Opis:
Vaccination is a most cost-effective way of controlling infectious diseases in fish. However, some vaccination techniques when applied to hatchery conditions are not as effective as we expect them to be. Modern molecular biology techniques offer a number of opportunities for improving existing bacterial or viral vaccines or creating new ones. One of the most promising trends in vaccinology is development of DNA vaccination. DNA vaccines are based on the gene encoding specific antigen, which is expressed in vaccinated organism and induces the host immune system. DNA vaccines, compared to conventional vaccines, have many advantages including ability to trigger wider immune response, bigger stability and possibility of large-scale production. To date, there are several reports indicating effectiveness of DNA vaccines used against fish pathogens.

The usability of surfactant-aided ultrafiltration for removal of fluoride ions from aqueous solutions has been investigated. The experiments were conducted with the use of cellulose and polyethersulfone membranes. Fluoride solutions containing 10 and 100 mg F/dm3 and cationic surfactants: octadecylamine acetate (ODA) and hexadecylpyridium chloride (CPC) were used in the experiments. The concentration of surfactants amounted to 320–960 mg/dm3. Ultrafiltration process was run under a trans-membrane pressure of 0.2 MPa. In the course of experiments, the effects of initial fluoride concentration, surfactant concentration and membrane type on the fluoride retention coefficient as well as the permeate flux were studied. It was found that the increase of the surfactant concentration effected in improvement of the separation efficiency. On contrary, the permeate flux decreased upon increase of the surfactant concentration. The process efficiency was also strongly influenced by the membrane and surfactant type. The permissible concentration of fluoride in the final product (below 1.5 mg F/dm3) was obtained for the test with initial solutions containing 10 mg F/dm3, polyethersulfone membrane and CPC con-centration equal to 644 and 966 mg/dm3.

In contrast to terrestrial animals, fish can ingest minerals from food or directly from water. Although micro- and macro-elements are needed in small quantities, they play a key role in many metabolic processes. Trace mineral and macromineral deficiencies may go unnoticed due to an absence of clear clinical symptoms in fish. Absorption processes are determined by various factors, mostly mineral concentrations in water but also other water parameters. The required dietary supplementation of macronutrients and micronutrients is very difficult to determine, and the amount of nutrients absorbed by fish from water is equally difficult to measure. Interactions between elements should also be taken into consideration. Many authors emphasize that phosphates may reduce the absorption of most micronutrients. Also, the current parameters of the water can affect the bioavailability. Some elements such as calcium, chlorine and sodium can be absorbed from ambient water in a quantity sufficient to meet the demand for this element. Other elements, however, require supplementation in a diet. For example, studies indicate the need for supplementation of phosphorus, zinc, copper and manganese. Most research concentrates on feedstuff as a source of micro- and macronutrients. Meanwhile, information concerning bioavailability of minerals directly from water is scarce. The aim of this study was to analyse literature from a different perspective, and concentrate on water as a source of minerals in fish nutrition. Measurements of water parameters such as temperature, pH, nitrate and nitrite levels and the amount of dissolved oxygen are a regular component of environmental control in fish farming. Determination of micro- and macro-element levels, however, remains uncommon in aquaculture. Measurements of these parameters could suggest which elements need to be supplemented and which are found in water in amounts that satisfy the needs of the fish.

Abstract Microsporidia are single-celled intracellular parasites which occur in a number of animals, both vertebrates and invertebrates. Several species of microsporidia can cause disease in humans in both immunocompromised and immunocompetent individuals. However, the sources of human infection and the routes of transmission of microsporidia have not yet been fully determined, although more and more researchers are of the opinion that microsporidia in humans is zoonotic. The aim of the presented study was to identify the most common microsporidial species in the droppings of raised and wild pigeons in Poland. A total of 139 collective samples of droppings (33 samples of droppings from feral pigeons and 106 samples from raised pigeons) were examined using conventional staining and molecular techniques. Using chromotrope staining and multiplex FISH techniques, the microsporidial spores were found in 12 (8.6%) of the 139 samples of droppings. The spores of Encephalitozoon hellem were detected in five samples of pigeon droppings (3.6%), spores of E. intestinalis in four samples of droppings (2.9%), while spores of E. cuniculi and E. bieneusi were only detected in two samples (1.4%). Furthermore, a mixed infection of E. bieneusi and E. cuniculi was found in a single sample of droppings (0.7%). The presence of microsporidial spores in droppings collected from raised pigeons indicates a risk of infection to humans, mainly pigeon fanciers.

