Lysophospholipids have long been recognized as membrane phospholipid metabolites, but only recently lysophosphatidic acids (LPA) have been demonstrated to act on specific G protein-coupled receptors. The widespread expression of LPA receptors and coupling to several classes of G proteins allow LPA-dependent regulation of numerous processes, such as vascular development, neurogenesis, wound healing, immunity, and cancerogenesis. Lysophosphatidic acids have been found to induce many of the hallmarks of cancer including cellular processes such as proliferation, survival, migration, invasion, and neovascularization. Furthermore, autotaxin (ATX), the main enzyme converting lysophosphatidylcholine into LPA was identified as a tumor cell autocrine motility factor. On the other hand, cyclic phosphatidic acids (naturally occurring analogs of LPA generated by ATX) have anti-proliferative activity and inhibit tumor cell invasion and metastasis. Research achievements of the past decade suggest implementation of preclinical and clinical evaluation of LPA and its analogs, LPA receptors, as well as autotaxin as potential therapeutic targets.

Opis:
Angiogenesis, the formation of new capillaries from pre-existing vascular network, plays an important role in physiological and pathological processes such as embryonic development, wound healing, and development of atherosclerosis. Extension of the circulatory network is also considered to be one the most important factors during cancerogenesis. Inhibition of angiogenesis may lead to inhibition of tumor growth whereas stimulation may improve wound healing. Research achievements suggest the use of plants and their extracts as potential therapeutic agents with pro- or antiangiogenic activity. Since the anticancer and antiangiogenic properties of many phytomedicines have been amply reviewed elsewhere this paper will focus on the treatment of vascular insufficiency in wound healing. Globally accepted herbal drugs are thought to be safe and effective, however, there is a need for more evidence-based confirmation in controlled and validated trials. Among the most frequently studied proangiogenic phytochemicals are ginsenosides from Panax ginseng, beta-sitosterol from Aloe vera, calycosin from Radix Astragali, and extracts from Hippophae rhamnoides L. and Angelica sinensis.

Opis:
Multidrug resistance-associated protein (MRP1) is a transmembrane pump protein responsible for the efflux of chemotherapeutic drugs, an important cause of anticancer treatment failure. Trying to circumvent MRP-mediated resistance we designed and synthesized hairpin loops forming antisense oligodeoxyribonucleotides (ODNs), both phosphodiesters (PO-ODNs) and their phosphorothioate analogues (PS-ODNs), to reduce the protein expression by targeting its mRNA in a sequence specific manner. Melting temperature measurements as well as polyacrylamide gel electrophoresis supported the preferential formation of a secondary structure, which was expected to protect ODNs against 3'-exonuclease degradation. ODNs and PS-ODNs designed in this work were successfully tested as antisense inhibitors of the expression of MRP1 in the leukaemia HL60/ADR cell line. Foreseeing the necessity to perform clinical studies with such ODNs we investigated their stability against the 3'-exonuclease activity of fetal calf serum and human plasma. Under the conditions, corresponding to physiological ones, we observed high stability of hairpin loop forming ODNs, especially those containing longer (e.g. 7 base pair) stems. Comparative studies on the stability of chemically unmodified hairpin loop forming ODNs and their PS-counterparts indicated that endonuclease activity did not play any important role in the process of their nucleolytic degradation. Our studies provide strong evidence for high stability of chemically unmodified hairpin loop ODNs, making them an attractive alternative to phosphorothioate analogues commonly used in antisense strategy.

Multidrug resistance-associated protein (MRP1) causes cellular drug resistance in several cancer cell lines. In this paper we show that antisense oligonucleotides decrease MRP1 expression in human leukaemia cells. We investigated biological activity of a series of 12 linear phosphorothioate oligonucleotides, complementary to several regions of MRP1 mRNA. The oligonucleotides were administered to leukaemia HL60/ADR cells overexpressing MRP1 protein. Then, the level of MRP1 mRNA was determined by means of semiquantitative RT-PCR and the protein level by reaction with specific monoclonal antibodies. Some of the investigated antisense oligonucleotides decrease the expression level of the MRP1 protein by 46% and its mRNA level by 76%.

Opis:
The use of nucleotides and their analogs in the pharmacological studies of nucleotide receptors (P2 class) should be preceded by detailed studies on their degradation connected with ecto-enzymes of a given cell type. In the present studies we have analyzed stability of some phosphorothioate and phosphonate analogs of ATP and ADP in the HeLa epitheloid carcinoma and endothelial HUVEC cells cultures. Our studies have revealed that ecto-nucleotide pyrophosphatase (E-NPP) is one of the main enzymes involved in the extracellular degradation of ATP and other nucleotides in the HeLa cells. On the other hand, the ecto-ATPDase is responsible for the hydrolysis of extracellular nucleotides in human endothelial cell cultures, while the E-NPP-like enzymes of the HUVEC cells are not essential to this degradation. The concerted action of the aforementioned ecto-enzymes and nucleotide pyrophosphatase, 5'-nucleotidase and adenosine deaminase present in fetal bovine serum (FBS) supplied to the culture medium, results in partial or complete degradation of the phosphorothioate (ATPγS) and phosphonate analogs of adenosine nucleotides (α,β-methylene-ATP and β,γ-methylene-ATP) in the cell cultures. Only ADPβS appears to be resistant to these enzymes. The influence of some nucleotides and their analogs on the proliferation of the HeLa cells in presence or absence of FBS is also discussed.

