Opis:
Plant RNA interference has been a very well studied phenomenon since its discovery. We are well versed with the types of small noncoding RNAs that are prevalent in the plant systems and their pathways of biogenesis and subsequent actions. However, apart from model plant systems such as Arabidopsis and Oryza, very little information is available regarding the other members of the RNA interference machinery; specially Argonaute proteins which acts as the major stabilizing factor for execution of the interference. This work focuses on the exploration of the sequenced crop genomes available on the web using a hybrid approach of computational protein fishing and genome mining. The results indicate that this hybrid approach was successful in the identification of argonaute proteins in the crop genomes under study.

Opis:
RNA silencing is one of the important phenomenon in plant defense mechanism, it actively protect host plants against viral infections. Existing viruses must have developed counter defense strategies to survive this arms race. Such counter defense strategy is the viral silencing suppressor (VSRs) which have been reported to directly interfere with the various steps leading to the interference of viral RNAs. Most identified VSRs are multifunctional, besides being RNA-silencing suppressors, they often perform essential roles by functioning as coat proteins, helper components for viral transmission, replicases and movement proteins, proteases or transcriptional regulators. One such identified VSR is AC4 of Sri Lankan cassava mosaic virus strain. Trivial knowledge about the structure –function relationship of this VSR leads to this work, where we focus on the structure generation by modelling to identify the mode of interactions with the various effector molecules of the silencing pathways. Structural analyses have been performed to screen interacting residues. Results indicate conserved structural features which signify propensity of functional interactions and further shows that this VSR can be a potent tool for the analysis of RNA silencing mechanisms and the relationships between different silencing pathways and VSRs.

Opis:
Pteridophytes and more specifically ferns represent a large but threatened group of plants which often serve as important environmental markers for pollution. Reports regarding stress responses in ferns are rare, apart from a few studies involving the ecological distribution and molecular marker studies. This work isolates a glutathione peroxidase enzyme from an aquatic fern widely distributed in fresh and polluted water bodies adjacent to sources of environmental polluted sources. Further computational analyses were performed to study the structure of the protein encoded by the open reading frame. Results indicate the presence of a large number of binding pockets which serve as important binding sites in the interactions with the cognate ligands.

Purpose: In India, the cost of manpower is very low; hence Manual Material Handling (MMH) is the cheapest solution. This study aimed to quantify the cardiac strain and postural stress of male building construction workers associated with MMH tasks. Materials and methods: Mean (SD) age (years) and job experience (years) of the workers were 31.0(4.65) and 8.8 (3.23), respectively (n=35). Working peak heart rate was recorded by polar heart rate monitor, posture analysis was done by the Ovako Working Posture Analysis System (OWAS) and the Rapid Entire Body Assessment (REBA) method, body part discomfort was assessed by the Nordic questionnaire, and perceived exertion was evaluated by the Borg scale. Two-tailed unpaired Student’s t test was performed between peak heart rate of workers associated with MMH tasks and upper extremity intensive tasks (n=31). Results: Results revealed that mean peak heart rate of the workers was significantly different (higher) compared with that of the upper extremity intensive workers (p<0.05). This study showed that most of the working postures were hazardous. The magnitude of risk for musculoskeletal disorders (MSD) was much higher as per REBA compared with OWAS. Most of the workers suffered from pain in the head, neck, shoulder, lower back, and arm region. As per the Borg scale, the rate of perceived exertion was ‘hard and heavy’ among most of the workers (68.57%). Conclusions: Postural stress and cardiac strain beyond the safe limit indicates the heavy nature of the job. Load limit optimization, ergonomic lifting technique, and rescheduled work-rest cycle are essential to reducing physiological and perceived work load.

Two new species of Phleobum (Apicomplexa: Eugregarinida: Hirmocystidae) are described from adult grasshoppers in Kalyani, India. Phleobum globuloepimeritum n. sp. is described from Oxya hyla hyla (Insecta: Orthoptera: Acrididae) and Phleobum elliptica n. sp. from Atractomorpha crenulata (Insecta: Orthoptera: Pyrgomorphidae). In P. globuloepimeritum, the trophozoites are solitary, orangecoloured, have a maximum length of 352 μm with a hyaline globular epimerite and the protomerite in satellite is characteristically flanged. The gametocyst is orange, orbicular, averaging 300 × 250 μm in size, dehiscing through a pore after 50 h. The oocysts are uniformly smooth, ellipsoidal, measuring 9 × 5 μm. In P. elliptica trophozoites are also solitary, orange-coloured, having a maximum length of 157 μm with triangular epimerites. The protomerite flange is typically a bowel-shaped collar. The gametocyst is yellow-orange, ellipsoidal, averaging 240 × 165 μm and dehisces through a pore after 72 h, releasing smooth-walled, ellipsoidal, uniformly shaped oocysts measuring 5 × 3 μm. The two new species share traits, which define the genus such as: gamonts in association, satellites with characteristic flange and gametocysts dehiscence through a single pore. The epimerite of P. globuloepimeritum is hyaline and globular, which is unique among members of the genus. The gametocyst of P. elliptica is notably ellipsoidal. Basing on observation on all the described and present two species, generic characters of the genus Phleobum have been redefined. The genus is also transferred from the family Didymophyidae to the family Hirmocystidae as the gametocysts in all the species of Phleobum dehisce through a pore.

Two new myxozoan species – Myxobolus analfinus sp. n. and Myxobolus debsantus sp. n. are described from Heteropneustes fossilis (Bloch) and Catla-Rohu hybrid carp [Male parent fish Catla catla (Hamilton Buchanan) × Female parent fish Labeo rohita (Hamilton- Buchanan)], respectively. Spores of Myxobolus analfinus are oval with slightly acuminate anterior end and large prominent intercapsular notch. On the other hand, in Myxobolus debsantus spores are spherical to oval with intercapsular notch and posterior sutural markings. In both the myxobolid species polar capsules are unequal. The detailed light microscopic and SEM structures and measurements of these two myxozoans are given.

