    Glucagon-like peptide 1 receptor agonists (GLP-1RAs) are relatively new pharmacological agents used to normalize glucose level in type 2 diabetes. Recently, GLP-1RAs have been approved for the treatment of obesity to reduce body weight in non-diabetic patients. The extra-pancre-atic effects of GLP-1RAs, as well as their molecular mechanism of action, are still poorly understood. Thus this study was aimed to verify the hypothesis that the mechanism of action of the GLP-1RAs involves mitochondria and that GLP-1RAs administration can improve mitochondrial functions. For this purpose, preadipocytes CHUBS7 were differentiated to mature adipocytes and then stimulated with GLP-1RA, exendin-4 at 100 nM for 24 h. Oxygen consumption rates, mitochondrial membrane potential, intracellular ATP (adenosine triphosphate) level, SIRT1 and SIRT3 gene expression and the histone deacetylases' activity were measured. Exendin-4 was found to uncouple mitochondrial electron transport from ATP synthesis, slightly decreasing mitochondrial membrane potential in mature adipocytes. Routine respiration and uncoupled oxy- gen consumption rates were higher in exendin-4 treated adipocytes than in the non-treated cells. The ATP level remained unchanged. Exendin-4 enhanced SIRT1 and SIRT3 genes expression. Histone deacetylases' activity in the nuclear fraction was not affected by exendin-4, although the activity of class III histone deacetylases was increased. All of the effects on mitochondrial bioenergetics induced by exendin-4 were abolished by addition of glucagon-like peptide 1 receptor antagonist. In conclusion, exendin-4 activates the sirtuin pathway and increases energy expenditure in human adipocytes. Our results suggest another mechanism that may be responsible for body weight reduction observed in patients using GLP-1RAs.

    The article focuses on human resources management and motivational paradoxes explanations. A presentation of complexity of motivational processes and the identification of contradictions in motives of human's actions, as well as a consideration of paradoxical thinking consequences for the human resource management and motivation function realization are a crucial aims of this article. The management literature review also allowed to identify a couple of strategies of managing paradoxes, which are also pointed out and characterized in this text.

    Le motif de fuseau fait partie d’un vieux répertoire des figurations dans l’iconographie sépulcrale. Nous le retrouvons sur les peintures égyptiennes, sur la céramique grecque. Il apparaît aussi sur les sarcophages, sur les peintures et sur les mosaiques d’Asie Mineure, Palmyre, Grece et Rome. Dans l’iconographie antique, tenu dans la main ou déposé avec d’autres objets, le fuseau représente un attribut féminin. Il symbolise un engagement d’une femme au foyer, son soin et sa vie laborieuse. Cependant sur les représentations grecques et romaines, il apparaît dans les mains des Moires qui accompagnent les mortels et les dieux au moment de leur naissance et de leur mort. Les Moires des reliefs de sarcophages romains, tissant avec le fuseau et la quenouille, sont présentées aussi dans les scenes de la création de l’homme par Prométhée. Dans ce contexte, le fuseau prend une signification nouvelle et plus profonde. Il n’est plus qu’un attribut des femmes, leur servant dans des activités domestiques, mais un outil symbolisant le filage du destin et surtout l’inévitabilité de la mort et la renaissance de la vie. Depuis Veme siecle apres Jésus Christ, l’iconographie paléochrétienne reprend le motif de fuseau, comme symbole du travail féminin, accompli pour le bien propre, de son mari et de son Dieu (Tb 2, 11-12 ; Prov 31, 13-19) en le situant dans la scene de l’Annonciation. Les représentations mariales paléochrétiennes qui puisent a cette scene gardent un typique pour l’art antique schéma de personnages. La fileuse – ici la Vierge Marie - occupée du tissage, est assise ou debout et s’approche d’elle une femme ou un homme – dans cette scene l’Ange Gabriel. Dans ce groupe, une peinture, découverte a Vieille Dongola, montrant la Vierge Marie tissant et allaitant l’Enfant Jésus, constitue une représentation unique. Car la présence de l’Enfant dont la tete est entourée du nimbe cruciforme fait qu’il est difficile de le ranger parmi les scenes classiques d’Annonciation. Dans ce cas, il s’avere plus important la présence du fuseau qu’il faut expliquer en relation avec une symbolique antique riche et diverse. Le motif de fuseau, de tissage et la présence du nimbe cruciforme, démontrés sur cette peinture, peuvent faire penser a l’incarnation et a la mort de Jésus (en rapport avec la scene apocryphiques du tissage et de la déchirure de la pourpre). Ainsi, comme l’écrit saint Augustin: «ce qui fut accroché sur la quenouille, allait s’accomplir» (la mort de Jésus?) et «ce qui fut enroulé sur le fuseau s’accomplit déja» (l’incarnation?) (Sermo 37; PL 38, col. 227; trad. d’apres le texte lat. R. Grela). Donc, Mere de Dieu aurait-elle filé le destin sur le fuseau, comme le faisaient les Moires? L’art paléochrétien puise a l’art antique la façon de démontrer le fuseau en l’approfondissant sa signification d’une nouvelle symbolique.
