Venice, one of the most enchanting cities in the world, has always been a very popular destination for travellers. For Poles who travelled through Europe in the eighteenth century, Venice was an important and frequently visited city. Polish noblewomen of that era, who travelled as frequently as their male counterparts, wrote many memoirs describing their journeys. Very few of their diaries, however, have survived to this day. Memoirs describing their Venetian escapades, rarely published, are mostly buried deep in archives and libraries. Travel diaries of Teofila Konstancja Morawska, née Radziwiłł (1738–1818) and Katarzyna Platerowa, née Sosnowska (born c. 1748–1832), widely considered as two of the most interesting publications describing Venice of that time, are filled with thoughtful observations of life in eighteenth-century Venice. Details about the city’s landscape and architectural artefacts, the chronicles of theatrical performances, descriptions of works of art and local cuisine and customs represent a remarkable source of information about appearances, the mentality and everything that defined the life of the eighteenth-century Venetians. Their memoirs are on a par with those written by men of their time and their observant eyes make them an extraordinary source of information about life in Venice of the eighteenth century.

Stanisław Mikołaj, son of Agnieszka, nee Izbicki, and Stanisław Treter, the king’s chamberlain, was born on 19th November 1776. When he was seven years old it turned out that his mother was mentally ill, most probably suffering from schizophrenia. In the 18th century mental and nervous disorders were very rare, and foreigners visiting Poland even thought they were characteristic of Poles. Certainly, mental disease in a family does not only affect the one who actually suffers from it, it has an impact on each family member and somehow everyone is involved. Agnieszka’s psychosis started with aggressive behaviour towards her family, and especially her son. She would destroy things which either belonged to him or were in some way related to him, she would scream at him, physically and mentally harass him. His father would usually buy the things the boy needed most, such as underwear, clothes and shoes, in well-kept secret. Agnieszka categorically opposed this and even “became stubborn and restless which influenced the atmosphere in this home”. Because of his mother’s disease and its influence on the atmosphere in the family, the boy often felt anxiety and fear, and sometimes even annoyance and despair. In November 1786 Stanisław Treter decided that it would be better if his wife stayed in Warsaw for a while. The atmosphere at home was becoming worse and worse, and their ten-year-old son required systematic and extensive education, while Agnieszka herself needed “professional” care. In the 18th century people who suffered from mental diseases would usually live with their families and be provided with good care and a kind of particular respect, unless they were dangerous to others. However, Agnieszka was completely unpredictable. Her irrepressible aggression was understood by her son as a lack of love and acceptance. He often felt lost and very lonely. His mother’s mental disease did not create supportive conditions for the development of the child’s emotions and mentality. Certainly, like any other child he loved his “Mummy” very much, so parting with her was a very difficult experience for the boy. Disharmony in the Treter family, which was the consequence of Agnieszka’s disease, developed in Stanisław Mikołaj patterns of instability, hostility and neglect. Thus it is not surprising that he became oversensitive, egoistic, combative and aggressive. We learn about Stanisław Mikołaj Treter’s difficult childhood from his father’s notes. In 1785 he decided to write a history of his son’s life and education, and it took him four years to write in 13 letters which are now kept in the Central State Historical Archive of Ukraine in Lvov.

The author of the article presents the contents of an interesting treaty written in November 1785 in a mansion house in Mikołajów—situated in the Sandomierz area – by Stanisław Treter of the Lubomirskis (1744–1833), the chamberlain of the King Stanisław August Poniatowski. The purpose of the treaty was the presentation of life and education of his son Stanisław Mikołaj (1776–1861). The work devoted and dedicated to his, then nine-year-old, son, was written in a form of letters, very popular in the period of the Enlightenment. The letters are an interesting contribution to the history of upbringing, education, fatherhood, motherhood, childhood, travel and even medicine. They are a specific “voice of the epoch” reflecting the lifestyle, values, concerns and joys of the people of the Polish Enlightenment.

