Opis:
Objectives Perfluorooctane sulfonate (PFOS) and zinc oxide nanoparticles (ZnO-NPs) are frequently detected in the environment but few studies have assessed the joint toxicity of them. Oxidative stress and apoptosis to zebrafish (Danio rerio) embryos induced by the PFOS and ZnO-NPs were investigated in this study. Material and Methods The embryos were exposed to the PFOS (0, 0.4, 0.8 and 1.6 mg/l), ZnO-NPs (12.5, 25, 50 mg/l) and PFOS plus ZnO-NPs (0.4+12.5, 0.8+25 and 1.6+50 mg/l) mixture solutions until 96 h post-fertilization. Activities of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (Gpx) and malondialdehyde (MDA) content were measured in zebrafish embryos after exposure lasting for 96 h. At the same time, the genes expression related to reactive oxygen species (ROS) generation, oxidative damage and apoptosis were also measured. Results A significant induction of the ROS accompanied by the increase in the activity of the Gpx and MDA contents were found in co-treatment groups. Furthermore, several apoptosis pathway related genes such as Bax, p53, caspase-3 and caspase-9 were significantly up-regulated in the PFOS plus ZnO-NPs exposure groups, while anti-apoptotic gene Bcl-2 was significantly down-regulated in the PFOS plus ZnO-NPs exposure groups. In addition, some oxidative stress-related genes such as Cat, GSH peroxidase 1 (Gpx1a) and superoxide dismutase 1 (Sod1) were also significantly down-regulated after the PFOS plus ZnO-NPs co-treatments. Conclusions The results demonstrated that the PFOS plus ZnO-NPs co-exposure could cause more serious oxidative stress and apoptosis than the PFOS and ZnO-NPs exposure alone at the exposure concentrations above. The synergistic effects of the PFOS and ZnO-NPs may be the important mechanisms of their toxicity to zebrafish embryos. Int J Occup Med Environ Health 2017;30(2):213–229

Background. Polyethylenimine (PEI) plays important roles in the pharmaceutical design of non-viral gene delivery systems. Due to a set of unique physicochemical properties this cationic polymer has a great potential in modern gene therapies. Objective. The aim of the present study was to determine the distribution of branched PEI (0.8 kDa) in zebrafish embryos (Danio rerio). Material and methods. Zebrafish embryos at 3 hours post-fertilization (hpf) were incubated with PEI (10 μg/ml) for 24 and 48 hours and studied using the confocal laser microscopy. Results. The obtained results show that PEI effectively distributed into the layers of the chorion and yolk sac in developing embryos due to first 24 hours of exposure. In contrast, PEI was found in the yolk, head, trunk and tail of the embryos due to prolonged treatments (48 hours). Conclusion. The study evidences a high distribution of the branched PEI (0.8 kDa) polymer in the zebrafish embryo tissues.
Wprowadzenie. Polietylenoimina (PEI) odgrywa ważną rolę w projektowaniu niewirusowych transporterów genów. Ze względu na unikalne właściwości fizykochemiczne, ten kationowy polimer posiada duży potencjał aplikacyjny w nowoczesnych terapiach genowych. Cel badań. Celem niniejszego badania było określenie dystrybucji rozgałęzionego PEI (0,8 kDa) w zarodkach Danio pręgowanego (Danio rerio). Materiały i metodyka. Trzy godziny po zapłodnieniu zarodki Danio pręgowanego inkubowano z PEI (10 μg/ml) przez 24 godziny i 48 godzin po czym badano przy użyciu konfokalnej mikroskopii laserowej. Wyniki. Wykazano, że PEI ulega dystrybucji w warstwach chorionu oraz żółtku rozwijającego się zarodka w pierwszych 24 godzinach ekspozycji. W wyniku przedłużonej ekspozycji (48 godzin), wykazano obecność PEI w żółtku, głowie, tułowiu i ogonie zarodków. Wnioski. Badania wskazują na znaczącą dystrybucję rozgałęzionego polimeru PEI (0,8 kDa) w tkankach zarodka Danio pręgowanego.