The raw trimming waste from the leather industry is considered potential hazardous waste as a consequence of the chrome tanned leather process. On the other hand, leather waste contains a large amount of precious protein – collagen, which has many uses. Nowadays, collagen preparations obtained from leather waste are available on the market. This paper presents a procedure for the determination of amino acids in five collagen preparations of animal origin. Recent improvements in HPLC-based methods for analysing amino acids have made it feasible to analyse different sample types accurately. In this study the 6-aminoquinolyl-N-hydroxysuccinimidyl carbamate (AQC) derivatization procedure was applied. The amino acid analysis indicated the presence of 18 amino acids (Asp, Ser, Glu, His, Gly, Arg, Thr, Ala, Pro, Cys, Tyr, Val, Lys, Met, Ile, Leu, Phe and Hyp) in the collagen samples. Glycine, alanine, proline and hydroxyproline were the most abundant amino acid, whereas the lowest contents corresponded to serine, tyrosine, valine and izoleucine. The analysis proposed can be used with confidence in collagen quality control to guarantee appropriate amino acid content.
Niegarbowane odpady skórzane pochodzące z przemysłu garbarskiego są potencjalnym zagrożeniem dla środowiska naturalnego. Z drugiej jednak strony tego rodzaju odpady zawierają znaczące ilości cennego białka – kolagenu. Białko kolagenowe jest biopolimerem, który z uwagi na swoje właściwości znajduje zastosowanie w przemyśle spożywczym, kosmetycznym oraz w przemyśle biomedycznym. Obecnie na rynku dostępne są preparaty kolagenowe pozyskane z odpadów pochodzenia zwierzęcego. W pracy przedstawiono procedurę oznaczania aminokwasów w wybranych preparatach kolagenowych metodą wysokosprawnej chromatografii cieczowej i metodą spektrofotometryczną. We wszystkich próbkach oznaczono wysokie stężenia glicyny, alaniny, proliny i hydroksyproliny, a niewielkie ilości tyrozyny, seryny, waliny i izoleucyny. Zastosowana metoda chromatograficzna umożliwia szybkie i równoczesne oznaczenie 17 aminokwasów w badanych próbach. Opracowane w ramach pracy metody analityczne mogą być wykorzystane m.in. do szybkiej kontroli składu aminokwasowego kolagenu.

The subject of this paper is improvement in the growth and yield of three different types of legumes and rape in drought conditions by coating seeds with hydrolysed collagen from tanning waste. In addition, the impact of various additives in the seed shell on the growth of the plant was investigated. The encapsulation process of seeds was conducted on a disc granulator. A centrally placed seed was first coated with a layer of fungicides. The next layer was collagen hydrolysate, collagen hydrolysate with latex or a solution of yellow dextrin and polyvinyl alcohol. The outer layer was a mineral additive e.g. dolomite or kaolin. After the end of the encapsulation process on the disk granulator, all of the seeds tested were sown into soils. Seeds without coating were also sown as control seeds. Seedlings were maintained for 29 days with cultivation without irrigation. The length of the seedlings was analysed for all of the seeds sown. Higher seedling growth values were obtained for seeds coated with collagen hydrolysate in comparison with control seeds (without coating). The use of collagen hydrolysate gave slightly better results than in the case of a solution of dextrin with polyvinyl alcohol.
Specyficzne właściwości pozyskanego z odpadów skórzanych hydrolizatu kolagenowego umożliwiły jego zastosowanie w procesie otoczkowania nasion roślin strączkowych oraz rzepaku w celu zmniejszenia ryzyka związanego z niewykiełkowaniem nasion podczas suszy, zwiększenia plonów oraz wskazanie nowego kierunku zagospodarowania odpadowych produktów branżowych. Proces otoczkowania poszczególnych nasion przeprowadzono w granulatorze talerzowym. Centralnie umieszczone nasiona najpierw pokryto warstwą fungicydów. Kolejną warstwą był hydrolizat kolagenu, hydrolizat kolagenu z lateksem lub roztwór żółtej dekstryny i alkoholu poliwinylowego. Zewnętrzną warstwę stanowił dodatek mineralny. Analizę chromatograficzną składu aminokwasów przeprowadzono dla preparatu kolagenowego oraz dla losowo wybranych ziaren po otoczkowaniu. Po procesach powlekania nasiona wysiano do gleby uniwersalnej. Analizowano długość siewek dla zasianych nasion w okresie 29 dni bez dostępu wody. Wyższe wartości wzrostu siewek uzyskano dla nasion pokrytych hydrolizatem kolagenu w porównaniu z nasionami kontrolnymi tj. bez powłoki oraz w przypadku otoczek z roztworem dekstryny i z polialkoholem winylowym.
