This study was aimed to evaluate the effects of inulin used as prebiotic on the kidney in lipopolysaccharide (LPS)-induced endotoxemia model. Wistar Albino rats were divided into four groups: Control group, LPS (endotoxemia) group, Inulin + LPS group in which LPS (1.5 mg/kg, E. coli, Serotype 0111: B4) was treated after inulin (500 mg/kg) given by gavage for 21 days and Inulin group. The animals were sacrificed 24 h after the last LPS injection. Kidney samples were taken for biochemical and immunohistochemical analyses. Total antioxidant status (TAS), total oxidant status (TOS), oxidative stress index (OSI), malondialdehyde (MDA) and myeloperoxidase (MPO) values were determined. In addition, kidney sections were stained for inducible nitric oxide synthase (iNOS), tumor necrosis factor (TNF)-α and interleukine-6 (IL-6) expression, and leukocyte infiltration. LPS caused oxidative stress and inflammation. Inulin administration could prevent oxidative stress and lipid peroxidation. Moreover, inulin decreased iNOS, TNF-α and IL-6 expression. However, it did not change the distribution of leukocytes in kidney tissues. These results suggest to promising benefits of inulin as prebiotic in reducing the effects of endotoxemia. Further studies should be conducted to evaluate the capacity of prebiotics in endotoxemia.

Opis:
Phosphoric acid is utilized for production of fertilizers. It is mainly produced by wet processes, in which phosphate concentrate, produce by several operations including crushing, scrubbing, classification, attritioning, and desliming, is leached with sulfuric acid with coproduction of calcium sulfate dihydrate (phosphogypsum). The New Valley oxidized phosphate concentrate was processed for phosphoric acid production by addition of certain additives to improve gypsum filtration through modifying its morphology and particle size distribution. The additives used were cetylpyridinium chloride (CPC) as a cationic surfactant, sodium dodecyl sulfate (SDS) as an anionic surfactant and sulfonic acid (SA). The applied dosages ranged from 0.25 to 2.5 kg additive/Mg P2O5 in the phosphate concentrate. The filtration rate of the New Valley oxidized phosphate concentrate is 3.6 Mg P2O5 /m2day without additives. The filtration rate was increased by 33% and 31% with 1 and 0.8 kg/Mg P2O5 doses of CPC surfactant and sulfonic acid (SA), respectively. On the other hand, the filtration rate was decreased by 33% with addition of 1 kg/Mg P2O5 dose of SDS. The aspect ratios of gypsum crystals were 6:1, 3:1, 3:1 and 12:1 without additive, with CPC, SA and SDS surfactant, respectively. Reaction efficiencies without and with 0.8 kg/Mg P2O5 of CPC, SA and SDS dose of additives were 95.0%, 97.9%, 97.9% and 94.7%, respectively. The P2O5 recoveries without and with 0.8 kg/Mg P2O5 of CPC, SA and SDS dose of additives were 92.6%, 95.5%, 95.8% and 90.8%, respectively.

The cowpea aphid (Aphis craccivora Koch) is considered a serious insect pest attacking several crops. We carried out biochemical studies to elucidate the role of the metabolising enzymes in conferring resistance to thiamethoxam, in two strains (resistant and susceptible) of the cowpea aphid. Bioassay experiments showed that the thiamethoxam selected strain developed a 48 fold resistance after consecutive selection with thiamethoxam for 12 generations. This resistant strain also exhibited cross-resistance to the tested carbamates; pirimicarb and carbosulfan, organophosphorus (malathion, fenitrothion, and chlorpyrifos-methyl), and the neonicotinoid (acetamiprid). Synergism studies have indicated that S,S,S-tributyl phosphorotrithioate (DEF), a known inhibitor for esterases, increased thiamethoxam toxicity 5.58 times in the resistant strain compared with the susceptible strain. Moreover, the biochemical determination revealed that carboxylestersae activity was 30 times greater in the resistant strain than in the susceptible strain. In addition, the enzyme activity of glutathione S-transferase (GST) and mixed function oxidases (mfo) increased only in the resistant strain 3.7 and 2.7 times, respectively, in relation to the susceptible (the control). Generally, our results suggest that the higher activity of the detoxifying enzymes, particularly carboxylesterase, in the resistant strain of the cowpea aphid, apparently have a significant role in endowing resistance to thiamethoxam, although additional mechanisms may contribute.

The cowpea aphid, Aphis craccivora management relies mainly on chemical control. As a result extensive and repeated treatment of insecticides has led to the development of aphid resistance to commonly used insecticides. To investigate chlorpyrifos-methyl resistance in A. craccivora, a field strain was selected for 24-generations to achieve a resistance factor of 82.3 fold compared with a susceptible strain. In the resistant strain, malathion and lambda-cyhalothrin exhibited obvious cross-resistance; while fenvalerate and dinotefuran showed moderate cross-resistance. In contrast, slight or no cross-resistance was obtained with the other tested insecticides. To investigate metabolic resistance mechanisms, integration of biochemical and synergism assays was conducted. Results showed the key role of esterase (EST) and mixed function oxidases (MFO); however, glutathione-s-transferase (GST) contributed less to resistance. Cross-resistance studies showed the need for rotation with non-cross resistant insecticides as a resistance management tactic.

Varthemia iphionoides is a perennial plant that belongs to Asteraceae family. This study investigates the cytotoxic effect of V. iphionoides essential oil on breast (MCF7), prostate (PC3), and chronic myelogenous leukemia (K562) and normal human fibroblast cell lines using MTT assay and flow cytometric analysis. In addition, GC-MS of the oil was carried out. The IC50 values for PC3, MCF7, K562 and fibroblast were 145.3, 188.8, 87.88 and 173.3 µg/ml, respectively. V. iphionoides essential oil was most effective against K562. Flow cytometric results for IC50 dose of V. iphionoides oil on K562 cells showed 32.2 % apoptosis in 24 h. GC-MS analysis resulted in the identification of 25 compounds. 1,8-Cineole, borneol, and α-cadinol were the major constituents of V. iphionoides volatile oil. In conclusion, this study reveals for the first time the cytotoxic activity of V. iphionoides essential oil on K562 cell line which may occur through apoptosis induction.