This paper deals with the morphology and life history of two new species of septate greganines (Apicomplexa: Conoidasida: Gregarinidae) of the genus Unilobus Théodoridès, Desportes & Jolivet, 1984, obtained from two tenebrionid beetles of West Bengal, India.These are Unilobus gonocephali n. sp. from Gonocephalum sp. (Insecta: Coleoptera: Tenebrionidae) and Unilobus scleroni n. sp. from Scleron irregulate (Insecta: Coleoptera: Tenebrionidae). Total length of U. gonocephali varies from 200–467 μm. Gametocysts spherical, 410 μm in average diameter, dehisces through 7–9 sporoducts. U. scleroni is comparatively smaller, total length varies from 61–232 μm. Gametocyst spherical with average diameter 350 μm, dehices through 10 to 12 sporoducts. The two new species share traits which define the genus such as the absence of epimerite, expanded protomerite and late association. These two described species are not only different from the previously described species, but also oocysts (= spores) entirely different from each other in characters like measurements, number of sporoduct and in host range. This communication also confi rmed the validity of the genus Unilobus Théodoridès, Desportes & Jolivet, 1984 and its placement under the family Gregarinidae Labbé, 1899.

The mangrove fungi, morphologically and physiologically adapted in a different habitat, were isolated from mangrove habitat of Jharkhali, Sundarban, India. The physico-chemical properties like temperature, pH, soil colour, moisture content, carbon and nitrogen content of soil determines the load of microbial population. The soil sample was serially diluted and plated on potato dextrose agar plate with ampicillin to obtain fungal isolates. Total of six isolates were characterized microscopically by lacto phenol cotton blue staining. Two of them were identified as Aspergillus niger and Penicillium sp. and are subjected to biodegradation of fish scale, the major waste of fish processing industries. Aspergillus niger was found to be the best for degradation of fish scale powder by producing zone of clearance. Moreover, media without fish scale didn’t show any zone of clearance indicates the mangrove fungi are capable of degrading the fish scale component.

In situ high-temperature scanning tunnelling spectroscopy measure-ments recorded on the heavily reduced TiO$\text{}_{2}$(110) surface which contains Ti$\text{}_{2}$O$\text{}_{3}$ regions showed disappearance of the energy gap accompanied by substantial decrease in amplitude of the band edge states with increasing temperature. It indicates smooth insulator-metal transition caused by bands overlap in Ti$\text{}_{2}$O$\text{}_{3}$, which takes place at elevated temperatures. In situ high-temperature scanning tunnelling microscopy and spectroscopy were used to study the influence of temperature on the electronic properties of Ni$\text{}_{x}$Mn$\text{}_{3-x}$O$\text{}_{3-δ}$ (0.4< x<1) thin films deposited by rf magnetron sputtering at three different oxygen/argon (2.5%, 10%, 15%) containing ambient. The morphology and distribution of the local density of states of the observed films did not show any difference for the films deposited at different conditions. The distribution of the local density of states was temperature dependent. The changes in the shape of the local density of states observed at 473 K were reversible with temperature implying that no permanent change of the electronic structure occurred.

The present study deals with the estimation of antioxidant levels in blood of sewage and garage workers exposed to sewage water, washing water and vehicular air pollution compared with control group of population. The study areas were selected on the basis of drainage clearance and wards where workers are active as sewage workers. For garage workers, the garage selected was a large and busy one where continuous vehicles washing, repairing etc are carried out. These two exposed groups were compared to non-exposed group of population as control. The blood samples were collected from two groups of exposed population as well as control population and the antioxidant levels were estimated in blood. The present results clearly indicate that there was decreasing trend of the antioxidant level in blood for sewage and garage workers. In the control groups of population the antioxidant levels were found to be within normal range, but for sewage workers (0.14-0.36 mM) and garage workers (0.12-0.36 mM) the antioxidant levels were significantly lower. The range of antioxidant level in human blood is 0.5 to 2.0 mM. It was found to be significantly lower (P < 0.001 or 0.05) in addicted sewage and garage workers when compared with non-addicted groups. All the exposed groups were showed to be having significantly lower antioxidant level when compared to control groups. This study is a preliminary assessment to know the potent biomarkers for oxidative stress and estimation of antioxidant level in blood due to occupational exposure. This study shows a way for easy screening of biomarker assessment but further work is needed in relation to biochemical, enzymological and genetic damage study.

Introduction: Though the trace element strontium’s (Sr) exact role in health remains unknown, at certain levels, it is toxic and, given the body’s low threshold for Sr, has been cited to cause nervous disorders. At different stages of the life cycle, organisms vary in their ability to discriminate Sr and calcium, which can cause age-related differences in gastrointestinal absorption. Though data regarding the effects of such absorption remain limited, it can reportedly impact health, the immune system, and chromosomal aberration. Purpose: To investigate and correlate levels of Sr concentration and any health problems at certain levels among automobile workers. Materials and methods: The blood of 41 automobile workers and 26 healthy controls of the Kolkata region was collected and whole blood Sr concentration estimated by the energy dispersive X-ray fluorescence (EDXRF) technique. Results: Automobile workers showed significantly (p <0.001) higher blood SR levels than controls from the same area, despite similar dietary habits and drinking-water quality. Conclusions: Automobile workers are exposed to different pollutants that can harm their health, and a trace element imbalance of Sr and its elevated concentrations in the blood may cause different health complications.
