"Elaioplasts" observed in Vanilla planifolia, Funkia Sieboldiana and Althaea rosea exhibit all the features characteristic of lipotubuloids earlier described in Ornithogalum umbellatum. They are cytoplasmic domains containing aggregates of lipid bodies connected with microtubules. The immunogold technique confirmed the presence of tubulin in this domain. These structures do not have their own membranes but they are surrounded by a tonoplast at the side of a vacuole since they invaginate into it. In cytoplasm of this domain among lipid bodies there are numerous ribosomes, ER cisternae and vesicles as well as few mitochondria, Golgi structures and microbodies while at older developmental stages there are also autolytic vacuoles. The fact that they are so similar to O. umbellatum lipotubuloids suggest that "elaioplasts" of V. planifolia, F. Sieboldiana and A. rosea can also be named lipotubuloids.

Opis:
Badania dotyczyły analizy właściwości substancji humusowych (SH) oraz kształtowania struktury kwasów huminowych (KH) gleb po wprowadzeniu preparatu wytworzonego na bazie (85%) odpadowego węgla brunatnego. Do badań wykorzystano materiał glebowy z wieloletnich statycznych doświadczeń polowych i mikropoletkowych, a jako źródło materii organicznej Rekulter preparat wyprodukowany na bazie węgla brunatnego odpadowego i węgla brunatnego typu ziemistego z Zagłębia Konińskiego (KWB Konin). Niższe wartości intensywności fluorescencji na widmach 3-D EEM dla KH z gleby rdzawej (piaszczystej) wskazują na większy wpływ materii organicznej z węgla brunatnego na gleby bardzo lekkie piaszczyste w porównaniu do gleb cięższych (brunatna kwaśna, płowa). Wyniki ponad 20-letnich doświadczeń polowych na glebie rdzawej potwierdziły korzystne, długotrwałe oddziaływanie węgla brunatnego na właściwości gleby oraz na stan materii organicznej z tego źródła.
Soil is non-renewable and a very complex natural resource which performs many vital functions: food (99%) and other biomass production, storage, filtration and transformation of many substances including water, carbon, nitrogen. Soil as a top layer of the earth's crust is the first receiver of anthropogenic contaminants, and may or may not serve as a natural barrier to protect groundwater against contamination from the surface. One of the most important indicators of soil quality is soil organic matter (SOM), which is an important building block for the soil structure, formation of stable aggregates, and is able to improve the infiltration rates and the storage capacity of water. SOM presents a major pool carbon in the biosphere and can act both as a source and sink for carbon dioxide and other greenhouse gases. Agricultural intensification and cultivation in general results in a serious decrease in SOM as compared to that in the natural vegetation. Presently, the protection of SOM is one of the main tasks in EU, because SOM in addition to its soil fertilizing function can act in the elimination of the soil contamination and carbon sequestration. In this context, humic substances (especially humic and fulvic acids) and cohesive fractions (clay, silt, silt clay, etc.) present in soil are favourable components. It is because of their high sorption capacity with respect to many contaminants, including heavy metals, which may results in their immobilisation and, consequently protect groundwater against contamination. Antropogenically transformed soils often do not contain humic substances (HS), thus their role as a natural barrier is decreased. Therefore, re-generation of HS through humification of organic matter from diverse sources added to soils, may be the way to re-built the protective character of soil barrier, and consequently to reduce environmental and/or health risks at areas under anthropopression. Transformation processes of brown coal applied to soil were described to evaluate the use of this material as an effective source of organic matter and HS generation in degraded soils. The goal of this study was to discuss fine brown coal derived preparation as effective sources of organic matter (and humic substances) especially in soils that are anthropogenically transformed. The research concerned the analysis of properties of HS as well as formations of humic acids (HAs) in soils after addition brown coal. Soil materials used in the research was collected from the long term field experiments and microplots, as the source of organic matter the Rekulter that is based (85%) on brown coal (soft smudge type) obtained from the Konin Basin deposits preparation was used. For qualitative characteristic of SOM, particularly, in HAs, modern analytical technique such as fluorescence intensity (FI) for 3-D EEM spectra was used. The lowest fluorescence intensity (FI) for 3-D EEM spectra were observed in the case of HAs extracted from rusty soil. The 20-years research results carried out on rusty soil (sandy) confirmed long-term effect of brown coal on soil properties as well as on organic matter that originated from this source.

