Opis:
To improve the measuring accuracy in two-step phase-shifting interferometry (PSI), a new approach combining the extreme value of interference (EVI) and the least-squares iterative algorithm (LSIA) is proposed to extract the phase from two-frame blind phase-shifting interferograms. This method first evaluates the phase shift between two interferograms by the EVI algorithm, and then constructs the fitted interferogram by the addition of two interferograms after filtering the corresponding background intensities, so the phase with high precision can be retrieved by combining two real interferograms and this fitted interferogram using the LSIA method. The proposed algorithm expands the flexibility of the LSIA method and has the high-precision performance compared with the existing algorithms in two-step PSI. Simulation and experiment are performed to verify the feasibility of the proposed algorithm.

Opis:
The interferogram containing the noises often affects the accuracy of phase retrieval, leading to the degradation of the phase imaging quality. To address this issue, a new interferogram blind denoising (IBD) method based on deep residual learning is proposed. In the presence of unknown noise levels, during the training, the deep residual convolutional neural networks (DRCNN) in the IBD approach is able to remove the latent clean interferogram implicitly, and then gradually establish the residual mapping relation in the pixel-level between the interferogram and the noises. With a well-trained DRCNN model, this algorithm can deal not only with the single-frame interferogram efficiently but also with the multi-frame phase-shifted interferograms collaboratively, while effectively retaining interferogram features related to phase retrieval. Simulation and experimental results demonstrate the feasibility and applicability of the proposed IBD method.

Bim is a pro-apoptotic member of the Bcl-2 protein family. Overexpression of Bim proved to be highly cytotoxic for diverse cells.The AD293 cell line is derived directly from the HEK293 cell line but has been transfected with a gene that can improve cell adherence.We found that there was almost no apoptosis seen in Bim L-transfected AD293 cells, but more than half ofBim L-transfected HEK293 cells underwent apoptosis. Suppression subtractive hybridizationwas used to detect the different gene expression profile between these two cell lines. In 192 sequencedpositive clones, there were 30 clones repeating twice or more. Ten genes were selected for identification by semi-quantitative RT-PCR.Thetranscripts of two adhesion-relatedgenes (actin and parvin)and two apoptosis-related genes (cyclin 2 and protein phosphatase 1G) were up-regulated in AD293 cells. These results suggest that the high expression of cell adhesion-related proteins might be responsible for the different apoptosis status after the transfection of Bim L.Our data provide candidate genes responsible for the different apoptosis sensitivity of these two cell lines. Further investigation on thedifferential expression profile between AD293 and HEK293 might improve our understanding of cell apoptosis mechanism.

Opis:
Human cytomegalovirus (HCMV) is often a dangerous opportunistic pathogen that causes significant morbidity and mortality in newborn children and immunocompromised patients. The different symptoms and tissue tropisms of HCMV infection may result from genetic polymorphism. This study investigated the sequence variability of the HCMV US28 ORF, which shows sequence homology to the G protein-coupled receptor. HCMV isolated from suspected pediatric cases and isolates from AIDS patients were compared in order to examine the possible associations between polymorphisms and pathogenesis. Seventy children with suspected congenital HCMV infection, who suffered from jaundice (47), megacolon (10), and microcephaly (13), and 17 AIDS patients, were studied. Mutation was prevalent among the sequences of US28, with a focus on the two ends of US28. The important functional groups of US28 are highly conserved. An unrooted tree showed that all sequences from suspected congenitally infected infants and AIDS patients were divided into three groups. Comparison showed that most of the sequences (12/17) from pediatric patients were included in the first group (G1), whereas most of the sequences (11/17) from AIDS patients were included in the third group (G3). The specific high mutation sites in US28 from children were located at the C terminus of the protein, whereas those from AIDS patients were located at the N terminus. We demonstrated the existence of polymorphisms among the US28 genes of clinical isolates of HCMV from infants with suspected congenital infection. Comparison of US28 sequences from AIDS patients with those from children showed that both sequences have their own specific high mutation points.

Inflammation plays an important role in the development of many obesity-related diseases. This study aimed to investigate the effect of ezetimibe on inflammation and myocardial remodeling in obese rats. A rat model of obesity was established, and myocardial damage was examined by transmission electron microscopy and Masson staining. Twenty obese rats were divided into two groups (n=10): obese group and ezetimibe group. Ten SD rats were used as controls. Western blot was performed to monitor the expression of P-p38MAPK and interleukin (IL)-6. Immunohistochemical staining was used to monitor the expression of intercellular adhesion molecule-1 and vascular cell adhesion molecule-1. In the obese rats group, we observed increased inflammatory factors and myocardial hypertrophy. In contrast, the ezetimibe group exhibited decreased expression of inflammatory factors and an improvement in myocardial remodeling compared to the obese group. Mechanistically, we found that ezetimibe decreased P-p38MAPK, IL-6, intercellular adhesion molecule-1, and vascular cell adhesion molecule-1 levels in the hearts of the obese rats. Taken together, these results indicate that ezetimibe may improve myocardial remodeling in obese rats by inhibiting inflammation.
