Opis:
Swine influenza virus (SIV) of H1N1 and H3N2 subtypes are dominated in European pigs population. "Classical swine" H1N1 subtype was replaced by "avian-like" H1N1 subtype. It co-cir- culates with H3N2 reassortant possessing "avian" genes. In the present study, 41 SIV strains isolated from pigs with pneumonia, raised in 20 Polish farms, were identified and characterised. Since it was evidenced that isolates from the same geographic district and the same year of isolation are in 100% similar, 15 strains representing different district and different year of isolation were chosen to construct phylogenetic trees. Two genes, conservative matrix 1 (Ml) and the most variable, haemagglutynin (HA), were sequenced and subjected into phylogenetic analysis. The results of the analysis confirmed that "avian-like" swine H1N1 strains evolved faster than classical SIV strains. HA gene of these isolates have been derived from contemporary strains of "avian-like" SIV. In contrast, the Ml gene segment may have originated from avian influenza viruses. H3N2 strain is located in swine cluster, in the main prevalent European group of H3N2 isolates called A/Port Chalmers/l/73-like Eurasian swine H3N2 lineage, which has evolved separately from the human H3N2 virus lineage around 1973.

Introduction and objectives. The parasite Cryptosporidium spp. is an intracellular protozoa which has a broad range of hosts and zoonotic potential. It presents a serious health risk for agricultural workers and veterinarians. The aim of the study was to identify the species and subtypes of Cryptosporidium occurring in a veterinary student who came into contact with calves on a farm. Materials and method. The Ziehl-Neelsen staining technique was employed to confirm the presence of Cryptosporidium oocysts. ELISA test was applied to detect coproantigen in faecal specimens. Nested PCR was used to amplify a small ribosomal subunit (SSU rRNA) and sequencing of the GP60 gene served to identify the zoonotic subtypes. Results. The nested PCR allowed to confirm the C. parvum species; subsequently, the IIdA15G1 zoonotic subtype was identified. Conclusion. This is the first confirmed case in Slovakia of human cryptosporidiosis caused by the unique subtype IIdA15G1.

The design of drugs with bioinformatics methods to identify proteins and peptides with a specific toxic action is increasingly recurrent. Here, we identify toxic proteins towards the influenza A virus subtype H1N1 located at the UniProt database. Our quantitative structure-activity relationship (QSAR) approach is based on the analysis of the linear peptide sequence with the so-called Polarity Index Method that shows an efficiency of 90% for proteins from the Uniprot Database. This method was exhaustively verified with the APD2, CPPsite, Uniprot, and AmyPDB databases as well as with the set of antibacterial peptides studied by del Rio et al. and Oldfield et al.

Opis:
In order to determine the genetic variability of Polish RHD virus strains and to confirm the presence of genetic variant (RHDVa) subtype the partial nucleotide sequences of capsid protein gene, including two highly variable regions C and E, were examined. Phylogenetic analyses of 15 viral strains obtained over 18 years revealed the presence of three genetic groups. The oldest RHDV strains exhibit very close amino acid sequence similarity (98-99%) to the German FRG89 reference strain and most of European strains of the same period, as well as Chinese isolate from 1984. The HA-negative strains and isolates with variable reactivity in the HA test belong to the second subgroup and exhibit an intermediate level of variability (about 3%) in the analysed VP60 gene fragment. The most genetically variable strains (6-7%) clustered to RHDVa subtype. The analysis of nucleotides and amino acid sequences demonstrated three pairs of well conserved RHDV strains, isolated over 3, 6 and 10-year period.

Opis:
Blastocystis spp. is a parasite that causes intestinal infection in humans and other animals. A few studies have been performed in Turkey on the distribution of Blastocystis in cattle. In this study, fecal samples were collected from 100 calves and subjected to analysis based on an SSU rRNA gene fragment. The overall prevalence of the disease was determined as 15% (15/100). This rate was 14.04% for females and 16.28% for males. In addition, three Blastocystis subtypes were identified: ST10, ST14, and novel subtypes ST25. To our knowledge, the ST25 subtype was reported with this study for the first time in Turkey. The nucleotide sequences (OM920832-OM920839) obtained in this study were deposited in GenBank. The results obtained will be useful for a better understanding of the epidemiology of Blastocystis spp., and its effects on public health.

Purpose:To examine lipid peroxidation, antioxidant protection and L-arginine-NO – system in patients with papulo-pustular rosacea.Materials and methods:The study included 128 women with papulo-pustular rosacea aged 40.1±0.99 years (range: 18-68 years). The patients were divided into three groups based on the severity of the symptoms: group I –patients with mild rosacea (n=42), group II – patients with moderate rosacea (n=49), group III – patients with severe rosacea (n=37). Indicators of lipid peroxidation and antioxidant protection were defined in all patients by a spectrophotometric method. Results:The first group of patients showed a significant decrease in superoxide dismutase (SOD) and ceruloplasmin when compared to the control group. A significant increase in diene conjugates (DC), malondialdehyde (MDA), liposoluble antioxidants (retinol, α-tocopherol) and decrease in SOD, catalase and ceruloplasmin were observed in the second group. Patients in the third group had similar dynamics with a worsening of lipid peroxidation. Conclusions: The changes in some parameters of lipid peroxidation and antioxidant protection were revealed in patients with papulo-pustular rosacea. The nature of these changes depends on the severity of the disease. Evaluation of the antioxidant imbalance may be informative to determine the understanding of the genesis of dermatosis and to study therapeutic strategy aimed at reducing the generation of reactive oxygen species (ROS), leading to a decrease in the capacity of the antioxidant defense.

Opis:
This work focused on the analysis of various gene expression-based cancer subtype classification approaches. Correctly classifying cancer subtypes is critical for understanding cancer pathophysiology and effectively treating cancer patients by using gene expression data to categorize cancer subtypes. When dealing with limited samples and high-dimensional biological data, most classifiers may suffer from overfitting and lower precision. The goal of this research is to develop a machine learning (ML) system capable of classifying human cancer subtypes based on gene expression data in cancer cells. These issues can be solved using ML algorithms such as Transductive Support Vector Machines (TSVM), Boosting Cascade Deep Forest (BCD Forest), Enhanced Neural Network Classifier (ENNC), Deep Flexible Neural Forest (DFN Forest), Convolutional Neural Network (CNN), and Cascade Flexible Neural Forest (CFN Forest). In inferring the benefits and rawbacks of these strategies, such as DFN Forest and CFN Forest, the findings are 95%.

Opis:
Blastocystis sp. is a commonly reported enteric protistan parasite in faecal specimens with a worldwide distribution afflicting both humans and a wide range of animals. The aim of this study is to characterize the subtypes (STs) of Blastocystis sp. isolates from asymptomatic individuals in an urban community in Pateros, Metro Manila, Philippines. The 600-bp small subunit ribosomal RNA (SSU rRNA) barcoding region of Blastocystis sp. isolates was amplified and sequenced using the primers RD5 and BhRDr. Subtypes were identified by uploading the sequences onto the Basic Local Alignment and Search Tool (BLAST) websites, the Blastocystis Subtype (18S) and Sequence Typing (MLST) Database and by construction of a phylogenetic tree. Twenty-nine (29) out of 35 individuals were detected positive for Blastocystis sp. ST3 is the most common among the three STs detected (65.5%), followed by ST1 (31.0%) and ST4 (3.44%). This study showed that DNA barcoding can serve as a helpful tool to investigate the diversity of Blastocystis sp. in the Philippines.
