Opis:
Animal models based on N-methyl-d-aspartate (NMDA) receptor blockade have been widely reported. Ketamine and MK-801, the two noncompetitive antagonists of NMDA receptors, produce behaviors related to schizophrenia and exacerbated symptoms in patients with schizophrenia. The study presented here investigated the effect of subchronic dosing (once-daily, 7 day) of histamine H3 receptor (H3R) antagonists, ciproxifan (CPX) (3 mg/kg, i.p.) and clobenpropit (CBP) (15 mg/kg, i.p) including clozapine (CLZ) (3.0 mg/kg, i.p.) and chlorpromazine (CPZ) (3.0 mg/kg, i.p.), the atypical and typical antipsychotic, respectively, on MK-801(0.2 mg/kg, i.p.)-induced locomotor activity, and dopamine and histamine levels in rats. Atypical and typical antipsychotic was used to serve as clinically relevant reference agents to compare the effects of the H3R antagonists. MK-801 significantly increased horizontal activity which was reduced with CPX and CBP. The attenuation of MK-801-induced locomotor hyperactivity produced CPX and CBP were comparable to CLZ and CPZ. Dopamine and histamine levels were measured in striatum and hypothalamus, respectively, of rat brain. The MK-801 induced increase of the striatal dopamine level was reduced in rats pretreated with CPX and CBP including CLZ. CPZ also significantly lowered striatal dopamine levels, though the decrease was less robust compared to CLZ, CPX and CBP. MK-801 increased histamine content although to a lesser degree. Subchronic treatment with CPX and CBP exhibited further increased histamine levels in the hypothalamus compared to MK-801 treatment alone. Histamine H3 receptor agonist, R-α methylhistamine (10 mg/kg, i.p.) counteracted the effect of CPX and CBP. The findings of the present study support our previous work showing positive effects of CPX and CBP on MK-801-induced schizophrenia like behaviors in rodents. However, clinical studies have reported no antipsychotic effects with histamine H3 receptor antagonists.

Opis:
Essential nutritional components are quantified in the study in addition to major antioxidants, carotenoids, phenols and flavonoids. Their antioxidant activities were also analyzed using DPPH, ABTS and hydrogen peroxide radical scavenging activities and by determining their iron reducing power. Results indicated that the highest quantity of nutritional and antioxidant components was obtained in red variety (4.63 mg/g ascorbic acid, 10.32 mg/g total carotenoid content, 61.50 mg/g total flavonoid content, 310.27 mg/g total phenolic content) followed by orange, yellow and green varieties. Similar trend was observed while analyzing their antioxidant activities by different methods. In studying correlation between components and antioxidant activity, a strong correlation was obtained for ABTS radical scavenging activity with total phenolic content (R2 = 0.722) and total carotenoid contents (R2 = 0.709), while moderate correlation was observed for ABTS radical scavenging activity with total flavonoid contents (R2 = 0.517) and ascorbic acid (R2 = 0.673).

Opis:
Toxoplasma gondii is a parasite that has been extensively studied due to its medical and veterinary importance in terminating pregnancies. Consequently, a satisfactory vaccine is required to control its adverse effects on pregnant animals. The microneme protein, MIC3, is a major adhesion protein that binds to the surface of host cells and parasites, and is therefore a potential vaccine against T. gondii. The viability of MIC3 as a vaccine is investigated in this study. Sheep were injected twice, intramuscularly, with plasmids containing DNA encoding for the mature form of MIC3 protein formulated into liposomes. Control sheep were injected with an empty vector or received no injections. The injection of sheep with DNA plasmids encoding for MIC3 elicited an immune response after the first and second injections as indicated by antibody responses and the production of IFN-γ. The immune response, as measured by the IgG2 and IgG1 serum levels, was boosted after the injection of the MIC3 DNA vaccine together with high anti-MIC3 antibodies. The results demonstrate that the intramuscular injection of sheep with a plasmid containing DNA coding for MIC3 protein induces a significant and effective immune response against T. gondii.

Bio-stimulants are in excessive demand for the sustainable production of floriculture crops. The current investigation was designed to find out the effect of naturally occurring growth stimulant moringa leaf extract (MLE) on the growth, flowering, post-harvest life and corm production of two gladiolus cultivars (cvs.) ‘Rose Supreme’ and ‘White Prosperity’. The research trial was laid out in the factorial arrangement under randomized complete block design. Corms were grown in the open field under local climatic conditions of Faisalabad, Pakistan. Potential of exogenously applied (MLE; 30 times diluted) was evaluated. Treatments including spraying MLE at 3 leaves, 5 leaves, floral bud stage, one combination of these three stages relative to control with no foliar application. Application of natural bio-stimulant produced maximum height in both cvs. against T4. Both cvs. gave maximum stalk length in response to T4. The number of florets spike–1 found the maximum (13) in T1 compared to control T0 which yield (9) florets. Maximum vase life in sucrose solution (13.33 days) was exhibited by T1 for both cvs. while minimum (10 days) in T0. In cultivar comparison earlier spike emergence was observed in ‘White Prosperity’. In ‘Rose Supreme’ maximum corm weight was attained in response to T3 (43.43 g) while minimum (30.33 g) in T0.‘White Prosperity’ produced maximum weight (40.33 g) against T4 whereas minimum by control. The cultivar mean comparison showed the superiority of cormel diameter in ‘Rose Supreme’ (10.93 mm) than ‘White Prosperity’ (9.13 mm). In treatment comparison, T4 produced maximum diameter (12.04 mm) in ‘Rose Supreme’ moreover, T2 induced maximum (9.57 mm) diameter in ‘White Prosperity’.

