For the reliable applications of silver nanowires, AgNW, which is used as a conductive transparent film in electronic devices, the isothermal degradation behaviors of AgNW films with and without overcoating were investigated. Accelerated isothermal degradation was performed as a function of temperature, time, and atmosphere. Electrical resistance and optical transmittance were measured and correlated with the microstructural damages, such as formation of oxide particles and fragmentations of AgNW, which were quantitatively determined from the scanning electron micrographs. The overcoating retarded the formation of oxide particles and subsequent fragmentations as well as resulting degradation in electrical resistance without affecting the optical transmittance.

Opis:
The effect of Ganoderma lucidum extract on glucose uptake was studied in L6 rat skeletal muscle cells. G. lucidum extract increased glucose uptake about 2-fold compared to control. The extract stimulated the activity of phosphatidylinositol (PI) 3-kinase which is a major regulatory molecule in the glucose uptake pathway. About 7-fold increased activity of a PI 3-kinase was observed after treatment with G. lucidum extract, whereas PI 3-kinase inhibitor, LY294002, blocked the G. lucidum extract-stimulated PI 3-kinase activity in L6 skeletal muscle cells. Protein kinase B, a downstream mediator of PI 3-kinase, was also activated by G. lucidum extract. We then assessed the activity of AMP-activated protein kinase (AMPK), another regulatory molecule in the glucose uptake pathway. G. lucidum extract increased the phosphorylation level of both AMPK α1 and α2. Activity of p38 MAPK, a downstream mediator of AMPK, was also increased by G. lucidum extract. Taken together, these results suggest that G. lucidum extract may stimulate glucose uptake, through both PI 3-kinase and AMPK in L6 skeletal muscle cells thereby contributing to glucose homeostasis.

Pleurostomatids are raptorial ciliates that form a very distinct group within the Haptoria. Traditionally, the order Pleurostomatida was divided into two families: the Amphileptidae with two perioral kineties and a suture formed by the right side ciliary rows, and the Litonotidae with three perioral kineties and without suture. However, molecular phylogenies depicted the “traditional” Amphileptidae as a paraphyletic assemblage nesting also the Litonotidae. To overcome this problem we have analyzed genealogy of pleurostomatids using morphological data and 18S rRNA gene sequences, including newly sequenced genera Acineria and Kentrophyllum. Specifically, we have combined a morphological and molecular approach and have used also some other phylogenetic tools such as phylogenetic networks, split spectrum analysis, quartet mapping as well as the likelihood method of tracing history of morphological characters. These analyses show that: (1) there are not two but three distinct pleurostomatid lineages – Epiphyllidae fam. nov., Amphileptidae and Litonotidae; (2) epiphyllids (Epiphyllum + Kentrophyllum) represent a basal pleurostomatid group which is defined by two perioral kineties, by the presence of a suture on both the right and the left side of the body, by the loss of the oral bulge extrusomes, and by the extrusome fringe extending all around the body except for the oral region; (3) the families Amphileptidae and Litonotidae are monophyletic each, and represent sister groups; (4) Acineria belongs to the Litonotidae, as already indicated by morphological data; (5) Loxophyllum is a monophyletic and crown genus of the Litonotidae; and (6) Litonotus is paraphyletic, which could be very likely caused by a rapid radiation event that did not allow primary nucleotide homologies to be fixed.

Complementary DNAs (cDNAs) of protein phosphatase 2C (PP2C) were cloned from two marine scuticociliates Uronema marinum and Miamiensis avidus. Both PP2C proteins showed structural characteristics of typical PP2C, such as highly conserved amino acid residues predicted for binding to phosphate and metal ions, 11 conserved PP2C motifs and 10 invariant residues. The phosphatase activity of recombinantly produced U. marinum PP2C (UmPP2C) was in proportion to the PP2C protein and Mg2+ concentrations, and was not sensitive to okadaic acid, but was inhibited by sodium fluoride, EDTA or Ca2+. The expression of UmPP2C was significantly up-regulated by exposure the ciliates with PMA suggesting that UmPP2C dephosphrylates proteins phosphorylated by protein kinases as in other eukaryotes and has a regulatory function against abrupt increase of protein phosphorylation triggered by strong stimulations.

Gamijeonssibaekchulsan (GJBS) is a typical Oriental medicine prescription which has been used in Korea for the treatment of allergic diseases and the development of physical strength. However, as yet there is no clear explanation of how GJBS affects the anaphylactic reaction and the immune function. In the present study murine models and MOLT-4 cells, a T cell line, were used to investigate these effects. Compound 48/80-induced systemic anaphylactic shock and ear swelling response were firstly analyzed. We also assayed histamine release and passive cutaneous anaphylaxis (PCA) in mice and cytokine productions in MOLT-4 cells. GJBS significantly inhibits compound 48/80-induced systemic anaphylactic shock and ear swelling response. GJBS also inhibits histamine release from rat peritoneal mast cells induced by compound 48/80. PCA activated by anti-dinitrophenyl immunoglobulin E is attenuated by GJBS. However, GJBS dose not affect the production of interferon-γ, interleukin (IL)-2, and IL-4 in MOLT-4 cells. These results indicate that GJBS has a potential regulatory effect on allergic reactions that are mediated by mast cells.