Background. The transformation period in Poland is associated with a set of factors seen as ‘socio-economic stress’, which unfavourably influenced cancer treatment and slowed down the progress of the Polish cancer care in the 90’s. These outcomes in many aspects of cancer care may be experienced till today. The results of the international EUROCARE and CONCORD studies based on European data prove evidence that there is a substantial potential for improvement of low 5-year survival rates in Poland. Since high survivals are related to notably efficient health care system, therefore, to improve organization and treatment methods seems to be one of the most important directions of change in the Polish health care system. Till today, cancer care in Poland is based on a network outlined by Professor Koszarowski in the middle of the last century, and is a solid foundation for the contemporary project of the Comprehensive Cancer Care Network (CCCN) proposed in the frame of CanCon Project. Objective. Analysis of the structure of health care system and the changes introduced within the network of oncology in Poland since the beginning of the post-commuinist socio-economic transformation in 1989. Materials and Methods. This study was conducted based on the CanCon methods aimed at reviewing specialist literature and collecting meaningful experiences of European countries in cancer care, including the main legal regulations. Results. The analysis provided evidence that the political situation and the economic crisis of the Transformation period disintegrated the cancer care and resulted in low 5-year survival rates. A step forward in increasing efficiency of the cancer treatment care was a proposal of the ’Quick Oncological Therapy’ together with one more attempt to organize a CCCN. With this paper the Authors contribute to the CanCon Project by exploration, analysis and discussion of the cancer network in Poland as an example of existing net-like structures in Europe as well as by preparation of guidelines for constructing a contemporary CCCN. Conclusions. (1) ‘Socio-economic’ stress adversely affected the efficiency of oncological treatment, both by reducing safety and slowing down the development of modern oncology. (2) Changing the current system into the contemporary form - CCCN could be an important step forward to optimise the oncological health care in Poland. (3) Introduction of the mandatory monitoring of organizational changes with the use of health standardized indicators could allow for the assessment of the effectiveness of implemented solutions and their impact on better prognosis for cancer patients. (4) Optimising the organization of the health care system is possible only by implementing necessary legislative corrections.
Wprowadzenie. Okres Transformacji w Polsce wiąże się z wieloma czynnikami postrzeganymi jako „stres społeczno- -gospodarczy”, które niekorzystnie wpłynęły na efekty leczenia nowotworów oraz spowolniły postęp w polskim lecznictwie onkologicznym w latach ‚90, co w wielu jego aspektach jest odczuwane do dziś. Wyniki międzynarodowych badań EUROCARE i CONCORD dowodzą, że wskaźniki 5-letnich przeżyć w Polsce mogą być znacząco wyższe. Wysokie wskaźniki 5-letnich przeżyć zależą od efektywnego systemu ochrony zdrowia, dlatego, poprawa organizacji i leczenia nowotworów, jest jednym z najważniejszych kierunków zmian w polskim systemie opieki zdrowotnej. System opieki onkologicznej w Polsce jest oparty na modelu sieci onkologicznej, której budowę w połowie ubiegłego stulecia rozpoczął Profesor Koszarowski. Stanowi on nadal solidny fundament do rozwoju nowoczesnej koncepcji sieci wielodyscyplinarnych centrów onkologii (ang. Comprehensive Cancer Care Network - CCCN) zaproponowanej w ramach projektu CanCon. Cel. Analiza struktury lecznictwa i zachodzących zmian w ramach sieci onkologicznej w Polsce po rozpoczęciu w 1989 postkomunistycznej transformacji społeczno-ekonomicznej po 1989 roku. Materiał i metody. Badanie zostało przeprowadzone w oparciu o metody przyjęte przez CanCon, tj. przegląd literatury specjalistycznej i analizę doświadczeń krajów europejskich w zakresie opieki onkologicznej, ze szczególnym uwzględnieniem polskich regulacji prawnych. Wyniki. Analiza wykazała, że sytuacja polityczna i kryzys gospodarczy w okresie Transformacji przyczyniły się do dezintegracji leczenia onkologicznego, a w efekcie złych wskaźników 5-letnich przeżyć. Analiza ta stanowi wkład Autorów do projektu CanCon poprzez analizę i omówienie sieci onkologicznej w Polsce, jako przykładu istniejących „podobnych do sieci” struktur lecznictwa w Europie. Zostanie ona wykorzystana przy opracowaniu wytycznych dotyczących współczesnej koncepcji CCCN. Postępem w zakresie organizacji leczenia onkologicznego w Polsce jest wdrożenie tzw. „Szybkiej Terapii Onkologicznej” oraz ponowna próba zorganizowania lecznictwa onkologicznego w ramach CCCN. Wnioski. (1) Stres społeczno-gospodarczy niekorzystnie wpływa na rokowanie u chorych na raka, zarówno poprzez obniżenie poczucia bezpieczeństwa socjalnego, jak i spowolnienie rozwoju nowoczesnej onkologii. (2) Zmiana obecnego systemu w nowoczesną formę - CCCN byłaby istotnym krokiem w stronę optymalizacji lecznictwa onkologicznego w Polsce. (3) Wprowadzenie obowiązku monitorowania zmian organizacyjnych, przy użyciu standaryzowanych wskaźników zdrowotnych w lecznictwie onkologicznym pozwoliłoby na ocenę skuteczności zastosowanych rozwiązań i ich wpływu na poprawę rokowania u pacjentów chorych na raka. (4) Optymalizacja organizacji systemu opieki zdrowotnej nie jest możliwa bez wprowadzenia koniecznych zmian legislacyjnych.

