Opis:
The aim of the research was to investigate interactions between pendimethalin (PE), an active substance of plant protection products, and the humin fraction (HM), which is the most recalcitrant component of soil organic matter. The soil material used for the study was collected from the mollic horizon of two Chernozems different in physicochemical properties. HM was obtained in the solid state after discarding humic acid (HA) and fulvic acid (FA) and digesting the mineral fraction in an HF-HCl mixture. Pure PE and two commercial products containing the substance (Penfox 330 EC and Stomp Aqua 455 SC), dissolved in hexane and added to the HM, were used in the research. The batch sorption experiment was carried out in accordance with OECD Guidelines for the Testing of Chemicals No.106. Spectroscopic methods (UV-Vis-nearIR and fluorescence) were used to assess the molecular characteristics of HM exposed to pesticides. The study showed significant differences in the molecular structures of the tested HM as a result of interactions with the pesticides used. The results revealed that PE had a different effect than the commercial products, and proved that the composition of commercial products significantly modified the strength and ability of organic matter to bind pesticides. This is probably related to the presence of adjuvants that are more competitive in forming bonds with the active sorption sites of HM, and thus strengthen or weaken the process of forming pesticide-HM complexes. As a result, the diversity of the molecular structure of HM and the presence of accompanying substances in commercial pesticide products interact, leading to different sorption mechanisms and durability of the resulting products.

Opis:
An extended second order finite difference method on a variable mesh is proposed for the solution of a singularly perturbed boundary value problem. A discrete equation is achieved on the non uniform mesh by extending the first and second order derivatives to the higher order finite differences. This equation is solved efficiently using a tridiagonal solver. The proposed method is analysed for convergence, and second order convergence is derived. Model examples are solved by the proposed scheme and compared with available methods in the literature to uphold the method.

Opis:
Great economic losses to the dairy industry are associated with bovine mastitis, which results in poor milk quality and high treatment costs. Anti-inflammatory proteins play an important role in the suppression of the immune response against invading pathogenic microorganisms and are therefore being studied for possible use in the early diagnosis of mastitis. In our study, we used milk samples from 15 cows of Holstein Friesian breed with different health status (5 healthy, 5 subclinical, and 5 clinical animals), and tested them using immunohistochemical (IHC) analysis to evaluate the presence of IL-2, IL-10, TGF-β1, βDEF-2, DEF-3, and Cathelicidin LL37 proteins. The calculation of positively and negatively stained cells for each biomarker was performed using the semiquantitative counting method. We found the presence of all factors with the exception of Cathelicidin LL37, which was almost absent in milk samples of all animal groups. The significant decrease of IL-10, β-def2, and β-def3 expression levels within the 3 days of sampling, found in the milk of animals with sub- and clinical mastitis, indicates the loss of antiinflammatory protection of the affected cow’s udder. In contrast, the stable increase of IL-2 and TGF-β1 positive cells observed in the milk of mastitis-affected cows, and the similar expression of these factors in the milk of healthy animals, indicate the possible lack of involvement of these cytokines at an early stage of udder inflammation.

Opis:
Goose astrovirus (GoAstV) is a novel avastrovirus that typically causes gosling gout and results in 2 to 20% mortality. GoAstV capsid protein is the sole structural protein, which is responsible for viral attachment, assembly, maturation as well as eliciting host antibodies. However, the epitopes within capsid protein have not been well studied. In this study, a monoclonal antibody, named 1D7, was generated against GoAstV capsid protein by hybridoma technology. Western blot results showed that this MAb could react with recombinant capsid protein expressed in E. coli. Also, it recognized the precursor of capsid protein, VP90 and VP70, in GoAstV-infected cells. Besides, excellent specificity of MAb 1D7 was further demonstrated in indirect immunofluorescence assay and immunohistochemical analysis. Epitope mapping results revealed that MAb 1D7 recognized the epitope 33QKVY 36 within Cap protein. Sequence alignment indicated that 33QKVY 36 is a conserved epitope among the isolates of goose astrovirus type 2 (GoAstV-2), suggesting the potential for its use in GoAstV-2 specific diagnostic assay. These findings may provide some insight into a function of the GoAstV capsid protein and further contribute to the development of diagnostic methods for GoAstV infection.

