Opis:
The SNIF-NMR method (site-specific natural isotope fractionation studied by Nuclear Magnetic Resonance) was used to examine the isotopic profile of glucoses derived from plants with different photosynthetic pathways. It is shown that the type of photosynthetic metabolism, either C3 (beet-root, orange, grape), C4 (maize, sugar-cane) or CAM (pineapple), exerts a strong influence on the deuterium distribution in the sugar molecules. The isotopic profile also depends, secondarily, on the physiological status of the precursor plant. Consequently, the isotopic fingerprint of glucose may be a rich source of information in mechanistic comparisons of metabolic pathways. Moreover, from an analytical point of view, it may provide complementary criteria with respect to the ethanol probe for origin inference of sugars.

In pursuing FHB resistance in wheat, 30 years of conventional breeding efforts in Eastern Canada have brought some progress. Substantial investment and the application in recent years of marker-assisted selection have to date, however, failed to produce agronomic lines that resist FHB as well as Sumai 3. We present here an alternative path, described as the systemic approach. Rather than seeking to introgress specific putative resistance genes, it subjects target germplasm to regimes of repeated cycles of multiple, interacting (biotic and abiotic) stresses in which desirable traits – not always adequately expressed in parental lines – are identified and selected. How can such a seemingly counterintuitive process work? The systemic approach views desired resistance as arising from the interactions of complex regulation mechanisms mediating how a host responds when a pathogen attacks. These constituents of resistance should thus not always be understood simply as discrete Mendelian units. In repeated rounds of selection, the systemic approach captures those rare individuals that embody optimal interactions of traits, and advances them as founders of lines that resist FHB more effectively than if selection focused on FHB alone. In Quebec, we have chosen to select wheat populations under combined pressure from barley yellow dwarf virus (BYDV) infection and FHB. Resistance to FHB and tolerance of BYDV are quantitative traits that interact. BYD increases both the direct losses from FHB and the production of mycotoxins. Selection under virus pressure, therefore, helps identify those individuals which express FHB resistance more effectively. Moreover, the correlates of virus tolerance (physiological efficiency, generalized stress tolerance and yield) point to those plants with better root traits, ability to produce biomass and yield stability. Together with numerous secondary criteria, such selection eliminates all but a few ‘winners’ in each round. Seen from a systemic perspective, the difficulty of identifying good progeny among descendants of crosses with Sumai 3 does not surprise. Deleterious linkages, pleiotropy and epistasis will usually combine in far from optimal expressions of the assembled genetic information. The systemic approach, by contrast, identifies in repeated cycles increasingly optimized expressions of genes, allowing all potential sources of resistance to be explored. Thus resistant lines can readily be derived from the crosses of susceptible parents, an objective rarely sought in conventional, focused approaches. Moreover, wheat plants respond to the systemic approach’s powerful stresses with enhanced epigenetic variation, raw material from which broader ranges of heritable traits can be selected. Germplasm that expresses a full range of attractive traits while resisting FHB as effectively as Sumai 3 can now be shown to be much more abundant than previously imagined. Perhaps this promise will entice more wheat workers to try a systemic approach...

Gullies developed on sandy lithologies are scarce. Such landforms have developed in the sandy deposits of the Cretaceous 'Utrillas' facies within the Segovia province of Central Spain. They appear at the slopes of a group of mesas and cuestas located at the edge of the northern piedmont of the Guadarrama Mountains of the Spanish Central System. The activity of the different geomorphic processes acting in these gullies, as well as their connectivity, are being characterized and quantified. This study was preceded by reconnaissance methods, whereas presently technologically advanced and more accurate techniques are being utilized. The new methods are applied in an experimental catchment (1.26 ha). They include: i) an automatic Reid-type (formerly termed Birkbeck) slot bedload sampler for continuous monitoring of bedload flux and for continuous sampling of bedload; siphons for sampling the suspended load; and a Parshall flume to monitor water discharge. Jointly, these instruments allow to study the fluvial dynamics at the catchment mouth; ii) topographic surveys undertaken by a Terrestrial Laser Scanner (TLS) to quantify the activity of gravitational, overland flow and fluvial processes in different sediment source areas within the gully basin; iii) micro runoff and erosion plots to monitor the infiltration and erosive response of different Hydrologic Response Units (HRU) comprising the interior of the catchment. This ensemble of novel methods has started providing patterns of sediment movement within the gully system.