The zebrafish (Danio rerio) has become known as an excellent model organism for studies of vertebrate biology, vertebrate genetics, embryonal development, diseases and drug screening. Nevertheless, there is still lack of detailed reports about usage of the zebrafish as a model in veterinary medicine. Comparing to other vertebrates, they can lay hundreds of eggs at weekly intervals, externally fertilized zebrafish embryos are accessible to observation and manipulation at all stages of their development, which makes possible to simplify the research techniques such as fate mapping, fluorescent tracer time-lapse lineage analysis and single cell transplantation. Although zebrafish are only 2.5 cm long, they are easy to maintain. Intraperitoneal and intracerebroventricular injections, blood sampling and measurement of food intake are possible to be carry out in adult zebrafish. Danio rerio is a useful animal model for neurobiology, developmental biology, drug research, virology, microbiology and genetics. A lot of diseases, for which the zebrafish is a perfect model organism, affect aquatic animals. For a part of them, like those caused by Mycobacterium marinum or Pseudoloma neutrophila, Danio rerio is a natural host, but the zebrafish is also susceptible to the most of fish diseases including Itch, Spring viraemia of carp and Infectious spleen and kidney necrosis. The zebrafish is commonly used in research of bacterial virulence. The zebrafish embryo allows for rapid, non-invasive and real time analysis of bacterial infections in a vertebrate host. Plenty of common pathogens can be examined using zebrafish model: Streptococcus iniae, Vibrio anguillarum or Listeria monocytogenes. The steps are taken to use the zebrafish also in fungal research, especially that dealing with Candida albicans and Cryptococcus neoformans. Although, the zebrafish is used commonly as an animal model to study diseases caused by external agents, it is also useful in studies of metabolic disorders including fatty liver disease and diabetes. The zebrafish is also a valuable tool as a model in behavioral studies connected with feeding, predator evasion, habituation and memory or lateralized control of behavior. The aim of the present article is to familiarize the reader with the possibilities of Danio rerio as an experimental model for veterinary medicine.

A higher sensitivity and the cytotoxic effects of garlic have been previously reported on culture cells. No studies, however, have investigated the developmental toxicity of fresh garlic in zebrafish model. In this study freshly made garlic juice, which chemically is a combination of sulphur containing compounds, was evaluated to investigate the developmental toxicity during a 5-day assay using zebrafish embryos, starting from 2 h post fertilization. The results showed that toxicity endpoints such as the hatching rate, survival rate, malformation rate and growth rate had a significant dose-response relationship with increasing concentrations of garlic juice. The average hatching rate and time to hatch of zebrafish exposed to lower concentrations (0.001% and less) were significantly increased around 48-60 h of treatment (P < 0.05), strongly emphasizing the acceleration of hatchability caused by garlic juice. Mortality was also increased with exposure to garlic, exhibiting the maximal toxicity after 12 h of exposure to the concentration of 0.08%. A significant reduction in the heart rate by 35.5% was recorded due to the higher concentrations of juice (above 0.01%) starting form 36-hour exposure (P < 0.05). Growth retardation, pericardial oedema with heart malformations, deformity of yolk, lack of pigmentation and body malformation have been noticed at the concentration of 0.03% starting from 48 h of exposure. No severe malformations in any of the experimental zebrafish were produced at concentrations less than 0.01% of garlic juice. The results indicate that developing zebrafish are sensitive to garlic juice and further studies are required to investigate the molecular basis of the observed effects that garlic constituents exert upon aquatic vertebrates.

The effect of dietary Echinacea purpurea (EP) on the response of zebrafish (Danio rerio) to a Flavobacterium columnare vaccine was investigated. Two hundred D. rerio with an average weight of 290 ± 40 g were selected and fed different levels of E. purpurea (5 g kg⁻¹ diet - group 1, 10 g kg⁻¹ diet - group 2, 20 g kg⁻¹ diet - group 3, 30 g kg⁻¹ diet - group 4, and 0 g kg⁻¹ diet - group 5). Experimental feeding was begun 3 weeks prior to bath immunization and continued until the end of the experiment. Twenty-eight days after immunization the fish were challenged by bath immersion with F. columnare at a concentration of 1x106 CFU/ml. The relative percent survival of the experimental groups (1, 2, 3, 4 and 5) was 5.0, 6.0, 30.0, 36.0 and 5.0, respectively. In conclusion, diet supplementation with E. purpurea may effectively enhance the response of zebrafish to a F. columnare vaccine.

Developmental abnormalities due to fresh garlic (Allium sativum L.) juice exposure have been previously reported on zebrafish (Danio rerio) embryos. Freshly made garlic juice (FGJ) containing allicin as a main ingredient and aged garlic extract (AGE) containing mainly S-allyl-L-cysteine were evaluated to investigate the lethality during a 5-day assay using zebrafish embryos, starting from 2 hours post fertilization (hpf), in a static system at an garlic concentration range of 0.00001-0.1%. In addition, lethality comparison between embryos and 3 days of post fertilization (dpf) larvae was performed. The results indicate that the mortality rate is positively correlated to the concentration of FGJ. Lethal concentration for the 24-h LC20 and 24-h LC50 in zebrafish embryos exposure was determined as 0.034% and 0.021%. The corresponding values for larval forms were significantly higher, 0.081% and 0.073% (P < 0.01), suggesting a greater sensitivity of developing embryos to FGJ treatment, as well as no protection from an embryo’s chorion. Below the FGJ concentrations of 0.02% (for embryos) and 0.06% (for larvae), no increase in mortality was observed over the entire exposure time. The mortality of AGE-treated embryos did not differ significantly (P = 0.571) from the control regardless of exposure duration and AGE concentration. The overall results indicate that zebrafish embryos constitute a reliable model for testing the developmental toxicity of garlic, and that different types of garlic preparations have different pharmacologic properties.

