In the present study, four new pegylated drugs were synthesized to investigate the anticancer activity. All the compounds were characterized on the basis of FTIR and 1HNMR. The new compounds were evaluated against HCT116 cell line by CellTiter-Glo® 3D cell viability assay. The sustained release pattern of newly synthesized compounds were also evaluated, slow released lasts for 240 h which was confirmed by in vivo and in vitro drug release profile. The sustained and controlled release behavior of pegylated drugs over the extended period of time showed a linear profile and this release pattern was not limited to diffusion process instead supporting chemically controlled release pattern which was dependent only on how fast the compound hydrolyzed. The newly designed pegylated drugs showed almost same killing effect against the cancer cells as the free drugs. The compounds with lower cLogP value showed more antiproliferative (lower IC50) effect due their higher cell permeability.

The big data concept has elicited studies on how to accurately and efficiently extract valuable information from such huge dataset. The major problem during big data mining is data dimensionality due to a large number of dimensions in such datasets. This major consequence of high data dimensionality is that it affects the accuracy of machine learning (ML) classifiers; it also results in time wastage due to the presence of several redundant features in the dataset. This problem can be possibly solved using a fast feature reduction method. Hence, this study presents a fast HP-PL which is a new hybrid parallel feature reduction framework that utilizes spark to facilitate feature reduction on shared/distributed-memory clusters. The evaluation of the proposed HP-PL on KDD99 dataset showed the algorithm to be significantly faster than the conventional feature reduction techniques. The proposed technique required >1 minute to select 4 dataset features from over 79 features and 3,000,000 samples on a 3-node cluster (total of 21 cores). For the comparative algorithm, more than 2 hours was required to achieve the same feat. In the proposed system, Hadoop’s distributed file system (HDFS) was used to achieve distributed storage while Apache Spark was used as the computing engine. The model development was based on a parallel model with full consideration of the high performance and throughput of distributed computing. Conclusively, the proposed HP-PL method can achieve good accuracy with less memory and time compared to the conventional methods of feature reduction. Opis:
Soil stabilization techniques are widely used for road construction to improve the properties of the subgrade materials. Using new additives and stabilizers to improve soil properties can reduce the costs of construction and reduce the possible negative effects of these materials on the environment. The purpose of this study was to evaluate the use of a liquid based nano-material called EarthZyme (EZ) and cement kiln dust (CKD) as admixtures to improve the soil properties. A mixture of two soils was used in this study which were prepared from mixing sand soil and fine-grained soil. Compaction tests were performed on the soil that was stabilized with the CKD to determine the density-water content relationships. Unconfined compression tests were also conducted on specimens without treatment, specimen treated with the CKD only, and specimens treated with the CKD and the EZ after curing period for seven days. The obtained results indicated that adding the CKD to the soil decreased the values of the unconfined compression strength (UCS) from 5 to 15 percent. However, adding the CKD reduced the maximum dry density (MDD) from 10 to 12 %. As discussed herein, soil stabilization with the EZ had insignificant effects on the results obtained from the unconfined compression test.

In Australia and others developed countries, concerns about global warming have increased, and these concerns influence the use of refrigerants as working fluids in mechanical vapour compression refrigeration systems. One of the most important aspects of refrigerant selection is to reduce its impact on the environment and the ozone layer. This paper provides a comprehensive review of various theoretical and experimental studies which have been carried out on air conditioning and refrigeration applications to investigate the effect of refrigerants on the environment. The analysis in this paper reveals that alternative refrigerants are the most suitable working fluids that can be used in refrigeration systems to meet the needs of the environment. This study also suggests that natural types of refrigerants such as water, carbon dioxide, and hydrocarbon will play a significant role in protecting the environment and providing alternative friendly refrigerants to be used in refrigeration and air conditioning systems.

Architectural structures’ nodal coordinates are significant to shape appearance; vertical overloading causes displacement of the joints resulting in shape distortion. This research aims to reshape the distorted shape of a double-layer spherical numerical model under vertical loadings; meanwhile, the stress in members is kept within the elastic range. Furthermore, an algorithm is designed using the fmincon function to implement as few possible actuators as possible to alter the length of the most active bars. Fmincon function relies on four optimization algorithms: trust-region reflective, active set, Sequential quadratic progra mming (SQP), and interior-point. The fmincon function is subjected to the adjustment technique to search for the minimum number of actuators and optimum actuation. The algorithm excludes inactive actuators in several iterations. In this research, the 21st iteration gave optimum results, using 802 actuators and a total actuation of 1493 mm.MATLAB analyzes the structure before and after adjustment and finds the optimum actuator set. In addition, the optimal actuation found in MATLAB is applied to the modeled structure in MATLAB and SAP2000 to verify MATLAB results.

