The measurement of the strength of bonds between biomaterials and cells is a major challenge in biotribology since it allows for the identification of different species in adhesion phenomena. Biomaterials, such as diamond-like carbon (DLC), titanium, and titanium nitride, seem to be good candidates for future blood-contact applications. These materials were deposited as thin films by the hybrid pulsed laser deposition (PLD) technique to examine the influence of such surfaces on cell behavior. The biomaterial examinations were performed in static conditions with red blood cells and then subjected to a dynamical test to observe the cell detachment kinetics. The tests revealed differences in behavior with respect to the applied coating material. The strongest cell-biomaterial interaction was observed for the carbon-based materials compared to the titanium and titanium nitride. Among many tests, a radial flow interaction analysis gives the opportunity to analyze cell adhesion to the applied material with the high accuracy. Analysis of concentrates helped to select materials for further dynamic tests on blood using an aortic flow simulator. In this case, the platelet adhesion to the surface and their degree of activation was analyzed. The quality of the selected coating was tested using a scratch test. The analyses of the microstructure were done using high resolution transmission electron microscopy. The phase composition and the residual stress were analyzed using X-ray diffraction methods.

When designing new biomaterials for tissue contact devices it is important to consider their architecture as it affects different cell response. Surface modification of tubular structures requires the use of different techniques than in the case of flat samples. Similarly, analytical techniques also need to be adapted to the specific shape of substrate. For blood contacting devices this issue is critical because of shear forces generated by fluid flow and responsible for blood components activation. This necessitates the use of diagnostic techniques dedicated for material analysis in dynamic conditions in order to simulate physiological conditions. In the frame of the work, the flat samples as well as tube like elements were considered. The flat samples were prepared for basic research. Based on the results of the basic research the thin coatings were selected for the internal side of the tube like elements which have been analysed in contact with blood using blood flow simulator. The cross section of the coating-substrate interaction was tested using transmission electron microscopy. The attachment of cells to coatings was determined by radial flow chamber. Hemocompatible analysis was carried out in two ways. The quality of the blood after the dynamic test was analysed using flow cytometry. In this case the aggregates formation, platelet consumption and apoptosis derived microparticles were considered. The amount of cells adhered to the materials surfaces was determined by confocal laser microscopy.

Since 1991, the Foundation for Cardiac Surgery Development in Zabrze has been implementing research on the artificial heart. In 1995, an artificial ventricle, POLVAD, was implanted to a patient, and in 1998, the prototype of a clinical controller, POLPDU-401, was created. A further development of the studies on an implantable artificial heart requires an integrated approach and an application of advanced methods of materials sciences in order to develop new materials suitable for the contact with blood, as well as to apply a multilateral biomedical diagnostics in hydrodynamic conditions. The estimation of the cell–material interaction plays an important role in the biomaterial design. An analysis of the influence of the carbon content in titanium nitride on the biological and biophysical properties of biomaterial coatings was studied. The cell-material reactions were considered in dynamic and static conditions. Three groups of materials were under examinations – titanium nitride (TiN), as well as titanium carbonitride with a low and high carbon content – of which the best properties were observed for TiN. We found a strong influence of the stoichiometry of TiN (atomic ratio of Ti/N) on the biocompatibility. A non-stoichiometric TiN could have a negative influence on the surrounding tissue.

The TiN/Si3N4 coatings were deposited by reactive single target magnetron sputtering. The TiSi10 and TiSi6 targets were prepared from titanium and silicon elemental powders. The transmission electron microscopy helped to confirm the nanocomposite nanocrystalline TIN/ amorphous Si3N4 microstructure of coatings. The experiments helped to determine the critical coatings thickness at close to 1 žm. The micro-hardness of such coatings deposited on high speed steel was estimated at 11.9 - 13 GPa and characterized by critical loading in the range of 13 - 14 N. The measurements of chemical compositions of coatings showed that the ratio of silicon to titanium was at least twice as high as those in the targets, what might have lowered the coatings mechanical properties.
Powłoki TiN/Si3N4 naniesiono techniką rozpylania magnetronowego przy użyciu targetu o składzie TiSi10 lub TiSi6 (% at.). Targety te zostały przygotowane na drodze spiekania na gorąco proszków krzemu i tytanu. Analizę mikrostruktury nanokompozytowych nanokrystalicznych powłok TiN/Si3N4 o krytycznej grubości delaminacji zbliżonej do l žm, przeprowadzono przy użyciu mikroskopu transmisyjnego. Mikrotwardość powłok na podłożu ze stali szybkotnącej określono na poziomie 11.9 - 13 GPa. Analiza składu chemicznego powłok wykonana techniką EDS wykazała, że stosunek zawartości krzemu do tytanu jest dwa razy większy niż w targetach, co może stanowić przyczynę ich niższej od oczekiwań twardości.