The paper analyses the geological conditions of study area, rock mass strength parameters with suitable support structure propositions for the under construction Nahakki tunnel in Mohmand Agency. Geology of study area varies from mica schist to graphitic marble/phyllite to schist. The tunnel ground is classified and divided by the empisical classification systems like Rock mass rating (RMR), Q system (Q), and Geological strength index (GSI). Tunnel support measures are selected based on RMR and Q classification systems. Computer based finite element analysis (FEM) has given yet another dimension to design approach. FEM software Phase2 version 7.017 is used to calculate and compare deformations and stress concentrations around the tunnel, analyze interaction of support systems with excavated rock masses and verify and check the validity of empirically determined excavation and support systems.

The performance of buckling load of tubular structures under quasi-static axial loading is quite appreciable, numerous tubes of various cross-section have been extensively investigated and corrugated sections have been designed to further improve the performance. In this paper, a carefully designed set of key performance indicators (KPIs) is utilized to assess and compare the buckling load of circular and corrugated tubes. A series of diagrams related to KPIs with various parameters of tubes are presented to demonstrate the influence of sectional configuration on the performance of tubes as well as the effect of the material on the potential of the same. The work is inestimable to engineering designs and applications, and further studies on the buckling load of other configurations.

The aim of study was to evaluate the release kinetics of domperidone maleate (DM) from immediate release (IR) tablets prepared by wet granulation method. Box behnken design (BBD) was used to optimize and evaluate the main, interaction and quadratic effects of independent variables i.e. crosslinked carboxymethyl cellulose (CMC) (X1), sodium starch glycolate (SSG) (X2) and starch (X3) on responses R2 of first order (YI) and β value of weibull model (Y2). Prepared tablets were characterized by various physical tests, in-vitro drug release, fourier transform infrared spectroscopy (FTIR), X-ray diffractometry (XRD) and differential scanning calorimetry (DSC). Accelerated stabilities studies were performed on optimized formulation D9. Y1 and Y2 were ranged from 0.9959 to 0.9994 and 0.041 to 0.912 respectively. β value of weibull model indicated the parabolic shape of dissolution curve. The quadratic model fit the data well and the resulting equations were used to predict the responses in the box behnken design. FTIR spectra showed the compatibility of DM with CMC and SSG. XRD presented diffraction lines indicates crystalline nature of drug. DSC thermograms indicated endothermic peak at 220 0C for DM. Stabilities studies revealed that no significant change in hardness, friability, disintegration time and dissolution release profile of DM. It is concluded that a combination of CMC and SSG can be used to enhance the dissolution and release kinetics of IR tablets of DM.

Coronavirus disease 2019 caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has affected 235.6 million people worldwide. In the present study, RNA-dependent RNA polymerase (RdRp) (PDB Id: 6M71) of SARS-CoV-2, an essential enzyme needed for subgenomic replication and amplification of RNA, was selected. Similar to other RdRps, it is a conserved protein and a popular target for antiviral drug therapy. Based on a computational approach, potential RdRp inhibitors were identified. The absorption, distribution, metabolism, excretion, and toxicity (ADMET) of selected molecules were determined using computation tools. The potential inhibitors were docked to the RdRp and later confirmed by Molecular Dynamics (MD) using the “Flare” module of Cresset software. Drummondin E and Flinderole B had higher drug similarity scores among the compounds selected in this study. Both these compounds are noncarcinogenic, nonirritant, nontumorigenic, and nonmutagenic. Molecular docking studies showed that both compounds can bind to RdRp. The best ligand interaction patterns were validated by MD using the “Flare” module. MD was performed for the period of 100 ns with the time step of 1 fs. The simulation results suggest that Thr-680, Arg-624, Lys-676, and Val-557 are key interacting partners in the Drummondin E-RdRp complex, while Asp-618, Asp-760, Asp-623, Arg-624, and Asp-761 are the interacting partners in the Flinderole B-RdRp complex. Based on the in silico drug-likeness score; ADMET properties; and molecular simulation result, we surmise that Flinderole B and Drummondin E could impede SARS-CoV-2 genome replication and transcription by targeting the RdRp protein.

Candida auris is a major public health concern due to its high transmission and mortality rates, as well as the emergence of pan-resistant strains. This study aimed to identify an antifungal compound from Sarcochlamys pulcherrima , an ethnomedicinal plant, that can inhibit the growth of C. auris. Methanol and ethyl acetate extracts of the plant were obtained, and high-performance thin-layer chromatography (HPTLC) analysis was conducted to identify the major compounds in the extracts. The major compound detected by HPTLC was subjected to in vitro antifungal activity testing, and its antifungal mechanism was determined. The plant extracts inhibited the growth of both C. auris and Candida albicans. HPTLC analysis revealed the presence of gallic acid in the leaf extract. Furthermore, the in vitro antifungal assay showed that gallic acid inhibited the growth of different C. auris strains. In silico studies indicated that gallic acid can bind to the active sites of carbonic anhydrase (CA) proteins in both C. auris and C. albicans, affecting their catalytic activities. Compounds that target virulent proteins such as CA can aid in the reduction of drug-resistant fungi and the development of novel antifungal compounds with unique modes of action. However, additional in vivo and clinical studies are required to conclusively determine gallic acid’s antifungal properties. Gallic acid derivatives may be developed in the future to possess more potent antifungal properties and target various pathogenic fungi.
