantify cell viability. Next, Colo-205 cells were incubated with PF-3845 alone or with PF-3845 together with selected antagonists: AM 251, AM 630, SB 366791, RN 1734 and G-15 (CB1, CB2, TRPV1, TRPV4 and GPR30 antagonists, respectively). Western blot assay was applied to identify the changes in CB1 and CB2 receptor expression. Migration and invasion assays were employed to characterize the effect of PF-3845 on colorectal cancer cell invasion. We found that of all the inhibitors used, the FAAH inhibitor PF-3845 reduced the Colo-205 cell line viability the most effectively (IC50=52.55 μM). We also showed that the effect of decreased cell viability was enhanced when Colo-205 cells were incubated with PF-3845 and RN-1734, a TRPV4 antagonist (IC50=30.54 μM). Western blot assay revealed significantly decreased CB1 receptor expression levels, while CB2 expression was increased in response to PF-3845 when compared to control. Furthermore, PF-3845 inhibited migration and invasion of Colo-205 cell line. These results suggest that pharmacological inhibition of FAAH and consequent enhancement of the endocannabinoid levels may reduce the colorectal cancer growth and progression.

Natural polyphenols and polyphenol-rich extracts have been found to possess preventive and therapeutic potential against several types of cancers, including colorectal cancer (CRC), which is an example of an inflammation-associated cancer. This study examines the chemopreventive effect of a Japanese quince (Chaenomeles japonica) fruit flavanol preparation (JQFFP) on colon cancer SW-480 cells. JQFFP, rich in procyanidin monomers and oligomers, was found to inhibit the SW-480 cell viability by 40% at 150 µM catechin equivalents (CE) after 72 h incubation when compared to control, but it was non-toxic to normal colon fibroblast CCD-18Co cells. Furthermore, 100 µM CE JQFFP suppressed COX-2 mRNA expression to 36.7% of control values and protein expression to 77%. In addition, JQFFP reduced the MMP-9 protein expression (to 24% vs. control at 100 µM CE) and caused inhibition of its enzymatic activity (to 35% vs. control at 100 µM CE). Not only did JQFFP inhibit the COX-2 and MMP-9 levels, but it also reduced the NF-κB protein expression (to 65% of control) and phosphorylation of its p65 subunit (to 51%) at 100 µM CE. These results provide the first evidence that JQFFP inhibits COX-2, MMP-9, and NF-κB expression, suggesting that it has cytotoxic, anti-inflammatory, and anti-metastatic activities towards the colon cancer SW-480 cells.

The present study concerns the determination of the characteristics of bioclimatic conditions, as well as the synoptic situations related to the occurrence of thermal stress conditions, in Poland. The study was based on daily data obtained from the Institute of Meteorology and Water Management – National Research Institute from the period 1966–2020 for 37 synoptic stations in Poland. Based on the obtained data, values of the Universal Thermal Climate Index (UTCI) were calculated. The occurrence of heat stress increases from the north to the south, corresponding with the variability of influx of solar radiation, and is modified by factors at a smaller spatial scale. The results of this paper evidently point to the cooling effect of the waters of the Baltic Sea. In circulation conditions favouring strong and very strong heat stress, e.g. in two of the designated circulation types (T1 and T2), the occurrence of an expansive high-pressure ridge in the Atlantic-European area is typical, stretching from the region of the Azores High towards the north-east, with a secondary high developed within its boundaries. In the third of the designated circulation types (T3), the high-pressure area extends from the Azores eastwards, reaching the Black Sea. Each of the three circulation patterns associated with the unfavourable biometeorological conditions of very strong and extreme cold stress in Poland is characterised by strong pressure centres formed in the Euroatlantic region, triggering the airflow from the northern (T4) or eastern (T5, T6) sector.

