The emergence of resistance in microorganisms on a global scale has made it necessary to search for new antimicrobial factors. Antimicrobial peptides (AMPs) seem to meet these expectations. AMPs are produced by bacteria, viruses, plants, and animals, and may be considered as a new class of drugs intended for the prophylaxis and treatment of both systemic and topical infections. The aim of this study is to review the results of studies on the use of peptides to combat infections in vivo. Antimicrobial peptides may be applied topically and systemically. Among the peptides used topically, a very important area for their application is ophthalmology. AMPs in ophthalmology may be used mainly for the protection of contact lenses from ocular pathogens. Many AMPs are in clinical trials for application in the therapy of local infections. There may be mentioned such preparations as: pexiganan (magainin analogue), MX-226 (based on indolicidin), NEUPREX (isolated from human BPI (bactericidal/permeability-increasing) protein), IB-367 (variant of porcine protegrin), P113 (based on histatin), daptomycin, polymyxins, as well as peptidomimetics. In the combat against systemic infections are used such peptides as: P113D (modified P113 peptide containing D-amino acids), colistin, peptoids, and peptides containing non-typical amino acids or non-peptide elements. AMPs are also used as antiprotozoal, antifungal, antitoxic and immunostimulatory agents. The limitations in the use of peptides in the treatment of infections, such as susceptibility to proteolysis, and resistance of microorganisms to the peptides, are also discussed. AMPs are a promising strategy in the fight against microbial infections.

Existing research for using the protective antigen (PA) of Bacillus anthracis as a vaccine component shows that protection against anthrax may be obtained using fragments of this protein. The aim of the research is to check whether the selected protein fragment of the protective antigen (domain 4) encoded by an appropriate nucleotide sequence of gene pag of B. anthracis, was expressed in the bacterial system of E. coli. In order to examine the selected sequence of the pag gene, a PCR reaction and a highly effective TOPO cloning strategy were used, followed by purification of the recombinant proteins and their detection by a western-blot method. In the planning of the PA4 antigen expression a higher level of effectiveness in production of small protein – domain 4 – was anticipated. As a result, the 139 amino acids protein fragment of B. anthracis PA (domain 4) was isolated. The research may have found the basis for in vivo research aimed at finding potential anthrax vaccine components.

Health care workers (HCW) worldwide are especially exposed to injury by sharp instruments in the course of their duty. The most often executed procedures with injury risk are: intramuscular or subcutaneous injection (22%), taking blood samples, or during intravenous cannulation (20%), and repeatedly replacing the cap on an already used needle (30%). Even a minor sharp injury with only a small loss of blood carries the risk of transfer of over 20 pathogens: Hepatitis B Virus (HBV), Hepatitis C Virus (HCV), HIV/AIDS virus, malaria, syphilis, tuberculosis, brucellosis, herpes virus and diphtheria. The World Health Organization (WHO) estimates that more than two million health care workers experience the stressful event of a percutaneous injury with a contaminated sharp object each year (25-90% of them, however, remain unreported). These exposures result in about 16,000 infections with HCV, 66,000 with HBV and about 1,000 (200-5000) with HIV, which lead to about 1,100 deaths or significant disability. Exposures to sharp injuries and their consequences are highly preventable through simple interventions, such as HBV vaccination, education and providing containers for sharp instruments. Specific guidelines, similar to the American Occupational Safety and Health Administration (OSHA) regulations that have lowered by up to 88% of needle stabbing incidents, should be introduced by the European Union (EU) and other countries. The results of a review of reports leads to the following conclusions: 1) elaboration and implementation of new State regulations, especially in EU countries and in countries where such regulations do not exist; 2) the training of health care personnel should always be undertaken for new employees, and periodically for those already employed; 3) periodical control by appointed inspectors of knowledge of procedures for the prevention of injuries by sharp instruments among health care workers; 4) introducing and training in the use of equipment, which can prevent the sharp injuries; 5) an advanced monitoring system of sharp injuries sustained by worker should also be introduced. Successful implementation of these prevention measures will result in progress for public health and HCW’s health and safety.