Opis:
Seeding of cells on functional, biocompatible scaffolds is a crucial step in achieving the desired engineered tissue. The authors show the constructional solutions of the Nd:YAG pulse laser system with the Q-switch modulation for direct and interferential shaping of the surface of biocompatible materials. The two-channel interference system is distinguished by high control, simplicity and repetitiveness regarding laser energy level and dimensions of the surface structures. The experiments were conducted on hard, biocompatible substrates of amorphous carbon (diamond-like carbon, DLC) and were preliminarily tested on smooth muscle cell depositions.

The paper presents the main achievements of the author on the development of blood contacting materials. The main objective of the work is to elaborate materials dedicated for the heart support systems. Appropriately designed biomaterial surfaces enable fully controlled cellular differentiation, proliferation, and even restoration of the tissue structure on solids. The paper presents two approaches to modify the surface, which can control the life processes of tissue. The first solution considers the topography in the form of cell niches. The main objective of the study is a modified surface of thin films deposited on the polymer substrate constituting the microenvironment for the cells caused by residual stress and optimized stiffness of the surface using the plasma methods. The research hypothesis was the plasma surface modification method generating a controlled contribution of residual stress in the coating affect the surface topography in the form of nano- wrinkles similar to the niches in the tissue environment. Topography and stiffness of the surface coating allows the targeted cellular differentiation. The properly formed surface topography effectively inhibits blood clotting processes. The second solution considers implementation of self-organizing feature of extracellular matrix like coatings and selective cell mobilization. The multiscale analysis and phenomenologic description were performed to experimental research. For this purpose, the deposition method was based on electrostatic interactions in polyelectrolytes. This type of cell-polymer structure imitate the native structures.
Praca przedstawia najważniejsze osiągniecia autora dotyczące rozwoju materiałów do kontaktu z krwią. Głównym celem prowadzonych prac są materiały o przeznaczeniu w komorach wspomagania serca. Odpowiednio zaprojektowana powierzchnia biomateriału umożliwia w pełni kontrolowane różnicowanie komórkowe, proliferację i nawet odtworzenie struktury tkanki na ciele stałym. W pracy przedstawiono dwa podejścia modyfikacji powierzchniowej, które pozwalają sterować procesami życiowymi tkanki. Pierwszym rozwiązaniem są powłoki o topografii nisz komórkowych. Celem głównym badań jest zmodyfikowana powierzchnia materiałów cienkowarstwowych nałożonych na podłoże polimerowe stanowiąca mikrośrodowisko dla wychwytu i kontrolowanego różnicowania komórek uzyskane przez odpowiedni udział naprężeń własnych i zoptymalizowaną sztywność powierzchniową. Ukierunkowana mikrostrukturą i właściwościami powierzchniowymi monowarstwa hamuje procesy wykrzepiania krwi. Drugie rozwiązanie dotyczy powłok o strukturze macierzy zewnątrzkomórkowej. Celem badań jest wieloskalowa analiza i opis fenomenologiczny samoorganizujących się powłok z funkcją selektywnej mobilizacji komórkowej. Jest to nowoczesne podejście na poziomie badań eksperymentalnych, dotyczące wytworzenia materiałów biologicznie kompozytowych i ich kompleksowej analizy z wykorzystaniem metod inżynierii materiałowej i inżynierii biomedycznej. Do tego celu została zastosowana metoda oparta o oddziaływania elektrostatyczne. Tego typu komórkowo-polimerowa struktura imituje struktury natywne tkanek.

The work is a review of the methods of the surface modification performed by the authors dedicated for for cardiac support system. It presents the evolution of designing the surface dedicated to direct contact with blood. Initially thin and ultrathin coatings were developed. They were designed as a blood-polymer barrier. The pneumatic heart assist devices are made of a medical grade polyurethane. A major milestone was to create advanced ceramic thin films expressing the flexible effects deposited by physical techniques. Coatings have evolved. Another milestone was the surface reproducing the microenvironment to capture progenitor cells from the bloodstream. Thin coatings were prepared, using methods of ion been, controlled residual stresses were introduced. Wrinkles appeared without cracking. This enabled taking control over the process of cell differentiation. Alternatively, the tissue inspired structure resulted of the coating in the form of extracellular matrix. The outer surface was modified with synthetic materials. This enabled the effective proteins docking to induce cell growth, recreating the luminal side of the blood vessel. Coagulation processes have been slowed down. In addition, it was found pro-angiogenic effect.