W diecie Polaków chleb jest głównym źródłem błonnika, który wraz z kwasem fitynowym jest uznawany za składnik pożywienia upośledzający przyswajanie składników mineralnych. Celem pracy było określenie zależności zawartości błonnika pokarmowego i kwasu fitynowego w chlebie od odmiany pszenicy, z której otrzymano mąkę użytą do wypieku. Analizie poddano chleby biały i sitkowy, wypieczone z mąki otrzymanej z ziarna 7 odmian pszenicy orkisz i 1 odmiany pszenicy zwyczajnej. Oznaczono w nich zawartość całkowitą błonnika, frakcji rozpuszczalnej i nierozpuszczalnej oraz kwasu fitynowego. W chlebie orkiszowym jasnym zawartość błonnika pokarmowego (4,65 – 6,52 % s.m.) i kwasu fitynowego (0,01 – 0,02 mg·g-1) nie zależała od odmiany pszenicy, z której mąkę użyto do wypieku. Chleb orkiszowy sitkowy zawierał, w zależności od odmiany pszenicy, z której otrzymano mąkę, statystycznie istotne różne ilości błonnika całkowitego (9,55 – 11,63 % s.m.), jego frakcji nierozpuszczalnej i kwasu fitynowego (2,18 – 5,94 mg·g-1). Zawartość frakcji rozpuszczalnej błonnika pokarmowego nie zależała od odmiany pszenicy, z której wyprodukowano mąkę użytą do wypieku chleba sitkowego. Chleb orkiszowy okazał się lepszym źródłem błonnika pokarmowego niż chleb wypieczony z mąki pszenicy zwyczajnej.
In the diet of Poles, bread is a main source of dietary fibre, and together with the phytic acid, they are regarded as components deteriorating the assimilability of mineral elements. The objective of the study was to determine the dependence between the content of dietary fibre and phytic acid in bread and the wheat variety that was used to make flour for bread to be baked. White and brown breads were analyzed; they were baked of flour produced from grains of 7 spelt wheat varieties and of 1 common wheat variety. In the breads studied, total fibre, its soluble and insoluble fractions, and phytic acid content were determined. In the white spelt bread, the content of total dietary fibre (4.65 - 6.52 % of dry mass) and phytic acid (0.01 - 0.02 mg·g-1) did not depend on the wheat variety used to make flour for the breads baked and investigated. Depending on the wheat variety, amounts of total fibre (9.55 - 11.63 % of dry mass) and its insoluble fraction, and of phytic acid (2.18 - 5.94 mg·g-1) statistically significantly varied in the brown spelt breads investigated. The content of soluble dietary fibre fraction did not depend on the wheat variety of flour used to bake brown breads. The spelt bread appeared to be the best source of dietary fibre than the bread baked of common wheat flour.

The degradation of pentoxifylline (PTX) using H2O2, UV, H2O2/UV, Fenton and photo-Fenton processes has been examined in aqueous solutions. The influence of oxidation agent and initial PTX concentration, on H2O2, H2O2/UV, Fenton and photo-Fenton reactions was investigated. The addition of inorganic ions (Cl, NO3, SO42– and CO32–) on the degradation efficiency of PTX was tested for H2O2/UV, Fenton and photo-Fenton processeses. The results indicate that the photo-Fenton reaction is the most sufficient for PTX removal. The complete pharmaceutics decomposition is achieved after 5 min under optimized concentration of FeSO4,·7 H2O2 and H2O2. The degradation of PTX is inhibited in the presence of inorganic matter in H2O2/UV and Fenton reaction. In the photo-Fenton process addition of above compounds does not affect the reaction rate. Structures of ten products of photo-Fenton reaction have been proposed.

Bodies of water are very sensitive to environmental changes and play an important role in nutrient cycle in their catchment. Due to their natural features such as reduction of the land fill, they may cumulate organic matter from the catchment. Excess supply of nutrients to water bodies contributes to gradual water eutrophication. This process can occur rapidly, particularly in shallow lakes, which are more susceptible to degradation. This also results in deterioration of habitat conditions for many organisms and limits the economic use of water. Eutrophication may lead to gradual shallowing and shrinkage of water bodies, and, consequently, to disappearance of lakes. Thus, proper diagnosis and reduction in the amount of pollutants flowing into lakes are key factors to improve water quality and prevent their further degradation. Lake Głęboczek, like many other urban lakes year were powered by sewage. As a result, the tank has ceased to be an attractive element of urbanized land, and became the object of nuisance for residents, often acting health risks. For this reason, the lake has undergone numerous Głęboczek reclamation treatment and protection. The object of this study was rehabilitated water from the lake Głęboczek in Tuchola. Samples were taken 12-fold in 3 places during the period from March 2011 to February 2012. In the tested water was determined by the colorimetric method phosphorus, phosphate, nitrogen, nitrate, nitrite, and aluminum. Additionally, the pH and temperature. Lake Głęboczek is a reservoir, which is heavily influenced by human activities, direct and indirect. This is due to the central location of the water, the entire drainage area is densely populated and intensively exploited for agricultural purposes. One of the most important elements harboring an increase in trophic status of the lake, is its low average depth. In addition to the lake Głęboczek not affect any river, which prevents water exchange and causes the accumulation of pollutants. Both the results of their own and other authors suggest that the degree of eutrophication of the lake Głęboczek apparently decreased. The level of biogenic elements and their compounds was higher than the results obtained in 2001 (after restoration), which may indicate improper sewage and agricultural economy in the catchment of the lake. Oversize nutrient content indicate a renewed increase in trophic lake.

The aim of the study was to determine the effects of platelet-activating factor (PAF) on selected quality parameters of cryopreserved bull semen with reduced sperm motility used for artificial insemination. The aim of experiment 1 was to identify the optimal concentration of the phospholipid able to preserve sperm viability. Cryopreserved semen was treated with different PAF concentrations: 1x10-5M, 1x10-6M, 1x10-7M, 1x10-8M and 1x10-9M. The experiment demonstrated that PAF at concentration 1x10-9M increased most the sperm viability parameters (motility parameters, plasma membrane integrity and mitochondrial function) after 120 min of incubation of thawed semen at 37oC. Cryopreserved bull semen with reduced sperm motility (below 70%) was supplemented with PAF in a concentration of 1x10-9M. A statistically significant increase in sperm motility, percentage of linear motile spermatozoa and VSL value was observed after 120 min incubation of sperm with 1x10-9M PAF. Sperm supplementation with PAF also had positive effects on plasma membrane integrity and percentage of spermatozoa with preserved mitochondrial transmembrane potential, but the differences were not statistically significant. The results indicated positive effects of PAF supplementation at a concentration of 1x10-9M on the selected sperm quality parameters in cryopreserved bull semen with reduced motility.
