Tobacco use is a leading cause of preventable mortality worldwide. New cost-effective smoking cessation treatments are needed especially in some low-to-middle income countries where smoking rates are rising, and current pharmacotherapy treatments remain cost-prohibitive. Since the 1960’s, cytisine has been used as an effective nicotine substitution agent to aid in smoking cessation albeit limited to a selected few Eastern/Central Europe and Central Asian countries. Cytisine is a biologically active alkaloid of plant origin and is known to be a ligand of nicotinic acetylcholinergic receptors (nAChRs). For several decades, the properties of cytisine have been investigated and reported in the biomedical and pharmaceutical literature. Due to the beneficial impact of cytisine on smoking cessation and its costly multistep synthesis, there is a growing interest in extraction from natural sources as well as in analytical identification and quantification for clinical medicine and forensic toxicology. In this paper, we present several current analytical approaches to cytisine extraction and identification from biological samples of plant and human origin. The development of extraction techniques will allow for the widespread use of the drug in experimental and clinical pharmacology, toxicology and forensic medicine.

Opis:
The structure of Austempered Ductile Iron (ADI) is depend of many factors at individual stages of casting production. There is a rich literature documenting research on the relationship between heat treatment and the resulting microstructure of cast alloy. A significant amount of research is conducted towards the use of IT tools for indications production parameters for thin-walled castings, allowing for the selection of selected process parameters in order to obtain the expected properties. At the same time, the selection of these parameters should make it possible to obtain as few defects as possible. The input parameters of the solver is chemical composition Determined by the previous system module. Target wall thickness and HB of the product determined by the user. The method used to implement the solver is the method of Particle Swarm Optimization (PSO). The developed IT tool was used to determine the parameters of heat treatment, which will ensure obtaining the expected value for hardness. In the first stage, the ADI cast iron heat treatment parameters proposed by the expert were used, in the next part of the experiment, the settings proposed by the system were used. Used of the proposed IT tool, it was possible to reduce the number of deficiencies by 3%. The use of the solver in the case of castings with a wall thickness of 25 mm and 41 mm allowed to indication of process parameters allowing to obtain minimum mechanical properties in accordance with the PN-EN 1564:2012 standard. The results obtained by the solver for the selected parameters were verified. The indicated parameters were used to conduct experimental research. The tests obtained as a result of the physical experiment are convergent with the data from the solver.

Nowadays nanostructures are more and more often designed as carriers for drug delivery, especially to improve the drug pharmacokinetics and pharmaco-dynamics. Numerous kinds of nanostructures are considered a good prospect for medical applications thanks to their small size, acceptable biocompatibility and toxicity. Due to the fact that nanotechnology is a new field of science, every nano-scale product must be thoroughly examined regarding its toxicity to the human body. This study provides new insights into effects of exposing endothelial cells to the selected nanostructures. Dendrimers of the fourth generation (PAMAMs), multi-walled carbon nanotubes (MWCNTs) and silver nanoparticles (SNPs) were used to evaluate nanostructures influence on endothelial cells in vitro. The nanostructures were evaluated via transmission electron microscopy and dynamic light scattering technique. The cells previously exposed to the nanostructures were observed and analyzed via the atomic force microscopy and scanning electron microscopy to obtain a quantitative evaluation of the cells morphology. The presence of multi-walled carbon nanotubes and silver nanoparticles on the cells surface was confirmed by the scanning electron microscopy. Our results confirm that the surface association and/or uptake of nanostructures by the cells resulting from physicochemical and biological processes, affect the cells morphology. Morphological changes can be induced by the membrane proteins interaction with nanomaterials, which trigger a sequence of intracel-lular biological processes.