The aim of this study was to investigate the impact of feed supplements with alfa-amylase and beta-glucanase (Optipartum C+ 200) on ingestive-related behaviour biomarkers registered with real-time sensors: rumination behaviours and reticulorumen parameters (pH and temperature). Cows (n=20) in the treatment group (TG) were fed with Optipartum C+ 200 (Enzymes feed supplement: Alfa-Amylase 57 Units; Beta-Glucanase 107 Units) from 21 days before calving until 30 days after calving with a feeding rate of 200 g/cow/day. Cows (n=22) in the control group (CG) were fed a feed ration without feed supplement. Measurements started from 6 days before calving and continued until 21 days after calving. The following indicators were registered: with the RumiWatch System: Rumination time; Eating time; Drinking time; Rumination chews; Eating chews; Drinking gulps; Bolus; Chews per minute; Chews per bolus. With the SmaXtec system: the temperature, pH of the contents of the cows’ reticulorumens, and cows’ walking activity. According to our results, feed supplementation with alfa-amylase and beta-glucanase (Optipartum C+ 200) in the TG group resulted in increases in the following parameters: 9% rumination time and eating time, 19% drinking time, 11% rumination chews, 16% eating chews, 13% number of boluses per rumination, 5% chews per minute and 16% chews per bolus. The rumination time showed a strong, positive relation with rumination chews and bolus indicators in both groups (TG and CG) (p<0.001); while the rumination time in both groups of cows showed an opposite direction and was negatively related to eating time and eating chews (p<0.05). We found a 1.28 % lower reticulorumen pH and a 0.64 % lower reticulorumen temperature in cows fed with the supplement compared with cows in the control group. Cows in TG were 8.80% more active than those in the CG group. For improvement of ingestive-related behaviour we suggest adding a feed supplement with alfa-amylase and beta-glucanase (Optipartum C+ 200).

Purpose Mathematical modelling of the process of deformation of pipeline, transporting gas-liquid mixtures with aggressive components and a comparative analysis of the value of the specified velocity depending on the dynamic viscosity of the multicomponent gas mixture is conducted. Design/methodology/approach A mathematical model of the process of leakage of the transported product due to the loss of tightness of the pipe based on the system of Navier-Stokes equations with boundary conditions with considering the geometry of the leakage zones and the value of the leakage rate is implemented. Findings Models of the process of deformation of the pipeline due to displacements of a certain set of surface points by specifying different types of functions, describing the geometry of deformed sections are constructed. The method of calculating the tensely deformed state based on the data on the movement of surface points by comparing different ways of setting functions, taking into account the actual configuration of sections and axes is improved. The change of flow characteristics in the pipeline when changing the structure of the mix, transported by studying of influence of change of dynamic viscosity is investigated; The method of calculating the rate of leakage of the mixture in case of loss of tightness due to the occurrence of critical stresses in the pipe material is improved. Research limitations/implications Building a model of the deformation process, information about the nature, duration of forces and loads affecting the pipeline is not used. The law of the pipeline movement was constructed having taken into account the deformation of the sections in three directions. The necessity to take wind loads into account, estimating the real tensely deformed state was displayed. Practical implications Using the method of calculating the tensely deformed state based on the data on the movement of surface points by comparing different ways of setting functions, taking into account the actual configuration of sections and axes. Originality/value According to the computational algorithms created on the basis of the specified models, the calculations of the tense state of the pipelines and the flow rate of the mixture depending on its composition were performed. An analysis of the results of calculations - tense intensity and flow rate depending on the dynamic viscosity of the mixture is performed. The influence on the flow parameters - the flow rate of the mixture and the force of hydraulic resistance - changes in the dynamic viscosity of the mixture is analyzed.

Mental health is equally important with physical health. Many reports found out that poor mental health is one of the major factors of depression among seafarers. This study ascertains the factors affecting mental health and the strategies applied by Filipino seafarers to combat these issues. A survey was used as a research design. A total of 100 seafarers taken through convenience and snowball sampling answered the validated researcher-made questionnaire. The questionnaire was sent in the JBLFMU Alumni Portal, messenger, and Gmail. Frequency, percentage, and rank were used. Themes were used to classify the strategies applied by seafarers to counteract mental health issues onboard ship. The results revealed that the major contributory factor affecting the mental health of Filipino seafarers is stress, while they believed that a positive outlook in life makes one’s self free from mental health problems onboard. The top three factors are stress, fatigue, and separation from families and friends. The top three strategies to counteract factors that affect seafarers’ mental health onboard ship are staying positive, socializing with crewmates, and communicating with family.

