Opis:
Currently it is acknowledged that rural environment, inadequately to possessed natural conditions, creates worse standards for the development of general physical fitness of both examined girls and boys compared to what the urban environment offers. In turn, children raised in the city, have a relatively low level of various aspects of strength and stamina. It can be also assumed that both mentioned environments shape different fitness profiles. In this article I will try to verify a hypothesis which assumes a diversified level of motoric fitness of a child in the pre-school age depending on the place of residence as well as on the educational institution. Alternatively, it can be assumed that, in comparison to the rural environment, the fitness level of children from the urban environment will be higher and the children who attend kindergarten will be fitter than those who fulfil the compulsory schooling obligation at school.

The aim of this study was to determine the influence of place of living, type of school, and family background on the physical activity and sedentary behavior of adolescents. 1,280 adolescents took part in the research: 694 boys and 586 girls aged 16–19 from Poland. Physical activity (PA) and sedentary behavior (SB) were evaluated by IPAQ-long version. A multiple regression analysis was used to evaluate the influence of socio-economic factors on physical activity and sedentary behavior. The results suggested a difference between boys and girls in all domains of physical activity and sedentary behavior. The results also indicated that social factors influence physical activity and sedentary behavior in different ways. After adjusting the model to BMI and age tested, the key determinants for boys’ physical activity were: type of leisure time in the family and place of living, while for girls – type of school and type of leisure time in the family. Sedentary behavior of both sexes were strongly dependent on type of school, and – for boys – parents’ education and type of leisure time in the family were also important. The results suggest the need for continuous monitoring of physical activity and seated behavior as well as taking into account a wide spectrum of social factors in which adolescents grow. Interventions and strategies to promote physical activity should focus on family involvement and uniting family and educational institutions operating in local environments.

Mouse thymidylate synthase R209K (a mutation corresponding to R218K in Lactobacillus casei), overexpressed in thymidylate synthase-deficient Escherichia coli strain, was poorly soluble and with only feeble enzyme activity. The mutated protein, incubated with FdUMP and N5,10-methylenetetrahydrofolate, did not form a complex stable under conditions of SDS/polyacrylamide gel electrophoresis. The reaction catalyzed by the R209K enzyme (studied in a crude extract), compared to that catalyzed by purified wild-type recombinant mouse thymidylate synthase, showed the Km value for dUMP 571-fold higher and Vmax value over 50-fold (assuming that the mutated enzyme constituted 20% of total crude extract protein) lower. Thus the ratios kcat, R209K/kcat, 'wild' and (kcat, R209K/Km, R209KdUMP)/( kcat, 'wild'/Km, 'wild'dUMP) were 0.019 and 0.000032, respectively, documenting that mouse thymidylate synthase R209, similar to the corresponding L. casei R218, is essential for both dUMP binding and enzyme reaction.

DNA base analogs, 2,4,5,6-substituted pyrimidines and 2,6-substituted purines were tested as potential inhibitors of E. coli Fpg protein (formamidopyrimidine -DNA glycosylase). Three of the seventeen compounds tested revealed inhibitory properties. 2-Thioxanthine was the most efficient, inhibiting 50% of 2,6-diamino-4-hydroxy-5N-methyl-formamidopyrimidine (Fapy-7MeG) excision activity at 17.1 μM concentration. The measured Kgi was 4.44 ± 0.15 μM. Inhibition was observed only when the Fpg protein was first challenged to its substrate followed by the addition of the base analog, suggesting uncompetitive (catalytic) inhibition. For two other compounds, 2-thio- or 2-oxo-4,5,6-substituted pyrimidines, IC50 was only 343.3 ± 58.6 and 350 ± 24.4 μM, respectively. No change of the Fpg glycosylase activity was detected in the presence of Fapy-7MeG, up to 5 μM. We also investigated the effect of DNA structure modified by tryptophan pyrolysate (Trp-P-1) on the activity of base excision repair enzymes: Escherichia coli and human DNA glycosylases of oxidized (Fpg, Nth) and alkylated bases (TagA, AlkA, and ANPG), and for bacterial AP endonuclease (Xth protein). Trp-P-1, which changes the secondary DNA structure into non-B, non-Z most efficiently inhibited excision of alkylated bases by the AlkA glycosylase (IC50 = 1 μM). The ANPG, TagA, and Fpg proteins were also inhibited although to a lesser extent (IC50 = 76.5 μM, 96 μM, and 187.5 μM, respectively). Trp-P-1 also inhibited incision of DNA at abasic sites by the β-lyase activity of the Fpg and Nth proteins, and to a lesser extent by the Xth AP endonuclease. Thus, DNA conformation is critical for excision of damaged bases and incision of abasic sites by DNA repair enzymes.
