Opis:
The squid hepatopancreas, containing 20-60% lipids, is not suitable on board vessel for producing fodder meal and is often discarded, although it constitutes the major part of the viscera and is a rich source of proteolytic enzymes. The proteolytic activity against hemoglobin at pH 3.0 of the extract of the he pato pan ere a s of frozen stored squid, calculated per mg of protein, is about 20 times higher than that of the extract of the rest of the viscera. Due to incubation of the homogenate at pH 3.0 at 30 and 40°C in 2 days a clear watery phase was formed, well separated from the oily fraction containing floating solid particles of the autolysate. In 2 days about 35 and 50% of the protein was hydrolysed at 20°C and 30-40°C, respectively, to degradation products not precipitated by TCA. The proteolytic activity of the autolyzed homogenate after 2 days at pH 3.0 is about 80 % of that of the original homogenate. Incubation at 40°C at pH 3.0 decreased the activity after 6 days by about 50%. In the autolysate of squid hepatopancreas the proteolytic activity at pH 3.0 in the absence of reducing compounds was almost totally due to aspartic proteases. In the presence of dithiothreitol the activity increased by about 50%, but the increase was only partly due to the presence of thiol proteases.
Zbadano aktywność proteolityczną ekstraktów enzymów z wnętrzności mrożonych kalmarów Illex argentinus wobec hemoglobiny przy pH 3 oraz określono wpływ temperatury i czasu inkubacji na stabilność enzymów i autolizę białek homogenatu z wątrobo-trzustki. Proteolityczna aktywność przy pH 3 ekstraktu enzymów z wątrobo-trzustki kalmarów jest w przeliczeniu na 1 mg białek około 20 razy większa niż aktywność ekstraktów z pozostałych wnętrzności. Po dwóch dniach inkubacji homogenatu z wątrobo-trzustki w temperaturze 20°C i 30—40°C hydrolizuje odpowiednio około 35% i 50% białek. Proteolityczna aktywność w zautolizowanym homogenacie obniża się w tych warunkach o około 20%. Aktywność proteolityczna w autolizacie wykazywana przy pH 3 w nieobecności substancji redukujących pochodzi nieomal całkowicie od proteaz asparaginowych. Autoliza białek wątrobo-trzustki kalmarów jest prostą i tanią drogą uzyskiwania częściowo oczyszczonego preparatu enzymów proteolitycznych aktywnych w kwaśnym środowisku. Jednocześnie ułatwia ona oddzielenie lipidów i części stałych od klarownego autolizatu. Autolizat może stanowić półprodukt do otrzymywania preparatu enzymów proteolitycznych o większej czystości.

Opis:
The muscles and internal organs of marine animals contain many proteases. These enzymes fulfill different metabolic functions in the living organisms and affect the sensory quality and functional properties of the catch. Their activity depends on the species, life cycle, and the feeding status of the organism. Many proteases of marine origin differ from their counterparts in terrestrial animals in being more active at lower temperature and less resistant to thermal denaturation. Of practical importance in the industry is their role in ripening of salted fish, fish sauces and marinades, modyfying fish protein concentrates, ensilaging of seafood offal, and deskinning of fishery products. The negative effects comprise mainly the early post morten quality degradation of the catch and deterioration of the rheological properties of fish gels. Proteolytic preparations form different marine sources may be used successfully in food processing and as industrial enzymes, e. g. tanning agents.

Opis:
The studies were carried out on guinea pig males. The animals were infected with 30 larvae (L3) of Anisakis simplex. After 6 hrs of invasion the animals were dissected. In homogenized pancreas and duodenal contents activities of trypsin were determined. In stomach content activities of pepsin were determined. The activities of trypsin in duodenal contents and in pancreas homogenate from infected animals were lower in comparison with the control animals. The activities of pepsin were higher in infected animals.

Numerous results on membrane lipid composition from different regions of autopsied Alzheimer's disease brains in comparison with corresponding fractions isolated from control brains revealed significant differences in serine- and ethanolamine-containing glycerophospholipid as well as in glycosphingolipid content. Changes in membrane lipid composition are frequently accompanied by alterations in membrane fluidity, hydrophobic mismatch, lipid signaling pathways, transient formation and disappearance of lipid microdomains, changes in membrane permeability to cations and variations of other membrane properties. In this review we focus on possible implications of altered membrane composition on β-amyloid precursor protein (APP) and on proteolysis of APP leading eventually to the formation of neurotoxic β-amyloid (Aβ) peptides, the major proteinaceous component of extracellular senile plaques, directly involved in Alzheimer's disease pathogenesis.