In the rich prose of Tomasz Kajetan Węgierski (1755-1787), a talented poet and satirist, a special place is occupied by a French-language diary. This diary was in the form of letters written during a journey around Italy in 1779. His recipient was a certain Julia, a friend and lover of the poet living in Paris. His letters survived in the Manuscript Section of the Jagiellonian Library in Krakow, though, unfortunately, as an incomplete copy only: just seven letters, written in November and December from Verona, Vicenza, Padua, and Venice, have been preserved entirely. We also have the beginning of an eighth letter, written from Venice. Furthermore, we have the poet’s comments on several missing letters, four of which refer to the city on the lagoon and one to Ferrara. There is also a fragment of a letter from Rome devoted to the catacombs. From its content, we learn that the poet intended to return to the city of St. Mark to see the wedding ceremony of the Venetian Republic with the sea. The aim of this article is to present the motivations that prompted the poet to leave the PolishLithuanian Commonwealth, as well as to analyse his impressions from his trip around Veneto. It is worth noting that this region was only a stage, though a very important one, on the poet’s route leading to the capital of the Papal States. This Italian region fascinated, surprised, and shocked the Pole, and in his notes, which are full of anecdotes and jokes, we find extensive descriptions of the history and culture of Verona, Vicenza, Padua, and Venice. Also included are more-detailed characteristics of the inhabitants and their customs, as well as all kinds of information about everyday life on the road. Węgierski’s narrative, though different in many respects from the travel descriptions of his predecessors, is quite typical of the century in which he lived.
Nella ricca produzione in prosa di Tomasz Kajetan Węgierski (1755-1787), poeta e satirico di talento, occupa un posto particolare il diario redatto in francese in forma epistolare durante il suo viaggio in Italia nel 1779. La sua destinataria era una certa Julia, amica parigina e amante del poeta. Le lettere italiane di Węgierski si sono conservate nelle collezioni della Sezione Manoscritti della Biblioteca Jagellonica di Cracovia, purtroppo come copia incompleta. Disponiamo di sette lettere intere, scritte nel novembre e nel dicembre del 1779 da Verona, Vicenza, Padova e Venezia, nonché della parte iniziale di un’ottava, anch’essa datata a Venezia. Inoltre, ci sono pervenuti i commenti di Węgierski ad alcune lettere mancanti, di cui quattro riguardano la città sulla laguna e uno Ferrara. Si è conservata anche una parte di una lettera scritta da Roma e dedicata alle catacombe. Dal suo contenuto veniamo a sapere che il poeta intendeva tornare nella città di San Marco per assistere alla cerimonia dello Sposalizio di Venezia con il mare, che cadeva in concomitanza con la festa dell’Ascensione. Scopo del presente articolo è presentare le ragioni che indussero il poeta nel 1779 a lasciare la Confederazione polacco-lituana e a recarsi in Italia, nonché svolgere un’analisi delle sue lettere, scritte durante il soggiorno nel Veneto. Vale la pena di sottolineare che questa regione costituiva una semplice tappa, per quanto significativa, lungo l’itinerario che conduceva Węgierski nella capitale dello Stato Pontificio. La Venezia Euganea affascinava il polacco, lo sorpendeva e scioccava. Nelle sue note di viaggio, piene di aneddoti e facezie, troviamo ampie descrizioni della storia e della cultura di Verona, Vicenza, Padova e Venezia, estesi ritratti dei suoi abitanti e delle loro usanze, nonché le più svariate informazioni sulla vita quotidiana, raccolte lungo la via. La narrazione di Węgierski, pur differendo per diversi aspetti dalle descrizioni di viaggio dei suoi predecessori, è piuttosto tipica dell’epoca in cui viveva.

