Opis:
The thermal stability of enzyme-based biosensors is crucial in economic feasibility. In this study, thermal deactivation profiles of catalase Aspergillus niger were obtained at different temperatures in the range of 35° C to 70° C. It has been shown that the thermal deactivation of catalase Aspergillus niger follows the first-order model. The half-life time t 1/2 of catalase Aspergillus niger at pH 7.0 and the temperature of 35° C and 70° C were 197 h and 1.3 h respectively. Additionally, t 1/2 of catalase Aspergillus niger at the temperature of 5° C was calculated 58 months. Thermodynamic parameters the change in enthalpy ΔH*, the change in entropy ΔS* and the change Gibbs free energy ΔG* for the deactivation of catalase at different temperatures in the range of 35° C to 70° C were estimated. Catalase Aspergillus niger is predisposed to be used in biosensors by thermodynamics parameters obtained.

Opis:
The paper presents a comparative analysis to determine the optimal temperatures and the activationenergies for various origin endo-inulinases fromAspergillus niger. The parameters were estimatedbased on the literature of the activity curves vs. temperature for hydrolysis of inulin. It was assumedthat both the hydrolysis reaction process and the deactivation process of endo-inulinase were first-orderreactions by the enzyme concentration. A mathematical model describing the effect of temperature onendo-inulinases fromAspergillus nigeractivity was used. Based on the comparison analysis, valuesof the activation energies Ea were in the range from23:53+/-3:20kJ/mol to50:66+/-3:61kJ/mol, thedeactivation energies Ed were in the range from88:42+/-5:03kJ/mol to142:87+/-2:75kJ/mol andthe optimum temperatures Topt were obtained in the range from317:12+/-0:83K to332:55+/-0:72forendo-inulinase A. niger.

Opis:
Effective diffusion coefficients (De) for different heavy-metal salts: Cd, Pb, Zn in calcium alginate beads were determined. Their values depend on the metal type, anion from the metal salt, and the alginate content in the beads. The results of calculations indicate a decrease in the values of De, caused by an increase in the alginate content in the alginate sorbent beads. This is in agreement with the mechanism of the diffusion process taking place in porous carriers. Experimental data were found to be in good agreement with the mathematical model, as indicated by high values of the correlation coefficient.

Opis:
The paper presents the use of modified silica gel for the production of immobilized trypsin from bovine pancreas. Silica gel was modified with 3-aminopropyltriethoxysilane, followed by glutaraldehyde. The influence of stirring time on activity of the prepared biocatalyst was determined for individual stages of the modification. Activity of both native and immobilized trypsin was measured using Kunitz method. At the temperature of 55℃ and pH 7.6 native and immobilized trypsin onto modified silica gel indicate optimum activity. The influence of multiple recycling and storage time on activity of immobilized trypsin was tested. After fourteen days of storage at the temperature of 4℃ immobilized trypsin exhibits 75% of its initial activity.

Opis:
The effect of supplementation of medium with rapeseed meal (RM) on production of biotechnologically important enzymes was investigated in submerged cultures of the white rot fungus Cerrena unicolor. The addition of RM (3.5% w/v) distinctly stimulated the activities of laccase, chitinase, and β-glucosidase. As compared to the control, the activities of chitinase, β-glucosidase, and laccase in the RM supplemented cultures were up to 4.1, 8.4, and 3.9 times higher, respectively. The results of the spectrophotometric and spectrofluorometric measurements were additionally confirmed by zymographic analysis of the samples. The level of sugars and phenolic compounds as well as the antioxidative ability of fungal preparations were also determined. The results obtained indicate that the submerged liquid fermentation of rapeseed meal can be proposed as an inexpensive and very effective method for biotechnological production of chitinase, β-glucosidase, and laccase by C. unicolor.