The origin of plastids is best explained by endosymbiotic theory, which dates back to the early 1900s. Three lines of evidence based on protein import machineries and molecular phylogenies of eukaryote (host) and cyanobacterial (endosymbiont) genes point to a single origin of primary plastids, a unique and important event that successfully transferred two photosystems and oxygenic photosynthesis from prokaryotes to eukaryotes. The nature of the cyanobacterial lineage from which plastids originated has been a topic of investigation. Recent studies have focused on the branching position of the plastid lineage in the phylogeny based on cyanobacterial core genes, that is, genes shared by all cyanobacteria and plastids. These studies have delivered conflicting results, however. In addition, the core genes represent only a very small portion of cyanobacterial genomes and may not be a good proxy for the rest of the ancestral plastid genome. Information in plant nuclear genomes, where most genes that entered the eukaryotic lineage through acquisition from the plastid ancestor reside, suggests that heterocyst-forming cyanobacteria in Stanier’s sections IV and V are most similar to the plastid ancestor in terms of gene complement and sequence conservation, which is in agreement with models suggesting an important role of nitrogen fixation in symbioses involving cyanobacteria. Plastid origin is an ancient event that involved a prokaryotic symbiont and a eukaryotic host, organisms with different histories and genome evolutionary processes. The different modes of genome evolution in prokaryotes and eukaryotes bear upon our interpretations of plastid phylogeny.

Microbial amylases are enzymes produced by microorganisms to hydrolyze starch. There are three types of microbial amylases: alpha-amylase, beta-amylase and glucoamylase. Each of these amylases has a unique way of acting on starch to yield simple glucose monomers. Microorganisms, plants, and animals are sources of amylases, but much attention is given to microorganisms since the amylases produced by them have greater thermal stability and give rise to different sugar profiles, thus meeting industrial demands. Two major groups of microorganisms play pivotal role in amylase production: bacteria and fungi. Starch is the substrate used in amylase production. Between the two fermentation processes used in amylase production (i.e. submerged fermentation and solid state fermentation), the latter is more advantageous as it saves cost, generates little effluent, and has high volumetric productivity. Microbial amylases are greatly applied in pharmaceutical, food, chemical, paper and distilling industries.

This study focused on determining the amylase-producing ability of the intestinal microbes in cultured fresh water fishes – Oreochromis niloticus and Clarias gariepinus. The bacterial isolates were identified on the basis of standard cultural, morphological and biochemical characteristics. The amylase production ability of the bacterial isolates was determined using starch agar. The mean viable count of the intestinal microbes ranged from 1.2 × 105 CFU/ml to 7.1 × 105 CFU/ml for tilapias (Oreochromis niloticus) and from 2.0 × 104 CFU/ml to 8.9 × 104 CFU/ml for catfishes (Clarias gariepinus). Staphylococcus and Micrococcus were predominant for both tilapias and catfishes. Out of 24 isolates, 21 were amylase producers. These included the following bacteria genera: Bacillus, Micrococcus, and Staphylococcus. These results strongly suggest that intestinal microbes play a pivotal role in the digestion of starch in cultured freshwater fishes and should be explored for industrial amylase production.

The need to maintain economic growth and development is, in the current conditions, coupled with the need to maintain a quality environment and quality of life. Individual economies are transforming their production and consumption patterns towards a circular economy, carbon neutrality and the elimination of the negative impacts of climate change, with the aim of conserving resources, in the light of the principles of sustainable development. In this paper we focus on the green growth and the green economy, as a current and the preferred concepts on the road to sustainability. The main objective of this paper is to review the theoretical approaches to the concepts of green growth and green economy. To achieve the main objective, an analysis of documents and sources declaring different approaches and ways to presenting the green growth and green economy was used. This article presents views on the common points of convergence, divergence and directions of development of green concepts, using the literature sources.
Konieczność utrzymania wzrostu oraz rozwoju gospodarczego łączy się w obecnych warunkach z koniecznością zachowania jakości środowiska i jakości życia. Poszczególne gospodarki przekształcają swoje wzorce produkcji i konsumpcji w kierunku gospodarki o obiegu zamkniętym, neutralności węglowej i eliminacji negatywnych skutków zmian klimatu, mając na celu ochronę zasobów, w świetle zasad zrównoważonego rozwoju. W niniejszym artykule skupiamy się na zielonym wzroście i zielonej gospodarce, jako aktualnej i preferowanej koncepcji na drodze do zrównoważonego rozwoju. Głównym celem artykułu jest przegląd teoretycznych podejść do koncepcji zielonego wzrostu oraz zielonej gospodarki. Aby osiągnąć główny cel, wykorzystano analizę dokumentów oraz źródeł deklarujących różne podejścia i sposoby przedstawiania zielonego wzrostu oraz zielonej gospodarki. Artykuł przedstawia poglądy na temat wspólnych punktów zbieżności, rozbieżności oraz kierunków rozwoju koncepcji zieleni, korzystając ze źródeł literaturowych.