Purpose: This study was aimed to investigate the effectiveness of plastic waste as fine aggregates to partial replacement of sand reinforced with inorganic pigment from red stone to manufacture paving block for pedestrian application. This is an effort not only to reduce plastic waste in the environment but also as an innovative way to find out an alternative eco-friendly paving block material for public walkways with an attractive appearance while ensuring pedestrian comfort. Design/methodology/approach: Approaches were converted the plastic waste to plastic powder which is then used as fine particles to sand partial replacement. The red stone powder is used to give red color to the paving block surface. The paving block materials were completely mixed in a pan mixer and added water as much as 12% of the total mass of the materials used. The paving block was cast in a mold dimension size of 20 cm × 10 cm × 6 cm and pressed with a load of 6 tons using a pressing machine. The effect of natural river sand to plastic powder ratio and curing time on the compressive strength and water absorption were investigated. Findings: The study results confirmed that the replacement of sand with plastic powder decreased the compressive strength of paving block. By partial replacement of sand with plastic powder in the range of 10% to 50% by weight, the compressive strength and water absorption value of pavement after 30 days agitation were at range of 18.06-12.78 MPa and 4.28-3.25%, respectively. This value was still met the minimum requirement for pedestrian applications according to Indonesian National Standard. Research limitations/implications: Replacing sand up to 50% by weight with plastic waste produces paving blocks with compressive strength and water absorption suitable for sidewalks and pedestrians. It is needed to continue research in terms of durability tests in order to be comfortable with the practical use of the material. Practical implications: The use of plastic waste reinforced with red stone powder as fine aggregate makes it one of the alternative ways to reduce plastic waste in the environment and obtain eco-friendly paving blocks with an attractive appearance. Originality/value: It has been experimentally proven that replacing sand up to 50% by weight with plastic powder produces paving blocks that are suitable for pedestrians application. The addition of red stone powder pigment makes the color of paving block surface become more attractive appearance.

Opis:
A proof of concept for using paper test as a suitable method in the production of monoclonal antibodies (MAbs) is reported. The paper test which detects antibodies against porcine circovirus type 2 (PCV2) using colloidal gold-labelled capsid protein as the antigen probe was applied exclusively in the screening of anti-PCV2 MAbs. It allowed the detection of 118 single cell clones within 30 min using naked eyes. MAbs with specific binding to authentic epitopes on the virus were selected using a blocking strategy in which the antibody was pre-incubated with PCV2 viral sample before applying to the test paper. Five hybridomas secreting MAbs against the capsid protein were obtained, with only three of them capable of binding to PCV2. The results were validated and confirmed using enzyme-linked immunosorbent assay and immunofluorescence assay. The paper test is simple, rapid, and independent on professional technicians and proves to be an excellent approach for the screening of MAbs against specific targets.

Classical swine fever (CSF) and porcine reproductive and respiratory syndrome (PRRS) are responsible for major economic losses and represent a threat to the swine industry worldwide. Routine surveillance serology for CSF and PRRS viruses is critical to maintaining the health status of sow farms in Hunan Province, which is one of the top pig production provinces in China. The aim of our study was to investigate the serological statistics of CSF virus (CSFV) and PRRS virus (PRRSV) in Hunan Province. The cohort serum samples were collected from vaccinated and unvaccinated pigs. Our findings showed that the average rates of CSFV and PRRSV antibody seropositivity were 82.2% (95% CI: 80.1-84.3) and 84.8% (95% CI: 82.5-87.1), respectively, in the immunized group and that these rates were higher than those in the unvaccinated group (58.6% for CSFV and 47.8% for PRRSV). Additionally, the level of CSFV antibody in piglet serum declined gradually with age, whereas PRRSV-specific antibody level increased initially (1 to 2 weeks old) and then declined with age (2 to 4 weeks old). In summary, we investigated the difference in CSFV/PRRSV antibody levels among piglets at various weeks old (1 to 4 weeks) to further establish the duration of maternal immunity in piglets. In addition, routine monitoring of CSFV/PRRSV antibodies in immunized pigs was carried out to evaluate the efficacy of vaccination.
