Opis:
W ostatnich latach nową, dynamicznie rozwijającą się gałęzią nauki o odporności jest immunometabolizm. Dział ten bada jak przemiany metaboliczne zachodzące w komórkach układu odpornościowego, wpływają na ich przetrwanie, rozwój, ale także funkcje wykonawcze. W opracowaniu tym opisujemy przebieg podstawowych i pomocniczych szlaków pozyskania energii przez leukocyty, a w szczególności glikolizę, cykl Krebsa, szlak pentozofosforanowy oraz utlenienie kwasów tłuszczowych. Przedstawiamy znaczenie poszczególnych szlaków dla funkcjonowania leukocytów, rozwoju ich fenotypu (np. makrofagów M1 i M2), oraz przełączania szlaków podczas ich aktywacji. Zmiany te mogą wpływać na funkcje obronne w czasie reakcji zapalnej, infekcji lub uszkodzenia tkanek. Z drugiej strony, leukocyty mogą realizować różne programy metaboliczne, celem pozyskania energii do walki z patogenami. Zależność pomiędzy funkcjami obronnymi a metabolizmem rzuca także nowe światło na zrozumienie mechanizmów chorób metabolicznych, a przede wszystkim kompleksowej odpowiedzi immunologicznej.
In recent years, a new branch of immunology called immunometabolism has been established. The discipline focuses on intracellular metabolic changes in immune cells that impact - influence their survival, development, as well as defense mechanisms. Here we provide a brief summary of basic and ancillary metabolic pathways which leukocytes utilize to obtain energy, with a special focus on glycolysis, TCA cycle, penthosophosphate pathway and fatty acid oxidation. Significance of the given metabolic path for leukocyte functioning, phenotype changes (e.g. M1 vs. M2 macrophages) and biochemical changes during activation is discussed. The metabolic changes can in fact shape the effector functions during inflammation, infection or tissue injury. On the other hand, leukocytes can adopt different metabolic programs to gain energy required to eliminate pathogens. An interplay between immunity and metabolism sheds new light on understanding of metabolic diseases but foremost on complex immune responses.

Young people, regardless of their social environment, place of residence or work, are looking for values and key competencies that enable achieving goals in life. Therefore, an appropriate education system is important, which in the conditions of changing reality will meet these requirements effectively. The contemporary employer is interested in four groups of key competencies, such as intellectual, professional, personal and interpersonal. Geography is a field with great potential for the development of various competencies. In this context, questions about adjusting geographical education to the expectations of employers are justified. Therefore, the aim of the study is to assess the strengths and weaknesses of the current development of competencies and qualifications at the geography speciality of the Faculty of Geographical and Geological Sciences, Adam Mickiewicz University in Poznań, Poland. The reference points included a report on research carried out among 200 employers in 2012, as well as surveys among students graduating from master‘s studies on the assessment of the level of their competencies and qualifications. It was determined that the strength of the current geographical education at the faculty is to prepare mainly specialists with broad general and professional knowledge, and high self-esteem in terms of cooperation in the group and communication. The area of development for the geographical education are intellectual competencies, above all independent thinking and prioritising. The last year geography students fall out the most in terms of personal competencies. The authors suggest building students‘ awareness because, as the above results show, they are not fully aware of what expectations they may face in the labour market. It is worth modifying the study program so as to put more emphasis on soft competencies and support the development of various forms of extra activities of students. Attention was also paid to the importance of didactic competencies, which on the one hand raise the self-esteem of students, and on the other, increase the chance of finding a good job. An interesting aspect of the research is the wide spectrum of professions in which students work. This reflects their diverse skills, including spatial thinking. Combined with increasing attention paid to the issues related to the natural environment and sustainable development, graduates of geography have a strong position to enter the current labour market. The advantage of geographers is the use of modern technologies. Unfortunately, however, the results of the research presented above, both among Polish students and internationally, indicate insufficient skills in the use of modern technologies by students of geography.

The study aimed to check the effectiveness of anticancer therapy combining a vascular-disruptive drug (combretastatin phosphate, CA4P) and a liposomal formulation of a chemotherapeutic (doxorubicin). CA4P was synthesized in our laboratory according to a previously described procedure. The antivascular drug and long-circulating doxorubicin-loaded liposomes were used to treat B16-F10 murine melanoma experimental tumors. Seventy-four hours after drug administration, a decrease in the number of tumor blood vessels was apparent and necrotic areas within tumors were visible. Combination therapy consisting of alternate administrations of CA4P and liposomal doxorubicin yielded greater inhibition of tumor growth than monotherapies alone. The best therapeutic results were obtained with the antivascular drug administered intratumorally every second day at 50 mg/kg body mass. In the case of combined therapy, the best results were obtained when the vascular-disruptive agent (CA4P) and the antineoplastic agent (liposomal doxorubicin) were administered in alternation.

