Opis:
Objectives Diesel exhaust fumes emission is a significant source of ultrafine particles, the size of which is expressed in nanometers. People occupationally exposed to diesel exhaust particles include mainly workers servicing vehicles with engines of this type. This article presents the analysis of measurements of ultrafine particle concentrations occurring in the bus depot premises during the work connected with everyday technical servicing of buses. Material and Methods The measurements were carried out in the everyday servicing (ES) room of the bus depot before, during and after the work connected with bus servicing. Determinations included: particle concentrations in terms of particle number and particle surface area, and mass concentrations of aerosol. Results Mean value of number concentration of 10- to 1000-nm particles increased almost 20-fold, from 7600 particles/cm³ before starting bus servicing procedures to 130 000 particles/cm³ during the bus servicing procedures in the room. During the procedures, the mean surface area concentration of particles potentially deposited in the alveolar (A) region was almost 3 times higher than that of the particles depositing in the tracheo-bronchial (TB) region: 356.46 μm²/cm³ vs. 95.97 μm²/cm³, respectively. The mass concentration of the fraction of particulate matter with aerodynamic diameter 0.02–1 μm (PM₁) increased 5-fold during the analyzed procedures and was 0.042 mg/m³ before, and 0.298 mg/m³ while the procedures continued. Conclusions At the time when bus servicing procedures continued in the ES room, a very high increase in all parameters of the analyzed particles was observed. The diesel exhaust particles exhibit a very high degree of fragmentation and, while their number is very high and their surface area is very large, their mass concentration is relatively low. The above findings confirm that ultrafine particles found in diesel exhaust fumes may be harmful to the health of the exposed people, and to their respiratory tract in particular.

Objectives: The aim of this study was to investigate the cyto- and genotoxicity of nanocomposites (NCs) and generation of reactive oxygen species (ROS) as a result of particle-cell interactions. Materials and Methods: Titanium dioxide (TiO₂-Ag) and ion-exchange resin (Res-Ag), both coated with silver (Ag), were examined. The murine macrophage J774A.1 cells were incubated in vitro with NC at different concentrations for 24 h. Cytotoxicity was analyzed by the methylthiazolyldiphenyltetrazolium bromide reduction test (MTT reduction test). ROS generation was assessed by incubation of cells with dichlorodihydrofl uorescein diacetate (DCF) and fl ow cytometry. DNA damage was detected by comet assay and included single-strand breaks (SSB), alkali-labile sites (ALS) and oxidative DNA damage after formamidopyrimidine glycosylase (FPG) treatment. The tail moment was used as an indicator of DNA damage. Results: TiO₂-Ag was not cytotoxic up to 200 μg/ml, whereas IC₅₀ for Res-Ag was found to be 23 μg/ml. Intracellular ROS levels were elevated after 4 h of exposure to Res-Ag at the concentration of 50 μg/ml. Both types of NC induced fragmentation of DNA strands, but only one of the composites caused damage to purine bases. TiO₂-Ag induced SSB of DNA at concentrations of 10 and 5 μg/ml. For Res-Ag, a concentration-dependent increase in tail moments was observed. Conclusions: Silver-coated nanocomposites (both TiO₂- Ag and Res-Ag) may cause genotoxic effects in murine macrophages J774A.1. Res-Ag increased generation of ROS which suggested that toxicity of Res-Ag in murine macrophages is likely to be mediated through oxidative stress. This paper will support industry and regulators alike in the assessment of hazards and risks and methods for their mitigation at the earliest possible stage in material and product development.