Physiological and pathological roles of cysteine proteases make them important targets for inhibitor development. Although highly potent inhibitors of this group of enzymes are known, their major drawback is a lack of sufficient specificity. Two cysteine protease covalent inhibitors, viz. (i) Z-RL-deoxo-V-peptide-epoxysuccinyl hybrid, and (ii) Z-RLVG-methyl-, have been developed and modeled in the catalytic pocket of papain, an archetypal thiol protease. A number of configurations have been generated and relaxed for each system using the AMBER force field. The catalytic pockets S3 and S4 appear rather elusive in view of the observed inhibitors' flexibility. This suggest rather limited chances for the development of selective structure-based inhibitors of thiol proteases, designed to exploit differences in the structure of catalytic pockets of various members of this family.

Cysteine proteases (CPs) are responsible for many biochemical processes occurring in living organisms and they have been implicated in the development and progression of several diseases that involve abnormal protein turnover. The activity of CPs is regulated among others by their specific inhibitors: cystatins. The main aim of this review is to discuss the structure-activity relationships of cysteine proteases and cystatins, as well as of some synthetic inhibitors of cysteine proteases structurally based on the binding fragments of cystatins.

We report our progress in understanding the structure-function relationship of the interaction between protein inhibitors and several serine proteases. Recently, we have determined high resolution solution structures of two inhibitors Apis mellifera chymotrypsin inhibitor-1 (AMCI-I) and Linum usitatissimum trypsin inhibitor (LUTI) in the free state and an ultra high resolution X-ray structure of BPTI. All three inhibitors, despite totally different scaffolds, contain a solvent exposed loop of similar conformation which is highly complementary to the enzyme active site. Isothermal calorimetry data show that the interaction between wild type BPTI and chymotrypsin is entropy driven and that the enthalpy component opposes complex formation. Our research is focused on extensive mutagenesis of the four positions from the protease binding loop of BPTI: P1, P1', P3, and P4. We mutated these residues to different amino acids and the variants were characterized by determination of the association constants, stability parameters and crystal structures of protease-inhibitor complexes. Accommodation of the P1 residue in the S1 pocket of four proteases: chymotrypsin, trypsin, neutrophil elastase and cathepsin G was probed with 18 P1 variants. High resolution X-ray structures of ten complexes between bovine trypsin and P1 variants of BPTI have been determined and compared with the cognate P1 Lys side chain. Mutations of the wild type Ala16 (P1') to larger side chains always caused a drop of the association constant. According to the crystal structure of the Leu16 BPTI-trypsin complex, introduction of the larger residue at the P1' position leads to steric conflicts in the vicinity of the mutation. Finally, mutations at the P4 site allowed an improvement of the association with several serine proteases involved in blood clotting. Conversely, introduction of Ser, Val, and Phe in place of Gly12 (P4) had invariably a destabilizing effect on the complex with these proteases.

Staphylococcus aureus is a human pathogen causing a wide range of diseases. Most staphylococcal infections, unlike those caused by other bacteria are not toxigenic and very little is known about their pathogenesis. It has been proposed that a core of secreted proteins common to many infectious strains is responsible for colonization and infection. Among those proteins several proteases are present and over the years many different functions in the infection process have been attributed to them. However, little direct, in vivo data has been presented. Two cysteine proteases, staphopain A (ScpA) and staphopain B (SspB) are important members of this group of enzymes. Recently, two cysteine protease inhibitors, staphostatin A and staphostatin B (ScpB and SspC, respectively) were described in S. aureus shedding new light on the complexity of the processes involving the two proteases. The scope of this review is to summarize current knowledge on the network of staphylococcal cysteine proteases and their inhibitors in view of their possible role as virulence factors.

Proteases (peptide hydrolases) aside from known catabolic functions and protein processing play numerous roles in many tasks imposed by a parasitic life cycle; this includes parasite immunoevasion, excystment/encystment, tissue penetration and digestion of host tissue for nutrition. They have been identified in biological systems from viruses to vertebrates. Proteases are divided into four major groups on the basis of the catalytic mechanism used during the hydrolytic process — serine, aspartic, cysteine and metalloproteinases; other ‘undefined’ or cryptic proteases may also exist. There are a number of excellent reviews covering general parasite proteinases but by far the largest number of papers in the literature report on enzymes belonging to the papain superfamily of cysteine proteinases. It turns out, however, that although many different proteases have been characterized in parasitic helminths many of them have yet to be discovered.
