Opis:
His296 of Zymomonas mobilis levansucrase (EC 2.4.1.10) is crucial for the catalysis of the transfructosylation reaction. The three-dimensional structures of levansucrases revealed the His296 is involved in the substrate recognition and binding. In this study, nine mutants were created by site-directed mutagenesis, in which His296 was substituted with amino acids of different polarity, charge and length. The substitutions of His296 with Arg or Trp retained partial hydrolysis and transfructosylation activities. The positively charged Lys substitution resulted in a 2.5-fold increase of sucrose hydrolysis. Substitutions with short (Cys or Ser), negatively charged (Glu) or polar (Tyr) amino acids virtually abolished both the activities. Analysis of transfructosylation products indicated that the mutants synthesized different oligosaccharides, suggesting that amino acid substitutions of His296 strongly affected both the enzyme activity and transfructosylation products.

Objectives This study’s objective was to examine association between sleep duration and sleep quality, and metabolic syndrome (MetS) and its components in Taiwanese male police officers. Material and Methods Male police officers who underwent annual health examinations were invited to join the study and eventually a total of 796 subjects was included in it. The study subjects were divided into 5 groups according to the length (duration) of sleep: < 5, 5–5.9, 6–6.9, 7–7.9 and ≥ 8 h per day, and the global Pittsburgh Sleep Quality Index was used to categorize their sleep quality as good or poor. To analyze the association between sleep problems and MetS, adjusted odds ratio and respective 95% confidence intervals (CI) were computed. Results The prevalence of MetS in Taiwanese male police officers was 24.5%. Abdominal obesity had the highest proportion (36.2%) among 5 components of MetS. More than 1/2 of the police officers (52.3%) had poor sleep quality. Police officers with higher scores of sleep disturbances had a higher prevalence of MetS (p = 0.029) and abdominal obesity (p = 0.009). After adjusting for age, low-density lipoprotein cholesterol, smoking status, alcohol drinking habit, physical habitual exercise, snoring and type of shift work, the police officers who slept less than 5 h were 88% more likely to suffer from abdominal obesity than those who slept 7–7.9 h (95% CI: 1.01–3.5). Sleep quality was not associated with MetS and its components. Conclusions The police officers who slept less than 5 h were more likely to experience abdominal obesity in Taiwan, and those with higher scores of sleep disturbances had a higher prevalence of MetS and abdominal obesity. It is recommended that police officers with short sleep duration or sleep disturbances be screened for MetS and waist circumference in order to prevent cardiovascular diseases.

With the rapid development of communication technology, the Train-centric Communication-based Train Control (TcCBTC) system adopting the train-train communication mode to reduce the transmission link of control information, will become the direction of urban rail transit field development. At present, TcCBTC system is in the stage of key technology research and prototype development. Uncertain behavior in the process of system operation may lead to operation accidents. Therefore, before the system is put into use, it must undergo strict testing and security verification to ensure the safe and efficient operation of the system. In the paper, the formal modeling and quantitative analysis of train tracking operation under moving block are carried out. Firstly, the structure of TcCBTC system and the train tracking interval control strategy under moving block conditions are analyzed. The subsystem involved in train tracking and the uncertain factors in system operation are determined. Then, based on the Stochastic Hybrid Automata (SHA), a network of SHA model of train dynamics model, communication components and on-board controller in the process of train tracking is established, which can formally describe the uncertain environment in the process of system operation. UPPAAL-SMC is used to simulate the change curve of train position and speed during tracking, it is verified that the model meets the safety requirements in static environment. Finally, taking Statistical Model Checking (SMC) as the basis of safety analysis, the probability of train collision in uncertain environment is calculated. The results show that after accurately modeling the train tracking operation control mechanism through network of SHA, the SMC method can accurately calculate the probability of train rearend collision, which proves that the method has strong feasibility and effectiveness. Formal modeling and analysis of safety-critical system is very important, which enables designers to grasp the hidden dangers of the system in the design stage and safety evaluation stage of train control system, and further provides theoretical reference for the subsequent TcCBTC system design and development, practical application and related specification improvement.