In Stanislaw August Poniatowski’s time, the upbringing and education of wealthy noblewomen were commonly entrusted to foreign governesses. There was a strong conviction in the Polish-Lithuanian Commonwealth that command of foreign languages was the foundation of proper and careful upbringing of young women. Many parents were not discerning clients when it came to selecting a governess. Oftentimes, foreign origin sufficed. As a result of attaching so much importance to command of foreign languages accompanied by acquiring foreign manners, costumes and customs, young Polish women grew indifferent to their mother tongue, native culture and traditions. There was also a lack of sense of responsibility for the country’s future.

Neuroblastoma is the most common extra-cranial solid tumor of childhood and it is characterized by the presence of a glycosphingolipid, GD2 ganglioside. Monoclonal antibodies targeting the antigen are currently tested in clinical trials. Additionally, several research groups reported results revealing that ganglioside-specific antibodies can affect cellular signaling and cause direct cytotoxicity against tumor cells. To shed more light on gene expression signatures of tumor cells, we used microarrays to analyze changes of transcriptome in IMR-32 human neuroblastoma cell cultures treated with doxorubicin (DOX) or a mouse monoclonal antibody binding to GD2 ganglioside 14G2a (mAb) for 24 h. The obtained results highlight that disparate cellular pathways are regulated by doxorubicin and 14G2a. Next, we used RT-PCR to verify mRNA levels of selected DOX-responsive genes such as RPS27L, PPM1D, SESN1, CDKN1A, TNFSF10B, and 14G2a-responsive genes such as SVIL, JUN, RASSF6, TLX2, ID1. Then, we applied western blot and analyzed levels of RPS27L, PPM1D, sestrin 1 proteins after DOX-treatment. Additionally, we aimed to measure effects of doxorubicin and topotecan (TPT) and 14G2a on expression of a novel human NDUFAF2 gene encoding for mimitin protein (MYC-induced mitochondrial protein) and correlate it with expression of the MYCN gene. We showed that expression of both genes was concomitantly decreased in the 14G2a-treated IMR-32 cells after 24 h and 48 h. Our results extend knowledge on gene expression profiles after application of DOX and 14G2a in our model and reveal promising candidates for further research aimed at finding novel anti-neuroblastoma targets.

Opis:
The civilization diseases of the 21st century are non-infectious disorders, affecting a large part of modern society. They are associated with the significant development of industry and technology, and hence with environmental pollution and an unhealthy lifestyle. These factors have led to the development of many civilization diseases, which currently include: cardiovascular diseases, respiratory diseases, diabetes, obesity, malignant tumors, gastrointestinal diseases, mental disorders and allergic diseases. The development of technologies, including modern therapies and new drugs, resulted in increase in life expectancy. This creates a global problem of an aging population with an increasing number of diseases of the old age, i.e. dementias. In addition, sedentary lifestyles and changing diets are the reasons why more and more people develop metabolic diseases, as well as neurological and cognitive disorders characterized by progressive damage to nerve cells and dementia. Currently, problem on a global scale is also the growing resistance to existing antimicrobial drugs. Therefore, the scientists face many challenges related to searching for the causes of these diseases, their diagnosis and treatment. Scientific research conducted at the Department of Biomedical Chemistry at the Faculty of Chemistry of the University of Gdańsk is part of this research trend. In this publication, we discuss various research topics with the long-term aim of solving the problems associated with the diseases mentioned above. The following chapters are dedicated to (i) looking for new effective fluorophores with diagnostic and anti-cancer activity; (ii) designing of new compounds with antibacterial and antiviral activity and their synthesis; (iii) investigating the mechanisms of amyloid deposit formation by human cystatin C and possibilities of inhibition of this process; (iv) designing and studies of compounds activating the proteasome with the potential to suppress the development of neurodegenerative diseases; (v) designing peptide fibrils and hydrogels as drug carriers; (vi) searching for peptide inhibitors of immune checkpoint as potential drugs for immunotherapy; (vii) studying the mechanism of action of selected herpesviruses by determining the structure of viral proteins and (viii) studying the composition of natural glycans and glycoconjugates in order to better understand the mechanisms of interaction of bacteria with the environment or with the host.
