Opis:
We report the synthesis of isotopomers of L-phenylalanine and L-tyrosine selectively labelled with hydrogen isotopes in the 2-position of the side chain. The deuterium or tritium label was introduced using reductive amination activity of enzyme L-phenylalanine dehydrogenase (EC 1.4.1.20). This way p-phenylpyruvic acid was converted into [2-2H]-, [2-3H]-, and doubly labelled [2-2H/3H]-isotopomers of L-phenylalanine, using deuteriated, tritiated, and mixed (DTO) incubation media, respectively. Similarly, p-hydroxyphenylpyruvic acid was converted into [2-2H]-, [2-3H]-, and [2-2H/3H]-L-tyrosine. Deuterium labelled isotopomers of L-phenylalanine and --tyrosine can be used as markers in the investigation of abnormal metabolism of these amino acids observed in patients with inborn genetic diseases such as phenylketonuria and tyrosinemia.

Opis:
The enzymatic deamination of 5-fl uorotryptamine and 5-hydroxytryptamine, 5-HT, catalysed by enzyme monoamine oxidase A (MAO-A, EC 1.4.3.4) was investigated using the kinetic (KIE) and solvent (SIE) isotope effects methods. The numerical values of deuterium isotope effects in the (1R) positions of 5-F-tryptamine were determined using non-competitive spectrophotomeric method. Isotopologue 5-F-[(1R)- -2H]-tryptamine, needed for kinetic studies was obtained by enzymatic decarboxylation of 5'-fl uoro-L-tryptophan, 5'-F-L-Trp, in fully deuteriated medium.

Opis:
Deuterium kinetic isotope effects (KIE’s) in the reaction of oxidation of dopamine (DA) catalyzed by the enzyme horseradish peroxidase (HRP, EC 1.11.1.7) was determined using a non-competitive spectrophotometric method. For kinetic studies, the deuterium ring labelled isotopomer [2’,5’,6’-2H3]-dopamine was synthesized using acid catalyzed isotopic exchange between authentic dopamine and heavy water. Deuterium solvent isotope effects (SIE’s) for dopamine and norepinephrine were determined separately for the enzymatic oxidation carried out in the presence or absence of hydrogen peroxide. Some mechanistic details of enzymatic oxidation of dopamine and norepinephrine to corresponding catecholchromes catalyzed by HRP were discussed.

Opis:
The kinetic isotope effect (KIE) method was applied to study the mechanism of elimination of bromine from p-nitro-erythro-alfa,beta-dibromocinnamic acid labeled at the alfa-carbon. This compound was obtained starting from [2-14C]malonic acid via [2-14C]cinnamic acid and subsequent addition of bromide. The value of 14C KIE determined for alfa-position of side chain of p-nitro-erythro-alfa,beta-dibromocinamic acid proves that elimination of bromine leading to formation of (E)-p-nitrocinnamic acid proceeds via E2 mechanism.

The synthesis of specifically ring labeled isotopomer of L-tyrosine, (l-Tyr), using chemical and enzymatic methods is reported. The carbon-14 labeled [1’-14C]-L-Tyr has been synthesized by a 6-step conversion of [2-14C]-malonic acid into [4’-14C]-phenol and its subsequent condensation with S-methyl-L-cysteine using enzyme tyrosine phenol lyase from Citrobacter freundii.

The tritium kinetic isotope effect on position 2 has been determined in the reaction of decomposition of L-tryptophan, L-Trp, catalyzed by enzyme TPase, (EC 4.1.99.1). We report the studies on the mechanism of oxidation of dopamine (DA) to 3’,4’-dihydroxyphenylacetaldehyde (DOPAL) by enzyme tyramine oxidase (EC 1.4.3.6) using kinetic isotope effects (KIE) and solvent isotope effects (SIE) methods. For kinetic studies, the selectively deuterium labelled isotopologues of dopamine, i.e., [(1R)-2H]-, and [(1S)- -2H]-DA were used. The numerical values of KIE’s for the (1R)- and (1S)-isotopologues of dopamine in the enzymatic oxidation of DA to DOPAL were determined using the non competitive spectrophotometric method. Also, the SIE’s for this reaction carried out in heavy water were obtained spectrophotometrically. Some mechanistic details of enzymatic oxidation of DA to DOPAL were discussed.

Opis:
12C/14C kinetic isotope effects on the hydrolytic cleavage of tyrosine to phenol and ammonium pyruvate catalyzed by Citrobacter freundii tyrosine phenol-lyase have been determined in positions 2, 3 and ring-1' of L-tyrosine. The competitive method with dual-label approach was applied with 3',5'-ring 3H as remote label. The results revealed the change of the effect on carbon atom in position 2 during the reaction course from the high normal values to low inverse values. On the other hand, the effect values on 3 and ring-1' position remained constant during the reaction course. The discussion of these results regarding the reaction mechanism is presented.

Opis:
Sea ice conditions in the Baltic Sea during six latest winters – 2010/2011 to 2015/2016 are analysed using coupled ice– ocean numerical model 3D CEMBS (3D Coupled Ecosystem Model of the Baltic Sea). Simulation results are compared with observations from monitoring stations, ice charts and satellite data. High correlation between model results and observations has been confirmed both in terms of spatial and temporal approach. The analysed period has a high interannual variability of ice extent, the number of ice days and ice thickness. Increasing number of relatively mild winters in the Northern Europe directly associated with climate change results in reduced ice concentration in the Baltic Sea. In this perspective, the implementation and development of the sea ice modelling approach (in addition to standard monitoring techniques) is critical to assess current state of the Baltic Sea environment and predict possible climate related changes in the ecosystem and their influence for human marine–related activities, such as fishery or transportation.