Opis:
The mechanical, chemical and biological properties of prostheses can be modified by thin coatings. Presently, carbon based coatings are often applied mainly to improve mechanical properties of the covered parts of the artificial prostheses. Particularly useful mechanical properties (such as good adhesion, high hardness, elasticity, etc.) are obtained when the coatings are formed by ion techniques such as the IBSD – Ion Beam Sputter Deposition and IBAD – Ion Beam Assisted Deposition. The chemical and biological properties of carbon coatings can be modified by metallic additives. In the ion-based techniques the metallic additives can be introduced into the coating in a few ways: by ion implantation of the chosen metal into the carbon substrate, by the IBAD technique working with two beams in the dual beam mode, or by using a complex sputtered target, composed of carbon and the selected metal. The final properties of the complex carbon – metal coating are related to the method used for its formation. In this work carbon–Ti and carbon–Ag coatings were investigated. All the coatings were formed by the IBSD or by DB IBAD techniques on UHMWPE and PU substrates. For each type of coatings the depth composition, chemical bonds and mechanical properties were determined. The morphology and thermal stability of the carbon–metal coatings were investigated mainly by the confocal dispersive Raman microspectrometry.

Opis:
Surface modification of medical implants is often required to improve their biocompatibility or, through bioactive properties of the surface material, facilitate its intergrowth with the living tissue. Bioactive-glass coatings can serve that purpose for the bone implants. We report a successful preparation of silicate-phosphate bioactive-glass coating on titanium substrate using the pulsed laser deposition method and present the coating characterization in terms of bonding configuration and chemical activity. The former was studied with high-resolution Raman microspectroscopy and revealed the presence of structural units responsible for the material's bioactivity. The bioactivity was also tested directly, in vitro, by soaking the samples in the simulated body fluid and examining the result with the Raman spectroscopy. The Raman spectrum, typical of hydroxyapatite was observed proving that the bone-like-material formed on the coating's surface.

The research, in 2004, was focused on the whole population of Anemone sylvestris (Snowdrop Windflower) present in the area of a kettle hole (0.8 km2) in north-eastern Poland (52°50’00’’N; 23°11’20’’E). The location of 56 clumps of generative specimens was surveyed. They numbered from several to approx. 1500 specimens. The number of generative specimens in this population exceeded 10 000. On account of this, it is a unique stand of A. sylvestris in Poland. The relationship between the occurrence of the Snowdrop Windflower and the presence of kame hills and ridges has been proven. Low slopes of kame with an inclination of 10-20o, and a north-eastern or eastern exposition has been found as the local ecological optimum for this species. Research carried out in 1970-2001 on 5 permanent plots (25 m2 each) has shown that: 1) Anemone sylvestris is an essential element of xerothermic grasslands of the order Festucetalia valesiacae; 2) the species suddenly withdraws from the parts of land on which woody species have already reached the shrub layer; 3) the greatest danger for the population of A. sylvestris in this area is the process of secondary succession initiated by the discontinuation of grazing on the kame hills.

