Opis:
Marine accidents are caused by complex and diverse causes. Therefore, it is necessary to analyse accidents from the perspective of the whole system rather than the existing analysis models that focus on cause-effect. The STAMP analysis is a constraint-based model that focuses on improper management or enforcement of safety-related constraints in system operation. Thus, as a case study of the application of the STAMP technique in marine accident analysis, the Milano Bridge pier contact accident that occurred on April 6, 2020 in Busan New Port of South Korea was analysed by the STAMP technique in this study. Through this study, it was proposed to break away from the domestic accident investigation, which focused only on the punishment of the ship operator, and to suggest improvements that enable organizations related to the marine accidents to take initiative to take efforts for safety.

Opis:
Seminal plasma (SP) proteins are responsible for sperm functional quality. Developing a reliable method to determine the degree of oxidative damage of these proteins is important for establishing semen fertilizing ability. The main aim of the study was to verify the applicability of protein carbonyl derivatives measurement in the SP of canine and stallion, using a method with 2,4-dinitrophenylhydrazine (DNPH). The research material consisted of ejaculates obtained from eight English Springer Spaniels, and from seven half-blood stallions during the breeding and non-breeding season. The content of carbonyl groups in the SP was measured on the basis of the reactions with DNPH. The following reagent variants were used to dissolve protein precipitates: Variant 1 (V1) – 6M Guanidine solution and Variant 2 (V2) – 0.1M NaOH solution. It has been shown that to obtain reliable results for the measurement of protein carbonylated groups in the dog and horse SP, both 6M Guanidine and 0.1M NaOH may be used. A correlation was also found between the number of carbonyl groups and the total protein content in the canine (V1: r = -0.724; V2: r = -0.847) and stallion (V1: r = -0.336; V2: r = -0.334) SP. Additionally, the study showed a higher content (p≤0.05) of protein carbonyl groups in the stallion SP in the non-breeding season compared to the breeding season. The method based on the reaction with DNPH, due to its simplicity and cost effectiveness, appears to be suitable for large-scale application in the determination of the SP proteins oxidative damage in dog and horse semen.

Background: This proof of principle study aims to investigate the applicability of finite element analysis (FEA) in Oral and Maxillofacial (OMF) surgery, by studying the effect of mandibular body height and osteosynthesis positioning on unilateral mandibular body fractures based on Champy’s technique. Material and methods: Mandibles made of polyurethane foam (Synbone®), with heights of 18, 14, and 10 mm were used to create a FEA model with a unilateral straight-line fracture, fixated with a standard commercially available 6-hole 2 mm titanium miniplate. Two different FEA programs were used for the comparison, namely: Solidworks and Comsol Multiphysics. The FEA outcomes were compared with a series of mechanical tests with polymeric models fixed in a customised device and loaded onto a mechanical test bench. Results: First, the study illustrated that the optimal plate position appeared to be the upper border. Second, lower mandibular height increases instability and requires a stronger osteosynthesis system. Conclusions: FEA’s and polymeric model testing outcomes of unilateral non-comminuted fractures were highly comparable with current opinions of mandibular fracture management. The promising outcome of this study makes it worthwhile to do more extensive analysis in order to determine whether FEA alone is sufficient for optimisation of fracture management.

The new Reynolds-averaged Navier–Stokes (RANS)-based method has been developed for taking into account, in an approximate manner, the effect of external acoustic forcing on laminar-to-turbulent transition in a separated boundary layer. Experimental studies [33] report an increase of the turbulent shear stress within the separated boundary layer under the influence of acoustic forcing. Enhancement of flow disturbances in a reversed flow region was also reported in our experiment. Experimental findings stimulated the development of a reduced-order aero-acoustic strategy. The effect of acoustic forcing was incorporated into the modelling framework of an algebraic intermittency model. The model component was tuned based on our experimental data and validated on reference experiments. The results show the feasibility of the proposed model to simulate flow over a flat plate and the NACA0018 profile.

Opis:
Purpose: This work investigated carboxymethyl hydroxypropyl guar (CMHPG) as a potential base polymer for a fracturing fluid used in deep tight gas wells, synonym to high-temperature reservoirs. Fracturing is a common method to improve oil production by creating fractures in potentially productive reservoirs using fracturing fluids. Fracturing fluids are typically composed of water, sand, proppant, and many different additives to alter the capabilities of the fluid. Design/methodology/approach: A stable high-density fracturing fluid (HDFF) was developed to cope with scorching good temperatures up to 350°F (177°C)as well as to reduce the surface treating pressure. The HDFF consists of NaBr heavy brine, CMHPG, clay stabilizers, and zirconium and borate crosslinkers. A linear gel fluid was prepared by combining NaBr brine with CMHPG before adding the additives. The resulting fluid was then tested using a high-pressure, high-temperature (HPHT) rheometer. Findings: The results showed that the performance of the HDFF was optimum in an alkaline environment between pH 10 to 12. The efficiency of zirconium and borate crosslinkers were optimum at 0.7 ppm and 1.5 ppt, respectively. Concentrations higher than these values are not only uneconomical but will cause the fluid to be overly crosslinked, consequently reducing efficiency. In actual field operation, this is disastrous when the fluid does not flow to the fracturing column but instead swirls around the drill string as the drill string rotates. On the other hand, a reduction of concentration below the optimum values can cause fluid instability at high temperature reservoirs. Research limitations/implications: The study was limited to using two different kinds of crosslinkers and various concentrations. Future studies can be conducted using other kinds of crosslinkers, as well as an investigation into the effects of varying temperatures, pressures, and pH on the HDFF using CMHPG. Practical implications: It can be concluded that HDFF using CMHPG as the base polymer can be a potential use in formulating fracturing fluids. Originality/value: Information on the rheological behaviour of HDFF using NaBr and CMHPG can provide a reference point for future scientists in developing a new formulation of fracturing fluid.

