    Diverse carbonaceous microfossils, including exceptionally preserved remains of non−biomineralizing metazoans, are reported from a basal middle Cambrian interval of the Kaili Formation (Guizhou Province, China). The application of a gentle acid maceration technique complements previous palynological studies by revealing a larger size−class of acritarchs, a richer assemblage of filamentous microfossils, and a variety of previously unrecovered forms. Metazoan fossils include Wiwaxia sclerites and elements derived from biomineralizing taxa, including chancelloriids, brachiopods and hyolithids, in common with previously studied assemblages from the early and middle Cambrian of Canada. In addition, the Kaili Formation has yielded pterobranch remains and an assemblage of cuticle fragments representing “soft−bodied” worms, including a priapulid−like scalidophoran. Our results demonstrate the wide distribution and palaeobiological importance of microscopic “Burgess Shale−type” fossils, and provide insights into the limitations and potential of this largely untapped preservational mode.

    Sound velocity, densities of binary mixture of Tetrahydropyran (THP) with 1-hexanol has been measured over the entire range of composition at T = 298.15 to 318.15 K. The excess parameters viz., excess sound velocity, deviations in isentropic compressibility, excess molar volume, excess free length and excess acoustic impedance are deduced from experimental values and discussed intermolecular interactions present in the mixture. At the end all the parameters have been fitted to Redlich-Kister equation and their coefficients are obtained.

    Exocyclic adducts of DNA bases, such as etheno- and hydroxyalkano- ones, are generated by a variety of bifunctional agents, including endogenously formed products of lipid peroxidation. In this work we selectively modified cytosines in the 5'-d(TTT TTT CTT TTT CTT TTT CTT TTT T)-3' oligonucleotide using: chloroacetaldehyde to obtain 3,N4-α-hydroxyethano- (HEC) and 3,N4-etheno- (εC), acrolein to obtain 3,N4-α-hydroxypropano- (HPC) and crotonaldehyde to obtain 3,N4-α-hydroxy-γ-methylpropano- (mHPC) adducts of cytosine. The studied adducts are alkali-labile which results in oligonucleotide strain breaks at the sites of modification upon strong base treatment. The oligonucleotides carrying adducted cytosines were studied as substrates of Escherichia coli Mug, human TDG and fission yeast Thp1p glycosylases. All the adducts studied are excised by bacterial Mug although with various efficiency: εC >HEC >HPC >mHPC. The yeast enzyme excises efficiently εC ł HEC >HPC, whereas the human enzyme excises only εC. The pH-dependence curves of excision of εC, HEC and HPC by Mug are bell shaped and the most efficient excision of adducts occurs within the pH range of 8.6-9.6. The observed increase of excision of HEC and HPC above pH 7.2 can be explained by deprotonation of these adducts, which are high pKa compounds and exist in a protonated form at neutrality. On the other hand, since εC is in a neutral form in the pH range studied, we postulate an involvement of an additional catalytic factor. We hypothesize that the enzyme structure undergoes a pH-induced rearrangement allowing the participation of Lys68 of Mug in catalysis via a hydrogen bond interaction of its ε-amino group with N4 of the cytosine exocyclic adducts.

    W artykule przedstawiono nowy ręczny przyrząd do pomiaru parametrów fizycznych powietrza THP-2/R przeznaczony do okresowego monitorowania parametrów i zagrożeń wentylacji. Zamieszczono w nim informacje dotyczące działania urządzenia, tryby pracy, możliwości pomiarowe oraz krzywe kalibracyjne czujnika ciśnienia. Urządzenie posiada niezbędną dokładność pomiaru parametrów powietrza (ciśnienie bezwzględne, różnica ciśnień, temperatura i wilgotność) do obliczenia pola potencjałów aerodynamicznych. Przedstawione urządzenie może być wykorzystywane przy obliczeniach wentylacyjnych oraz przy wykrywaniu zagrożeń i akcji podejmowanych w celu likwidacji ich skutków.
    A new hand-held instrument of type THP-2/R for measurement of physical parameters of the air designed for periodic monitoring of ventilation parameters and related hazards has been presented in the paper. The paper includes information on operation of the instrument, modes of operation, measuring potential, and calibration curves of a pressure sensor. The instrument provides for essential measuring accuracy of the air parameters (absolute pressure, differential pressure, temperature, humidity) needed for calculation of aerodynamic potentials. The presented instrument may be used for calculations relating to ventilation as well as for detection of hazards and operations aiming at liquidation of effects of the hazards.

    Cyclic guanosine monophosphate (cGMP) is synthesized by two types of enzymes: particulate (membrane-bound) guanylyl cyclases (pGCs) and soluble (cytosolic) guanylyl cyclases (sGCs). sGCs are primarily activated by binding of nitric oxide to their prosthetic heme group while pGCs are activated by binding of peptide ligands to their extracellular domains. One of them, pGC type A (GC-A) is activated by atrial and brain natriuretic peptides (ANP and BNP, respectively). Human monocytes isolated from peripheral blood mononuclear cells have been found to display sGC expression without concomitant expression of GC-A. However, GC-A activity appears in monocytes under certain conditions but a molecular mechanism of GC-A expression is still poorly understood. In this report we show that phorbol ester (PMA) induces transcription of a gene encoding GC-A in human monocytic THP-1 cells. Moreover, we find that PMA-treated THP-1 cells raise cGMP content following treatment with ANP. Studies using pharmacological inhibitors of protein kinases suggest involvement of protein kinase C (PKC), mitogen extracellular kinases (MEK1/2), and extracellular signal-regulated kinases (ERK1/2) in PMA-induced expression of the GC-A encoding gene in THP-1 cells. Finally, we show that PMA stimulates binding of Sp1 transcription factor to GC-rich DNA sequences and mithramycin A (a selective Sp1 inhibitor) inhibits expression of the GC-A mRNA in PMA-treated THP-1 cells. Taken together, our findings suggest that the PMA-stimulated PKC and MEK/ERK signaling pathways induce Sp1-mediated transcription of the GC-A encoding gene in human monocytic THP-1 cells.