Reconstruction of the cardiovascular system: The aim of the work is to minimize thrombus formation life threatening in the pulsatile heart assist chamber, by the use of a new biomimetic heart valve made on the basis metal-polymer composites. The heart prostheses are dedicated to support the heart of patients with late heart disease failure. They are dedicated to therapy related to self-regeneration or as a bridge for transplantation. In the future, they will be able to help treatment with gene therapy for myocardial infarction. Systems with valves mechanically generate plaque activation by shear stress. This is due to the narrow gap that is between the petal and the ring. Composite materials and a new valve design can minimize this problem. Injected polyurethane with a metallic, titanium bonded insert optimal microscale flexibility with macro-stiffness for ensuring appropriate mechanical functions of the valve.

Seeding of cells on functional, biocompatible scaffolds is a crucial step in achievement the desired engineered tissue. In the present study, a pulsed laser modification onto inorganic substrate was made to promote endothelium cells migration and spread. Presented scaffolds were fabricated on carbon and titanium based coatings. Fabricated films provided very good mechanical properties together with a chemical stability preservation. The substrate modification consisted of grid-like template fabrication of micrometer size meshes. The microstructure analysis of laser traces revealed the grain size increase in the zone of laser beam interaction, which exerts an influence on a surface topography. Endothelium cells locomotion was observed within 10 day time period. As a result it was shown that the modified area enhanced cells adhesion with a preferred static behavior. The performed research work improved our understanding on the pulsed laser ablation process and template size influence on cells spatial arrangement. It constituted an important step towards fabrication of inorganic, biocompatible scaffolds for successful substrate endothelialization.

Basins without outlets are natural storage reservoirs that can hold rainwater surpluses in a catchment for a time. In Poland, they have been an object of detailed fieldwork in the upper Parsęta and Potok Oliwski catchments, where they occupy a substantial proportion of the area. It was found that the prevailing weather conditions had a notable effect on the chemical composition of the surface water in the kettle ponds examined. The basic method of indoor work was an analysis of topographic maps and the statistical processing of the material collected. The fieldwork included a cycle of measurements in the hydrological years 2006 and 2007 and for 8 kettle ponds in the upper Potok Oliwski catchment. Hydrochemical mapping was carried out for 20 kettle ponds located in two subcatchments of the upper Parsęta catchment and for 8 kettle ponds in the upper Potok Oliwski catchment. Ion concentrations in surface water were determined with the help of analytic methods complying with the ISO and EPA standards. On the basis of the results obtained for those two geographically and hydrographically distinct catchments, great hydrochemical diversity was recorded, visible not only in a wide range of ion concentrations or such indices as specific electrolytic conductivity and pH, but also in the hydrogeochemical types found among the surface waters of individual basins. Because of their isolation from the horizontal inflow, the chief cause of the chemical differences in their waters is the vertical exchange.

Structure changes in laser constituted surface layers of aluminium alloys of the systems: Al-Zn, Al-Fe, Al-Fe-Ni, Al-Si were examined. Significant structure refinements and even formation of characteristic precipitates for rapidly quenched microstructure were found. Fine martensitic microstructure was observed in constructional steels of the types: X45CrSi92 (H9S2) and C50 (50G) subject to a laser modification. The study of influence of the chemical composition on the solidification structure and hardness was carried out on a steel 45, as well as on a high speed steel M2 (SW7M), by laser alloying using carbides of the types: VC, Mo$\text{}_{2}$C and the borides of the types: CrB, VB$\text{}_{2}$, B$\text{}_{4}$C. Thermodynamic calculations of phase diagrams were made to predict the changes of the constitution due to laser alloying. A comparison of the solidification structures and the calculated phase diagrams revealed a good agreement, especially for carbides. Boride alloying led to very complex microstructures consisting of various metallic and intermetallic phases.

The nanoparticles of CH3NH3PbBr3 hybrid perovskites were synthesized. These perovskite nanoparticles we embedded in polymethyl methacrylate (PMMA) in order to obtain the composite, which we used as light converter for silicon solar cells. It was shown that the composite emit the light with the intensity maximum at about 527 nm when exited by a short wavelength (300÷450 nm) of light. The silicon solar cells were used to examine the effect of down-conversion (DC) process by perovskite nanoparticles embedded in PMMA. For experiments, two groups of monocrystalline silicon solar cells were used. The first one included the solar cells without surface texturization and antireflection coating. The second one included the commercial cells with surface texturization and antireflection coating. In every series of the cells one part of the cells were covered by composite (CH3NH3PbBr3 in PMMA) layer and second part of cells by pure PMMA for comparison. It was shown that External Quantum Efficiency EQE of the photovoltaic cells covered by composite (CH3NH3PbBr3 in PMMA) layer was improved in both group of the cells but unfortunately the Internal Quantum Efficiency was reduced. This reduction was caused by high absorption of the short wavelength light and reabsorption of the luminescence light. Therefore, the CH3NH3PbBr3 perovskite nanoparticles embedded in PMMA matrix were unable to increase silicon solar cell efficiency in the tested systems.
