Opis:
In this study, the proteomic, morphometric, and photosynthetic pigment data of purslane (Portulaca oleracea) accessions were combined together to show their impact on genetic variation in order to establish a relationship between protein patterns and phenotypic behavior of the plant. Seeds of 18 collected purslane accessions were cultivated based on a completely randomized design with three replicates. Before the flowering stage, the data on morphology, photosynthetic pigment content, and seed proteins were obtained. The results showed a significant difference among purslane accessions in terms of the most studied agronomic characteristics and the content of photosynthetic pigments and proteins. The cluster analysis of the 18 purslane accessions based on agronomic data, and photosynthetic pigment content, and protein pattern data produced three main clusters. Moreover, the seed protein analysis revealed that the two polymorphic protein bands of size 40 kDa (protein “a”) and 30 kDa (protein “b”) effectively diversified the agronomic, photosynthetic pigment, and phylogenetic relationships among the purslane accessions. Interestingly, protein “a” was produced in plants growing in low altitude areas and played a suppressive role for TDW, while protein “b” was produced in plants growing in high altitude areas and functioned as an activator agent for this trait. Overall, the outcomes of the present study indicated the presence of high genetic variability (77.6%) among the purslane accessions. These findings suggest that these proteins should be sequenced for further proteomic analyses and can be used for hybridization to generate useful recombinants in segregating generations and improve breeding varieties of P. oleracea.

Opis:
Sugar beet is one of the most important industrial crops in Iran. For the last two decades it has been mainly affected by a destructive virus, beet necrotic yellow vein virus (BNYVV). The Polymyxa betae is the only natural transmitting agent of the disease among the plants. Developing accurate diagnostic methods may have a major impact on the rising of resistant germplasms. In the present study, specific monoclonal recombinant antibodies in the form of single chain variable fragments (scFv) were obtained from naïve phage display libraries. The fungus specific glutathione-S-transferase (GST) protein was chosen as an antigen for developing antibodies and diagnostic purposes. To generate specific scFv, screening of Tomlinson phage display libraries was performed by applying both recombinant and native fungal GST. Using the recombinant GST in the panning process resulted in the isolation of an antibody only bound to recombinant GST but it failed to detect native GST in the infected plants. Alternatively, the process of panning was carried out by applying native fungal GST trapped to immunotubes through specific polyclonal antibody intermediate. The recent approach resulted in the selection of a specific scFv binding to native GST which was able to detect the presence of the fungi within infected plants. To the best of our knowledge, this is the first report on the generation of recombinant antibodies against Polymyxa betae, fungal vector of sugar beet rhizomania disease.

Opis:
Purpose: of this paper is to investigate the durability and the mechanical properties, including compressive and flexural strengths, of the locally compressed earth blocks manufactured from soil in Irbid, Jordan. Moreover, effect of volcanic tuff as new stabilizer material on properties of compressed earth block (CEB). Compressed earth block is a technique that was created to solve environmental and economic problems in construction sector. It is widespread in many countries around the world but hasn't been used in Jordan yet. Design/methodology/approach: 9 mixtures were carried out. One of this mixture is the control mix, beside other mixtures were performed by replacing soil with 40%, 10%, 10%, of sand, volcanic tuff, and lime respectively. In addition, polypropylene fibre was used. After 28 days of curing, the CEB were dried in oven at 105°C for 24 hours then tested. Findings: Show that absorption and erosion were decreased when the lime used in the soil. On the other hand, the fibres presence significantly improved the durability and mechanical properties in all mixtures. Moreover, the higher compressive strength was obtained in the mixtures which contain lime only while the higher tensile strength was obtained in the mixtures which contain lime with sand replacement. The using of volcanic tuffs produced average compressive strength values. The reason is that in the presence of lime and pozzolana (volcanic tuff) reactions take place at low and slow rate at early ages. Research limitations/implications: volcanic tuff can produce favourable compressive strengths at later ages and this is a point of interest in the future work. Originality/value: Searching for a new material as stabilizer material that improves the properties of the compressed earth block (CEB).

Opis:
Purpose: This work aims to study the effect of hard segments (HS) content on the thermal, morphological and mechanical properties of polyurethane polymers based on 1.5 pentanediol chain extenders. Design/methodology/approach: Two comparable series of polyurethanes were synthesised including homo-polyurethane (Homo-PU) and copolyurethane (Co-PU). The Homo-PU consists of 100% wt. of hard segments (HS). The Co-PU composes of 30%wt. of soft segments (SS) using a poly(ethylene glycol)-block-poly(propylene glycol)-block-poly(ethylene glycol) material. The effect of hard segments content on the morphology of Homo-PU and Co-PU was also studied. Findings: The Homo-PU and Co-PU materials show three distinct degradation steps with the higher thermal stability of the Co-PU compared to the Homo-PU. Enthalpy of fusion (ΔHM) and heat capacity (ΔCP) of polyurethane (PU) samples decrease with decreasing HS content. In the cooling cycle, the higher exothermic peak of crystallization is observed in the Co-PU. In contrast, the cold crystalline peak is observed in the 2nd heating cycle of the Homo-PU. Melting temperature (TM) increases with increasing SS content. Glass transition temperature (Tg) of PU samples shifts to higher temperature with increasing HS content. Storage modulus (E’) of the Co-PU is higher than E’ of the Homo-PU. All N-H groups in PU samples are hydrogen-bonded, whilst most of the C=O groups are hydrogen-bonded. The degree of hydrogen bonding in PU samples decreases with decreasing HS content. The Homo-PU shows better hardness than the Co-PU and higher brittleness at low temperature. WAXS results of the Homo-PU display better crystallinity compared to the Co-PU. Research limitations/implications: The main challenge in this work was how to synthesis Thermoplastic polyurethanes (TPUs) with specific properties to compete other common polymer such as Polyamides (PA) and Polypropylene (PP). Practical implications: Thermoplastic polyurethanes (TPUs) can be used in various application such as backageing, foot, automobiles and constructions. Originality/value: A new type of TPUs that synthesized using different type of chain extender (1.5 pentanediol). Two different types of TPUs were synthesized one contained 30% SS and 70% HS and a second one contained 100% HS.