Wprowadzenie: Alergeny roztoczy mogą łatwo gromadzić się w materacach i na powierzchniach tekstylnych, głównie na pościeli. Zatwierdzone strategie ograniczania narażenia na alergeny roztoczy obejmują stosowanie pokrowców z certyfikowanej tkaniny barierowej. Pokrowce mogą znacząco zwiększyć komfort i bezpieczeństwo nocnego snu oraz zapobiegać występowaniu zaostrzeń alergii atopowych i astmy. Celem pracy była ocena skuteczności blokowania alergenów przez nowo opracowane tkaniny barierowe przeznaczone na pokrowce antyalergiczne. Metody: Tkaniny barierowe z włókien poliestrowo-bawełnianych z jednostronną powłoką polimerową zostały opracowane i wytworzone przez Łukasiewicz-Instytut Włókiennictwa. Skuteczność barierową opracowanych tkanin i kontrolnej tkaniny bawełnianej, przed alergenem kurzu i roztoczy kurzu domowego Der p 1, badano zgodnie z wcześniej opisaną i opatentowaną metodą.[1,2]. W skrócie: próbki kurzu domowego odsysano odkurzaczem przez badane tkaniny, a kurz i alergeny zbierano na filtrach. Po ekstrakcji filtrów oznaczono stężenia alergenu roztoczy Der p 1 komercyjną metodą ELISA. Skuteczność barierową określono jako % przepuszczalności tkanin powlekanych w stosunku do kontrolnej tkaniny bawełnianej. Wyniki: Zbadano łącznie sześć rodzajów tkanin powlekanych i wybrano jedną o najlepszych właściwościach, zapobiegających przenikaniu kurzu i Der p 1. Ten opracowany materiał (nazwany Argo Plus) charakteryzował się wysoką skutecznością barierową wobec alergenu Der p 1 (0,15 % przepuszczalności ) oraz przepuszczalnością pary wodnej i powietrza. Po 20 cyklach prania tkanina Argo Plus zachowała swoją funkcję barierową. Wniosek: Stosując naszą oryginalną metodę badania tkanin barierowych, mogliśmy wybrać materiał włókienniczy o optymalnych właściwościach barierowych, który można wykorzystać do produkcji pokrowców na pościel i materace - barierowych wobec alergernów roztoczy kurzu domowego.
Background: Mite allergens can easily accumulate in mattresses and on textile surfaces, mainly on linen. Approved strategies for reducing exposure to dust mite allergens include the use of covers from certified barrier fabric. Covers may significantly increase the comfort and safety of nighttime sleep and prevent the occurrence of exacerbations of atopic allergies and asthma. The aim of this study was to evaluate the allergen-blocking effectiveness of newly developed barrier fabrics designed for antiallergic covers. Methods: The barrier fabrics made of polyester/cotton woven structure with one-sided polymer coating were developed and manufactured by the Łukasiewicz-Textile Research Institute. Barrier effectiveness of designed fabrics and control cotton woven fabric against dust and house dust mite allergen Der p 1 was tested according to the previously described and patent method.[1,2]. In brief, house dust samples were sucked onto the tested textile material with a vacuum cleaner and the allergen was collected on filters. Following a filter extraction, the mite allergen Der p 1 concentration was determined with commercial ELISA method. Barrier effectiveness was defined as the % of permeability of tested fabrics in relation to control cotton fabric. Results: A total of six fabrics types were tested and the one with the best performance in preventing dust and Der p 1 penetration was chosen. This developed textile material (named Argo Plus) was characterized by a high barrier effectiveness against Der p 1 allergen (0,15 % permeability), but at the same time had sufficient permeability to the air and water vapor. When subjected to 20 washing cycles the Argo Plus fabric retained its barrier function. Conclusion: Using our original method for barrier fabrics testing, we were able to select a textile material with optimal barrier characteristics, which may be used to manufacture mattress covers.