We have analysed allele distribution at the highly polymorphic variable number of tandem repeats (VNTR) locus D1S80 (pMCT118) in the Polish population using the, polymerase chain reaction (PCR) technique. Characteristics of the D1S80 locus makes it a very useful marker for population genetic research, genetic linkage studies and forensic identification of individuals. During our routine application of the D1S80 marker to paternity testing in several cases of homozygosity detected by polyacrylamide gel electrophoresis, heteroduplex formation for alleles 18 and 24 was also observed. Direct sequencing of PCR products revealed that alleles 18 and 24 of locus D1S80 actually represent a mixture composed of different sequences. Our observations indicate that identification of some 18 and 24 VNTR alleles based only on size estimated in electrophoretic analyses could lead to errors in paternity testing and DNA profiling.

Baker’s yeast beyond the traditional use in the food industry may be used to carry out biotransformations. The effectiveness of yeast as biocatalysts is based on the presence of large amounts of intracellular enzymes, whose efficiency can be repeatedly increased by permeabilization. It is the process of increasing the permeability of cell walls and membranes in order to facilitate reagents access to the intracellular enzyme. Alcohols permeabilization process allows for approx. 50-fold increase in catalase activity of baker’s yeast. In this paper, the influence of physical and chemical parameters on the effectiveness of permeabilization of baker’s yeast cells using alcohols was analyzed. Research has shown that with increasing temperature of permeabilization process better results are achieved using a lower alcohol concentration. Based on presented response surface graphs, we can also indicate a negligible impact of duration time on the process efficiency.

Planning and spatial management are very often associated with the best providing human needs that require adjustment to environmental conditions. Thus interpreted, the rational spatial management brings many benefits to the local community, often the price of it is the degradation of the environment. System solutions reduce the negative effects of the investment on the environment and society. As a part of that, an investment in the implementation of apply optimal solutions in terms of environmental and social protection are procedures for environmental impact assessments (EIA). The aim of this study was to determine the operations to minimize negative environmental impacts and methods of environmental compensation in EIA procedures in the planning and implementation of the linear investments in Poland on A2 highway from Stryków I to Konotopa section ‚E' case study. To determine the most effective mitigation methods the negative impact of the highway and activities in terms of environmental protection, simplified EIA was carried out. The most threatened elements of the natural and social environment were identified: groundwater, soil, human health, acoustic climate.

Screening for molecular changes within the adenomatous polyposis coli (APC) gene, exons 11-14 and the 5’ half of exon 15, encompassing the mutation cluster region within exon 15, was performed in 30 patients with Familial Polyposis Coli (FAP). All patients were studied by heteroduplex analysis (HA) and single strand conformation polymorphism (SSCP) and molecular changes were found in 7 cases. Protein truncation test (PTT) has been performed in 17 cases in which mutations have not been found earlier, and shortening of protein product was noted in 2 cases. In three cases common deletion of 5 bp at codon 1309 and in one 5 bp deletion at codon 1061 were found. In other cases the molecular changes were demonstrated as heteroduplexes in exon 14 (1 patient), in segments E and F (one patient each) of exon 15, and in two cases the heteroduplexes were within the overlapping sequences of segments E/F and F/G of exon 15, respectively. In families where the molecular changes were found by HA, 7 persons at high risk for FAP were found and advised to undergo regular endoscopic examinations. In three persons at risk the transfer of mutation was excluded.

The research was carried out in north-eastern Poland in the vicinity of the village of Haćki (52°50’00’’N; 23°11’20’’E). This area is distinguished by an exceptionally numerous agglomeration of kames (10). Previous archaeological and soil research showed that kames have been inhabited by man from the iron age, and that the following stages of colonization are reflected in three levels of fossil soils. In modern times local people use the base of kames for the storage of potatoes over the winter. The potato pits are a cultural curiosity found only in this area. They are dug out in the cohesive, fine-grained substratum, which is the building material of kames, down to 1.5-2 m deep. Research plots (16) with a surface area of about 2-5 m2 were established in the potato pits in 1990. In the summer of the same year 16 phytosociological surveys were performed and soil samples taken. The investigated habitats are characterized by very low humidity. The concentration of carbon in the samples from potato pits dug out at the foot of a kame hill (the site of a settlement in the past) results probably from the occurrence of soil from the Roman period. At the present time the pits are covered by poor ruderal flora (approx. 10 species in one phytosociological survey). It is distinguished from other communities of such type by a low participation of species occurring in the xerothermic grasslands.