Opis:
Minimal neighbour designs (NDs) are used when a response of a treatment (direct effect) is affected by the treatment(s) applied in the neighbouring units. Minimal generalised NDs are preferred when minimal NDs cannot be constructed. Through the method of cyclic shifts (Rule I), the conditions for the existence of minimal circular generalised NDs are discussed, in which v/2 unordered pairs do not appear as neighbours. Certain generators are also developed to obtain minimal circular generalised NDs in blocks of two different sizes, where k2 = 3, 4 and 5. All these designs are constructed using i sets of shifts for k1 and two for k2.

Ligninolytic enzymes are employed for the production of second-generation biofuel to minimize fuel crisis. Additionally, they play a crucial role in global carbon cycle and a variety of applications in food, agriculture, paper and textile industries. On a large scale production of ligninolytic enzymes, microorganisms can be cultured on lignocellulosic wastes. In the present study, proximate analysis including acid detergent lignin (ADL), acid detergent cellulose (ADC), acid detergent fiber (ADF) and acid insoluble ash (AIA) were performed for Platanus orientalis (chinar), Bauhinia variegata (orchid tree), Pinus roxburghii (chirpine), wheat straw and wheat husk. Platanus orientalis was selected as substrate because of higher lignin contents for the production of ligninolytic enzymes by Aspergillus flavus. Solid State Fermentation was used and Response Surface Methodology was employed for optimizing various parameters and enzymes production. Maximum production was achieved at temperature 32°C, fermentation period 120 hours, pH 4.5, inoculums size 3.5 mL, substrate mesh size 80 mm, substrate size 7 g. Maximum purification of laccase, manganese peroxidase (MnP) and lignin peroxidase (LiP) was achieved with 50%, 60% and 40% ammonium sulfate respectively and it was enhanced by gel filtration chromatography. Characterization of enzymes shows that Laccase has 35°C optimum temperature, 4.5 pH, 0.289 mM Km and 227.27 μM/ml Vmax. Manganese peroxidase has 30°C optimum temperature, 5.5 pH, 0.538 mM Km and 203.08 μM/ml Vmax. Lignin peroxidase has 30°C optimum temperature, 3 pH, 2 mM Km and 2000 uM/ml Vmax. Protein concentrations found in crude extracts and partially purified enzymes are respectively: laccase 1.78 and 0.71 mg/ml, MnP 1.59 and 0.68 mg/ml. LiP, 1.70 and 0.69 mg/ml.

Opis:
Decomposition of organic waste in dumping sites and landfills prompts the uncontrolled emission of greenhouse gases which enhances global warming and related issues. The present investigation estimated the total landfill gas, methane, carbon dioxide and non-methane organic compounds emissions from Mahmood Booti dumping site located at Lahore, Pakistan from 1996 to 2045. LandGEM 3.02 model was utilized to evaluate the gas emission with the volumetric methane 50%, production potential (170 m3/Mg), and methane generation rate (0.050/year). The findings demonstrated that organic and food wastes had the maximum quantity in the solid waste stream (63.46%). Methane measured from solid waste was 1.150E+03 (Mg/year) in 1997 in the first year after waste was accepted by landfill while the maximum methane generation rate happened from 2014 to 2018, i.e., from 4.049×104 to 4.654×104 Mg/year, respectively. The study finding highlighted that all gases emissions have an increasing/rising trend up to 2021 and then gradually their level will decrease because most of the organic fractions of solid waste would degrade with time. The findings of this study can be used to identify the impacts and contribution of MBODS in the global emission of greenhouse gases. The study concludes that proper management of landfill gas will not only decreases greenhouse gas emissions, diminishing adverse impacts on public health but can also be used as a sustainable energy source.

Opis:
Environmental risk assessment is one of the key tools in environmental engineering. This risk assessment can be qualitative or quantitative and it is based on preliminary studies i.e., baseline study for waste disposal sites. Even though the literature exists on baseline study in general, still there is a lack of guidance regarding development of a site-specific baseline study model for a waste disposal site. This study has two-fold aim, firstly, how to develop site-specific baseline study model for a selected dumping site, and secondly, how this site-specific baseline study can support the environmental engineering via mathematical risk estimation. Mahmood Booti Open Dumping Site (MBODS) is selected to demonstrate the development and application of site-specific baseline study model. This is followed by building a framework that shows how the output of the baseline study can lead to environmental engineering via mathematical risk estimation. The paper provides a mechanism of how to construct a bespoke baseline-study model that is readily useable, avoiding procurement of expensive computer software and yet smoothly connecting with the follow-on stages of the risk assessment. The work presented in this paper can be reproduced repeatedly to create site-specific baseline study models for risk assessment of other waste disposal sites in a cost-effective, consistent and cohesive manner.