W przypadku wystąpienia na stanowiskach pracy szkodliwych i niebezpiecznych czynników chemicznych i pyłów konieczne jest zapewnienie przez pracodawcę skutecznej ochrony pracowników narażonych na te czynniki. Priorytetowym rozwiązaniem są działania organizacyjne i zbiorowe środki techniczne. Na wielu stanowiskach pracy stosuje się jednak w tym celu środki ochrony indywidualnej. W szczególności dotyczy to sprzętu ochrony układu oddechowego, odzieży ochronnej, środków ochrony rąk i nóg oraz środków ochrony oczu i twarzy. Podstawowym problemem jest wybór przez pracodawcę optymalnego zestawu tych środków, biorąc pod uwagę narażenie na czynniki chemiczne i pyłowe oraz minimalny dyskomfort dla pracownika. Przydatne do tego celu są dane związane z charakterystyką poszczególnych rodzajów środków, sposobem ich klasyfikacji oraz procedurami doboru do różnego poziomu zagrożeń. Informacje te można znaleźć w licznych publikacjach tematycznych, ale ich analiza wydłuża czas potrzebny pracodawcom do podjęcia prawidłowej decyzji. W ramach V etapu programu wieloletniego zrealizowano zadanie 3.SP.08, którego celem było zebranie i usystematyzowanie wiedzy na temat środków ochrony indywidualnej. W artykule zaprezentowano próbkę informacji, które dotyczą doboru przykładowych środków ochrony indywidualnej do zagrożeń chemicznych i pyłów. Zakres tematyczny artykułu obejmuje zagadnienia zdrowia oraz bezpieczeństwa i higieny środowiska pracy będące przedmiotem badań z zakresu nauk o zdrowiu i inżynierii środowiska.
In the event of an emergence of harmful and hazardous chemical and dust factors at workplaces, the employer must provide adequate protection for employees exposed to these factors. As a rule, organizational measures and collective technical measures are a priority. However, many workplaces use personal protective equipment for this purpose. This applies particularly to respiratory protective devices, protective clothing, hand and leg protection, and eye and face protection. The selection of the optimal set of these measures, considering the exposure to chemical and dust factors and the need to ensure employees suffer from the minimum level of discomfort, is a fundamental problem. The data related to the characteristics of individual types of hazards, their classification and selection procedures for various levels of exposure is essential in this regard. This information can be found in numerous thematic publications, but their analysis extends the time needed by employers to make the correct decision. Therefore, as part of the 5th stage of the Multiannual Program, task no. 3.SP.08 was carried out to collect and systematize knowledge about personal protective equipment. The article presents an extract of information from the knowledge base concerning selecting personal protective equipment for chemical and dust hazards. This article discusses the problems of occupational safety and health, which are covered by health sciences and environmental engineering.

Patients with Crohn’s disease (CD) prefer carbohydrates and foods, which easy to digest, both risk factors of caries disease. The aim of this study was to evaluate the dental caries prevalence and oral hygiene habits in CD patients who live in urban and rural areas of the southern part of Poland in comparison to controls. Seventy one adult patients with CD and 61 age – and sex – matched volunteers were enrolled; 52% of patients were living in urban areas, 48% in rural regions. We studied: findings on clinical examination, calculation of BMI, blood hemoglobin and serum CRP levels, oral examination according to the WHO recommendations, with assessment of oral hygiene habits, dental history, visits to dentists. Presence of decayed (D), missing (M), filled teeth (F) were assessed, and the “Decayed, Missing, and Filled Teeth Index” (DMF-T) was calculated. Statistical methods included non-parametric tests and Spearman correlation coefficient estimation. Dental caries assessed by DMF-T index was higher in CD patients compared to controls. DMF-T was similar in patients from urban and rural areas (14.2±4.7 vs 15.4±2.1, respectively). M and F scores were comparable in both groups, but D score was higher in CD patients (p<0.01). No correlation was found between DMF-T index and place of living. Patients from rural areas less frequently used tooth brushing (68% vs 57%, p<0.05) and dental floss (11% vs 5%, p<0.05). Patients from urban areas visited dentists more frequently than those from rural regions. The prevalence of dental caries in the middle aged CD patients living in the southern region of Poland is higher in comparison to healthy controls. The prevalence of caries disease and oral hygiene habits in CD patients is comparable between patients living in urban areas and in rural regions.