The aim of the study was to compare the acute toxicity of diclofenac to juvenile and embryonic stages of the zebrafish (Danio rerio). Acute toxicity tests were performed on the aquarium fish Danio rerio, which is one of the model organisms most commonly used in toxicity testing. The tests were performed using a semi-static method according to OECD guideline No. 203 (Fish, acute toxicity test). Embryo toxicity tests were performed in zebrafish embryos (Danio rerio) in compliance with OECD No. 212 methodology (Fish, short-term toxicity test on embryo and sac-fry stages). The results were subjected to a probit analysis using the EKO-TOX 5.2 programme to determine 96hLC50 and 144hLC50 (median lethal concentration, 50% mortality after a 96 h or 144 h interval, respectively) values of diclofenac. The statistical significance of the difference between LC50 values in juvenile and embryonic stages of Danio rerio was tested using the Mann-Whitney non-parametric test implemented in the Unistat 5.1 programme. The LC50 mean value of diclofenac was 166.6 ± 9.8 mg/L in juvenile Danio rerio, and 6.11 ± 2.48 mg/L in embryonic stages of Danio rerio. The study demonstrated a statistically higher sensitivity to diclofenac (P<0.05) in embryonic stages compared to the juvenile fish.

The aim of this study was to investigate the effects of subchronic exposure to atrazine on fish growth and the development of histopathological changes in selected organs (gill, kidney, liver) in Danio rerio. Juvenile growth tests were performed on D. rerio according to OECD method No. 215. For 28 days, fish at an initial age of 30 days were exposed to the environmental atrazine concentration commonly detected in Czech rivers (0.3 μg/L) and a range of sublethal concentrations of atrazine (3.0, 30.0 and 90.0 μg/L). The results showed decreasing growth rates and morphological changes in the liver (dystrophic lesions of hepatocytes) at 90.0 μg/L of atrazine. The environmental concentration of atrazine in Czech rivers did not have any effect on fish growth and development of histopathological changes in D. rerio. The value of NOEC was 30.0 μg/L and the value of LOEC was 90.0 μg/L.

The effects of four different kinds of nanoparticles (NPs), namely, CuO, ZnO, TiO2, and Au, of the sizes ranging from <20 nm to 50 nm on Daphnia magna, early life stage of zebrafish, and various enzymes have been investigated. The experimental results showed that the NPs inhibited both the body length and hatching rate of zebrafish larvae; the small nanoparticles exhibited more toxicity. In a 21 day chronic toxicity test, metal ions of higher concentrations significantly reduced the number of Daphnia magna offspring. Studies on enzyme activity showed that the NPs reduced the glutathione content and inhibited catalase and superoxide dismutase activities, resulting in shorter body length, lower hatching success, and lower reproduction of zebrafish larvae. Therefore, studies should focus more on the potential toxicity of smaller NPs.

The aim of this work was to evaluate lethal and sublethal effects of benzo(a)pyrene, phenanthrene and phenanthrene derivatives (1-methylphenanthrene, 4-methylphenanthrene, 1-phenylphenanthrene and 4-phenylphenanthrene) on zebrafish (Danio rerio) larvae. We conducted acute toxicity test, using 96h static renewal exposure to a series of the PAH concentrations (0.05, 0.50, 5.00, 50.00µmol*l-1), to determine the No Effect Concentration (NEC) value for each compound examined. The mean NEC estimates obtained in the study were 5.16۪.45µmol*l-1 (B[a]P), 4.88۪.13µmol*l-1 (Ph), 40.24䔰.93µmol*l-1 (1P-Ph), 47.92ۭ.61µmol*l-1 (1M-Ph), 24.31۱.33µmol*l-1 (4P-Ph) and 3.11۫.01µmol*l-1 (4M-Ph) and suggested the following order of PAH toxicities on Danio rerio larvae: 4M-Ph>Ph˜B[a]P>4PPhP-Ph>1M-Ph. To gain insight into possible molecular mechanisms of apparent toxicity of phenanthrene derivatives on zebrafish larvae, we examined mRNA expression of cyp1a, cyp1b1, and vtg genes in the larvae exposed for 48h to a PAH concentration of 0.50µmol*l-1. Whereas the larvae exposed to each tested PAH displayed many developmental abnormalities (i.e. pericardial and yolk sac edema, dorsal curvature, or tail malformations), no significant upregulation of cyp1a and cyp1b1 mRNA was observed, except for zebrafish exposed to B[a]P. However, significant reduction of vtg mRNA was observed in the larvae exposed to phenanthrene and its 4P- derivative. The results may contribute to the development of a new knowledge about effects of structurally diverse phenanthrene derivatives on vertebrate organisms.