The effects of linoleic acid supplementation on protozoa counts, fermentation parameters and fatty-acid composition of Entodinium caudatum and Diploplastron affine were examined in in vitro cultures. Entodinium caudatum (EC) and Diploplastron affine (DA) were isolated from the rumen of sheep (Slovak Merino breed) and cultivated anaerobically in the presence of bacteria in caudatum-type medium. To test the effect of soluble linoleic acid (LA) on protozoan growth, both ciliate species were supplemented with LA (1.51 μg/L) on the day of dilution over a 30-day period. Twenty-four-hour fermentation parameters were examined on both ciliate cultures and their respective bacterial fractions. Ciliate counts of both EC and DA cultures were not significantly affected by supplemented LA. The major impact of the soluble form of LA supplement was found in the bacterial fractions of both ciliate cultures. LA supplementation had a greater effect on fermentation parameters and fatty-acid contents in the EC experimental groups than in the DA groups. Our results suggest that experimental rumen ciliates and their associated bacterial populations had different metabolic responses to the tested form and concentration of supplemented LA.

This review presents a brief account of the chemistry and mechanistic aspects of aryl H-phosphonates, and selected applications of this class of compounds as intermediates in the synthesis of a wide range of biologically important analogues of nucleoside phosphates, and oligonucleotides, in which the phosphate moieties are replaced by other structurally related groups. The aryl nucleoside H-phosphonates, compounds of controlled reactivity, have proven to be more versatile and superior to various mixed anhydrides as synthetic intermediates, particularly for preparation of nucleotide analogues bearing P-N or P-S bonds in various configurational arrangements at the phosphate moiety.

Liver transplantation is a well-established treatment of patients with end-stage liver disease and selected liver tumors. Remarkable progress has been made over the last years concerning nearly all of its aspects. The aim of this study was to evaluate the evolution of long-term outcomes after liver transplantations performed in the Department of General, Transplant and Liver Surgery (Medical University of Warsaw). Material and methods. Data of 1500 liver transplantations performed between 1989 and 2014 were retrospectively analyzed. Transplantations were divided into 3 groups: group 1 including first 500 operations, group 2 including subsequent 500, and group 3 comprising the most recent 500. Five year overall and graft survival were set as outcome measures. Results. Increased number of transplantations performed at the site was associated with increased age of the recipients (p<0.001) and donors (p<0.001), increased rate of male recipients (p<0.001), and increased rate of piggyback operations (p<0.001), and decreased MELD (p<0.001), as well as decreased blood (p=0.006) and plasma (p<0.001) transfusions. Overall survival was 71.6% at 5 years in group 1, 74.5% at 5 years in group 2, and 85% at 2.9 years in group 3 (p=0.008). Improvement of overall survival was particularly observed for primary transplantations (p=0.004). Increased graft survival rates did not reach the level of significance (p=0.136). Conclusions. Long-term outcomes after liver transplantations performed in the Department of General, Transplant and Liver Surgery are comparable to those achieved in the largest transplant centers worldwide and are continuously improving despite increasing recipient age and wider utilization of organs procured from older donors.

The aim of the study was to analyze indications and results of the first one thousand liver transplantations at Chair and Clinic of General, Transplantation and Liver Surgery, Medical University of Warsaw.Material and methods. Data from 1000 transplantations (944 patients) performed at Chair and Clinic of General, Transplantation and Liver Surgery between 1994 and 2011 were analyzed retrospectively. These included 943 first transplantations and 55 retransplantations and 2 re-retransplantations. Frequency of particular indications for first transplantation and retransplantations was established. Perioperative mortality was defined as death within 30 days after the transplantation. Kaplan-Meier survival analysis was used to estimate 5-year patient and graft survival.Results. The most common indications for first transplantation included: liver failure caused by hepatitis C infection (27.8%) and hepatitis B infection (18%) and alcoholic liver disease (17.7%). Early (< 6 months) and late (> 6 months) retransplantations were dominated by hepatic artery thrombosis (54.3%) and recurrence of the underlying disease (45%). Perioperative mortality rate was 8.9% for first transplantations and 34.5% for retransplantations. Five-year patient and graft survival rate was 74.3% and 71%, respectively, after first transplantations and 54.7% and 52.9%, respectively, after retransplantations.Conclusions. Development of liver transplantation program provided more than 1000 transplantations and excellent long-term results. Liver failure caused by hepatitis C and B infections remains the most common cause of liver transplantation and structure of other indications is consistent with European data.