The authors report two approaches, the first based on growth of lattice matched InGaAs/GaAsSb superlattice on InP substrate with tunable bandgap in the 2 to 3 μm range. The second approach is based on bulk random alloy InGaAsSb, which is tunable from 1.7 μm to 4.5 μm and lattice matched to the GaSb lattice constant. In each case, detector structures were fabricated and characterised. The authors have assessed the performance of these materials relative to commercially available extended short wave infrared devices through comparison to IGA-Rule 17 dark current performance level. A complementary barrier structure used in the InGaAsSb design showed improved quantum efficiency. The materials compare favourably to commercial technology and present additional options to address the challenging extended short wave infrared spectral band.

We describe two skull fragments of a new morganucodontan from the Cisco Mammal Quarry (Upper Jurassic Morrison Formation), preserving portions of the palate and snout in excellent 3D detail as well as the complete upper postcanine dentition. Morganucodontans are best known by isolated elements and relatively complete skulls of several species of Morganucodon from the Lower Jurassic of Wales and China; this group is fundamental to our understanding of the early evolution of mammals. Cifellilestes ciscoensis gen. et sp. nov. possesses derived features of the snout paired with plesiomorphic construction of the molars; the distal premolars are complex and there is an unusually low count (two) of strongly imbricated molars. This character combination expands craniodental variation for the group. We sampled mudstone from the Cisco Mammal Quarry for ash-fall zircon analysis and obtained a date of 151.50 ± 0.28 Ma. This dates the locality to the earliest Tithonian and slightly younger than other major dated mammalbearing localities in the Morrison Formation. Cifellilestes represents one of the youngest members of this group and extends the record of morganucodontans in North America by more than 30 Ma. Morganucodontans are a rare component of Late Jurassic faunas but display surprising dental diversity through variations in a tooth count and cusp morphology of a deeply conserved, generalized mammalian tooth pattern, which was fully established in brasilodontid (non-mammalian) ancestors at least 80 my prior.

The process of vitrification of the cumulus-oocyte complex (COCs) often results in cold shock. When warming, heat shock occurs which can disrupt the balance of intracellular calcium (Ca2+) intensity. Drastic changes in temperature cause Reactive Oxygen Species (ROS), affecting changes on Ca2+ in COCs. The role of calcium is needed for oocyte activation in the fertilization process. The purpose of this study was to measure the expression of Ca2+ and the intensity of Ca2+ in COCs after vitrification. The study was divided into 2 groups, the control group (C) of fresh COCs, and the treatment group (T) of COCs after vitrification. After vitrification for 24 hours, then thawing, the expression of Ca2+ was examined using the Immunocytochemistry (ICC) method and the intensity of calcium (Ca2+) with a Confocal Laser Scanning Microscope (CLSM). The research data obtained were analyzed statistically by T-Test. The results showed that the expression of Ca2+ in the control group (12.00±0.00) was different from the treatment group (0.35±0.79). The intensity of Ca2+ in the control group (1059.43±489.59) was different from the treatment group (568.21±84.31). The conclusion of this study is that cryopreservation affects calcium in COCs; there were differences in the expression and the intensity of Ca2+ between fresh COCs and COCs after vitrification. Ca2+ intensity of COCs after vitrification was concentrated in the nucleus, while in fresh COCs it was concentrated in the cytoplasm.

Amyloidosis, a serious and widespread disease with a genetic background , manifests itself through the formation of dangerous fibrils in various organs. Apart from the polluted environment and an unhealthy lifestyle, genetic factors may accelerate this process leading in some cases to lethal damages to the body. Recently, a growing interest in amyloidogenic protein research has been observed. Transthyretin (TTR) is a tetrameric protein that transports thyroid hormone thyroxine and retinol binding protein in plasma and the cerebral fluid. Sometimes TTR breaks apart and forms fibrils. Several single point mutations, having de stabilizing impact on the TTR complex, are involved in the amyloidogenic TTR cascade. Problems with the TTR tetramer stability and conformational space characteristics of the protein have not been addressed computationally before. We present selected results of our molecular dynamics (MD, ∼ 2000ns) and steered MD simulations ( SMD ) of three variants of TTR : Wild Type ( WT ), V 30 M and L 55 P . SMD has been used to enforce the dissociation of TTR . Conformational spaces of WT TTR and its amyloidogenic variants have been investigated using a novel “ One Place One Conformation ” ( OPOC ) algorithm based on a graph technique called Petrinet (PN) formalism. While the PN approach alone does not permit a direct identification of protein regions with reduced stability, it gives quite a useful tool for an effective compari son of complex protein energy landscapes explored during classical and/or SMD steered molecular dynamics simulations.