Introduction. Dobrava-Belgrade virus (DOBV) is one of the emerging pathogens which have been reported during the last decades in Europe and have attracted the attention of researchers. The course of infection among humans may have a varied course – from the completely asymptomatic to the more severe forms, such as haemorrhagic fever with renal syndrome (HFRS). DOBV is hosted and carried by rodents like Apodemus flavicollis or A. agrarius, which occur commonly in Europe. Objective. To-date, orthohantaviruses have been reported in Poland, both in humans and animals, but detailed countryscale studies have not yet been carried out. The aim of the study was molecular characterization of a strain which was found in A. flavicollis in south-eastern Poland. Materials and method. The phylogenetic analysis of the first Dobrava-Belgrade virus found in A. flavicollis in the subcarpathian region of south-eastern Poland, presented in this study, was performed after virus proliferation in cell culture and sequencing of specific PCR products. Results. Based on genetic sequences of fragments of three segments (S, M and L), the isolated virus was assigned to the Dobrava genotype, taking into consideration the most current classification of the DOBV species. Conclusions. The Dobrava-Belgrade virus strain isolated from A. flavicollis in the subcarpathian region of south-eastern Poland, has been molecularly characterized and assigned to Dobrava genotype, thereby the occurrence of that genotype in Poland has been confirmed by molecular techniques.

Celem pracy było określenie dynamiki zmian koncentracji albumin oraz frakcji wybranych białek o masach zarówno wyższych, jak i niższych od albumin. Zbadano również zmiany procentowego udziału tych frakcji w białku całkowitym osocza krwi. Badania przeprowadzono na siedmiu cielętach w pierwszych siedmiu dniach życia. Wykazano, że zmianom wraz z wiekiem ulega zarówno koncentracja, jak i procentowy udział albumin oraz białek o masach cząsteczkowych wyższych i niższych w białku całkowitym osocza krwi cieląt noworodków. Średnie stężenie albumin w osoczu krwi w pierwszym tygodniu wahało się w granicach od 29,22 do 31,72 g ・ l–1. Procentowy udział tej frakcji w białku całkowitym osocza krwi obniżał się od 38,14 do 29,43%. Stężenie frakcji białek wysokocząsteczkowych wykazywało tendencję wzrostową (od 8,43 do 14,47 g ・ l–1). Średni procentowy udział tej frakcji w białku całkowitym wahał się od 10,71 do 13,22%. Koncentracja białek niskocząsteczkowych wzrastała w pierwszych czterech dniach życia od 18,73 do 36,60 g ・ l–1, a ich procentowy udział w białku całkowitym wzrastał od 24,10 do 31,64%.
The aim of the present study was to evaluate dynamic changes in the concentration of albumin, and selected protein fractions with higher and lower molecular weight in comparison to albumin. The percentage of these fractions of the total blood plasma protein was also ex-amined. The experiment was carried out on 7 calves during their first seven days of life. Additionally, changes with age of the albumin and high and low molecular weight protein concentration and percentage of the total blood plasma protein were also determined. During the first week of life mean albumin concentration in the blood plasma varied between 29.22–31.72 g ・ l–1. The percentage of these fractions of the total blood plasma protein decreased from 38.14 to 29.43%. The concentration of high molecular weight fraction rose (from 8.43 to 14.47 g ・ l–1). Average percentage of these proteins of the total protein varied between 10.71–13.22%. Both low molecular weight protein concentration (from 18.73 to 36.60 g ・ l–1) and their percentage of the total protein (from 24.10 to 31.64%) increased during the first four days of life.

Introduction: In the autumn of 2009 the authors participated in a humanitarian operation in Western Ukraine by undertaking an epidemiological investigation of an influenza-like-illness (ILI) in the L’viv Oblast region. Mobile biological survey teams took samples from civilian patients with severe acute respiratory distress syndrome, rapid transportation of the samples, and their molecular analysis in Poland to provide accurate results. Objective: The aim of the study was the molecular and epidemiological analysis of the biological samples collected. Material and Methods: Real-time reverse transcriptase polymerase chain reaction (real-time RT-PCR), multiplex PCR techniques, traditional Sanger Sequencing and classical viral culture methods were used. Results: Among the 124 influenza-like illness cases, ~50% (58) were positive for influenza A virus in WHO-CDC molecular assay, including subtyping. The specimens were further analyzed to confirm results and determine the genetic sequence. Phylogenetically, the nucleotide similarity of both the Ukraine specimens and reference A/California/7/2009 (pH1N1) was 99.2–99.3%. Oseltamivir resistance was not registered. HA1 region characterization showed an overall protein identity of 98.5–99.4%. Conclusions: An unexpected high contribution of influenza A was confirmed among ILI patients, as well as a very limited number of other detected viruses, indicate that the 2009 epidemic in western Ukraine was strongly related to novel influenza A/H1N1. The importance of swift sharing of information and reference laboratories networking in surveillance, as well as serving governments and international agencies in pursuing adequate actions, should be stressed.