Scientific research and student education aimed at preparing students to practice their profession under the conditions of civilization and technological changes play a special role in geography teaching. It is important to be aware of the impact of key competences which are necessary for every person to function in the modern world and are needed for self-fulfilment, personal development, social integration, flexible adaptation to any changes and which determine the success in adult life. Proper development of such skills contributes to the correct interpretation of natural and socio-economic phenomena and processes. The aim of the article is to present and discuss research work and teaching activities pursued by the Department of Geography Didactics and Ecological Education at the Faculty of Geographical and Geological Sciences of the Adam Mickiewicz University in Poznań, aimed at the use by students of various educational concepts and the resulting key competences necessary for their future work. Students also have the opportunity to develop soft competences, such as communication, courage of expression, self-esteem or responsibility for the group, to which employers have paid special attention in recent years. Therefore, comprehensive preparation of the student requires the implementation of specific educational concepts. The most important ones include bilingual education; CLIL, inquiry based science education (IBSE), project method, fieldwork, geographical educational trails, participation, as well as the use of geoinformation technologies, GIS and ICT.

An important aspect of a senior's life is to be able to function independently in his or her own and family surroundings as long as possible, actively participate in social life without economic barriers. Numerous studies show that the quality of life and the biological condition of the elderly alongside nutrition are clearly dominant and have a positive effect on their functional physical fitness. In order to promote physical activity and physiological prevention among the elderly, a physiotherapeutic program was organized under the "Summer Sanatorium of Prevention of the spine pain syndrome", which was attended regularly by a group of senior citizens from the Karkonosze University of the Third Age in Jelenia Góra. The aim of the study was to compare the level of functional physical activity before and after treatment among the participants. The study was conducted among 20 women aged 60-75 years, before and after 6 weeks of treatment, which included: gymnastics for prevention of back pain syndromes, relaxation training, full spine massage, TENS electrotherapy and ultrasound in the lumbar and cervical spine. Research included; assessment of functional physical fitness by the Functional Senior Fitness Tests: „stretching behind" and "slope forward", ranges of motion within the selected joints of the spine, upper and lower limbs. The selected form of physiotherapeutic treatment improves the results obtained after test with the Functional Senior Fitness Test and influences the range of motion in the selected joints, which demonstrates that physical fitness is improved. Physical activity in the form of general-purpose gymnastics favors higher levels of functional fitness of elderly women, physical therapy and massage improves mood and makes older people more willing to participate in physio preventing programs. The conducted research proves that the developed program was optimal and showed a positive effect on the daily functional fitness of the examined persons.

Neoplastic cells which co-form tumors are usually subjected to various stress factors, mainly hypoxia and shortage of nutrient factors. Such cells employ different strategies that permit their survival under such conditions. Experiments in vitro are usually carried out in the presence of 21% oxygen and medium supplemented with 10% FBS. Altering these parameters can approximate the in vivo conditions found within tumor mass. The present paper reports certain properties (especially ability to metastasize) of B16-F10 cells able to grow upon exposure to altered growth conditions (medium supplemented with 0.06% FBS or presence of 1% oxygen for 24 or 72 hours). These properties were compared with those of control cells cultured in the presence of 21% oxygen and in medium supplemented with 10% FBS. Some properties of the cells exposed to medium supplemented with 0.06% FBS differ from those of cells cultured under low oxygenation conditions (ability to form metastases, to migrate, or to express various proteins). Only the partial deprivation of oxygen did increase both the number of migrating cells and the number of metastases formed. Serum deficiency enhanced only the cell ability to metastasize, but not to migrate. It appears that cultured B16-F10 cells employ different adaptation strategies under conditions of oxygen shortage and those of serum deficiency. Under oxygen deprivation, such cells most likely undergo an epithelial-mesenchymal transition, whereas serum deficiency ("starvation"), while increasing the tumorigenicity of B16-F10 cells, does not induce the epithelial-mesenchymal transition.

One of the preconditions of effective anticancer therapy is efficient transfer of the therapeutic agent (chemotherapeutic) to tumor cells. Fundamental barriers making drug delivery and action difficult include underoxygenation, elevated interstitial pressure, poor and abnormal tumor blood vascular network and acidic tumor milieu. In this study we aimed at developing an optimized scheme of administering a combination of an angiogenesis-inhibiting drug (vasostatin) and a chemotherapeutic (cyclophosphamide) in the therapeutic treatment of mice bearing experimental B16-F10 melanoma tumors. We report that the strongest tumor growth inhibition was observed in mice that received two, three or four vasostatin doses in combination with one injection of cyclophosphamide (i.e., V2 + CTX, V3 + CTX or V4 + CTX schemes). Double administration of vasostatin increases oxygenation of B16-F10 tumors. On the other hand, its five-fold administration lowers tumor oxygenation, breaks down tumor vascular network (increasing hypoxia) and leads in consequence to death of cancer cells and appearance of necrotic areas in the tumor. A decreased cyclophosphamide dose in combination with two doses of vasostatin (V2 + CTX scheme) inhibits tumor growth similarly to a larger dose of cyclophosphamide alone.