Opis:
Annular cavitator with water injection is one of the key parts of the long-range supercavitating vehicle powered by water ramjet. In this paper, hydrodynamic properties of annular cavitator are studied numerically. The standard k ~ ĺ turbulence model is coupled with the Reynolds Averaged Navier-Stokes (RANS) equations to model the natural supercavitation process. The multiphase flow is considered as a mixture of varying density and modeled by the mass exchange equations. To fully understand this process, numerical simulations were performed for different annular cavitators. Computational Fluid Dynamics (CFD) results, including the pressure distribution and forces acting on the cavitator surface, mass flow and pressure loss of water injection, various supercavity sizes, were obtained and analyzed. The pressure distribution on the cavitator surface was significantly changed which resulted in 4 ~ 6% increase of the total drag of the vehicle. The results show that the mass flow and velocity of the injection water is mainly dependent on the tube size, while the total pressure loss of the water injection is mostly related to the outlet pressure. Supercavity generated by annular cavitator is smaller than that of the discal one. Based on the correlation analysis of the supercavity size and other factors, it could be concluded that the contraction of the cavity size is mainly caused by the diffluent mass flow of the water injection.

Osteosarcoma is the most common primary malignant bone tumor in adolescents and young adults. However, the involvement of serine/threonine phosphatase type 5 (PP5) in osteosarcoma remains unclear. The aim of this study was to evaluate the functional role of PP5 in osteosarcoma cells. Firstly, we found that PP5 is widely expressed in several human osteosarcoma cell lines. Then we used lentivirus-delivered siRNA to silence PP5 expression in Saos-2 and U2OS cell lines. Knockdown of endogenous PP5 expression by shRNA-expressing lentivirus significantly decreased the viability and proliferation of the osteosarcoma cells. Moreover, FACS analysis showed that knockdown of PP5 expression induced a significant arrest in the G0/G1 phase of the cell cycle, which was associated with the inhibition of cell proliferation. Therefore, knockdown of PP5 is likely to provide a novel alternative to targeted therapy of osteosarcoma and deserves further investigation.

Effective recognition of tags in the dynamic measurement system would significantly improve the reading performance of the tag group, but the blurred outline and appearance of tag images captured in motion seriously limit the effectiveness of the existing tag group recognition. Thus, this paper proposes passive tag group recognition in the dynamic environment based on motion blur estimation and improved YOLOv2. Firstly, blur angles are estimated with a Gabor filter, and blur lengths are estimated through nonlinear modelling of a Generalized Regression Neural Network (GRNN). Secondly, tag recognition based on YOLOv2 improved by a Gaussian algorithm is proposed. The features of the tag group are analyzed by the Gaussian algorithm, the region of interest of the dynamic tag is effectively framed, and the tag foreground is extracted; Secondly, the data set of tag groups are trained by the end-to-end YOLOv2 algorithm for secondary screening and recognition, and finally the specific locations of tags are framed to meet the effective identification of tag groups in different scenes. A considerable number of experiments illustrate that the fusion algorithm can significantly improve recognition accuracy. Combined with the reading distance, the research presented in this paper can more accurately optimize the three-dimensional structure of the tag group, improve the reading performance of the tag group, and avoid the interference and collision of tags in the communication channel. Compared with the previous template matching algorithm, the tag group recognition ability put forward in this paper is improved by at least 13.9%, and its reading performance is improved by at least 6.2% as shown in many experiments.

Objective: To explore the association of the methylation status of MGMT and hMLH1 with chromosome damage induced by vinyl chloride monomer (VCM). Materials and Methods: Methylation of MGMT and hMLH1 was measured in 101 VCM-exposed workers by methylation-specifi c PCR. Chromosome damage in peripheral blood lymphocytes was measured by the cytokinesis-block micronucleus assay. The subjects were divided into chromosome damaged and non-damaged groups based on the normal reference value of micronuclei frequencies determined for two control groups. Results: MGMT promoter methylation was detectable in 5 out of 49 chromosome damaged subjects, but not in the chromosome non-damaged subjects; there was a signifi cant difference in MGMT methylation between the two groups (p < 0.05). Conclusions: We detected aberrant promoter methylation of MGMT in a small number of chromosome damaged VCM-exposed workers, but not in the chromosome non-damaged subjects. This preliminary observation warrants further investigation in a larger study.
