Opis:
The metabolism of oxygen in aerobic organisms leads to generation of reactive oxygen species (ROS). These entities are able to oxidize almost all classes of macromolecules, including proteins, lipids and nucleic acids. The physiological level of ROS is usually regulated by antioxidant defense mechanisms. There are at least three groups of antioxidant enzymes: superoxide dismutases, catalases and glutathione peroxidases (GSH-Pxs) which neutralize ROS. The trace elements (copper, zinc and selenium) bound to the active sites of the above listed enzymes play an important role in the antioxidant defense system. In mammals, a major function of selenium (Se) and Se-dependent GSH-Pxs is to protect cells from oxidative stress. Selenium concentrations and GSH-Px activities are altered in blood components of chronic kidney disease (CKD) patients. The Se level is frequently lower than in healthy subjects and the concentration very often decreases gradually with advancing stage of the disease. Studies on red cell GSH-Px activity in CKD patients reported its values significantly lower, significantly higher and lower or higher, but not significantly as compared with healthy subjects. On the other hand, all authors who studied plasma GSH-Px activity have shown significantly lower values than in healthy subjects. The degree of the reduction decreases gradually with the progression of the disease. High inverse correlations were seen between plasma GSH-Px activity and creatinine level. A gradual decrease in plasma GSH-Px activity in CKD patients is due to the fact that this enzyme is synthesized predominantly in the kidney and thus the impairment of this organ is the cause of the enzyme's lower activity. Se supplementation to CKD patients has a slightly positive effect in the incipient stage of the disease, but usually no effect was observed in end-stage CKD. Presently, kidney transplantation is the only treatment that may restore plasma Se level and GSH-Px activity in patients suffering from end-stage CKD. A few studies have shown that in kidney recipients, plasma Se concentration and GSH-Px activity are restored to normal values within a period of 2 weeks to 3 months following surgery and thus it can be acknowledged that Se supplementation to those patients has a positive effect on plasma GSH-Px activity.

To investigate the dynamics of a planar plasma diode system (PDS), a model based on the current density equilibrium at the interface was developed. The current densities and plasma boundary variations with the potential fields were obtained by simulating a single square pulse. The variation of an observed overshoot current density with the applied voltage is presented. Planar plasma diode system was also simulated for periodic, sine, square, triangular and saw tooth voltage patterns by varying the amplitude and frequencies. A method to find the lower bound of the electron density of plasma for a specified PDS is presented. Particle-In-Cell simulation technique was used to investigate the plasma particles and electric field distributions over the anode cathode gap for different intensities of external electric fields. The system became stable after few time steps and this time depends upon the intensity and polarization of the external field.

There is a growing trend in the engineering of biomaterials, focusing on surface modifications of biomaterials to improve their mechanical strength, corrosion resistance, and biological properties. Cold plasma treatment may improve biological properties of biomaterials for biomedical applications by enhancing their integration with host tissue. This study investigated the influence of different cold plasma treatments on the surface properties of a polysaccharides- -based biomaterial to improve cell adhesion to its surface. The samples were subjected to plasma treatment using three different reactors operating at atmospheric pressure: gliding arc discharge (GAD) reactor, dielectric barrier discharge (DBD) plasma jet, and DBD surface reactor. Next, surface chemistry of the biomaterial after plasma treatment was determined by ATR-FTIR analysis. Furthermore, a cell adhesion assay on the samples was carried out using normal human skin fibroblasts (BJ cell line). The attenuated total reflection Fourier transform infrared analysis (ATR- -FTIR) showed that new potential functional groups could be formed on the material surface after plasma treatment. However, plasma treatment of the samples did not enhance cell adhesion to the surface of the polysaccharides-based biomaterial. Thus, the obtained results indicate that plasma treatment using GAD reactor, DBD plasma jet, and DBD surface reactor was not effective for surface modification and cell responses.

Cell wall proteins of Candida albicans, besides their best known role in the adhesion of this fungal pathogen to host's tissues, also bind some soluble proteins, present in body fluids and involved in maintaining the biochemical homeostasis of the human organism. In particular, three plasma factors - high-molecular-mass kininogen (HK), factor XII (FXII) and prekallikrein (PPK) - have been shown to adhere to candidal cells. These proteins are involved in the surface-contact-catalyzed production of bradykinin-related peptides (kinins) that contribute to inflammatory states associated with microbial infections. We recently identified several proteins, associated with the candidal cell walls, and probably involved in the binding of HK. In our present study, a list of potential FXII- and PPK-binding proteins was proposed, using an affinity selection (on agarose-coupled FXII or PPK) from a whole mixture of β-1,3-glucanase-extrated cell wall-associated proteins and the mass-spectrometry protein identification. Five of these fungal proteins, including agglutinin-like sequence protein 3 (Als3), triosephosphate isomerase 1 (Tpi1), enolase 1 (Eno1), phosphoglycerate mutase 1 (Gpm1) and glucose-6-phosphate isomerase 1 (Gpi1), were purified and characterized in terms of affinities to the human contact factors, using the surface plasmon resonance measurements. Except Gpm1 that bound only PPK, and Als3 that exhibited an affinity to HK and FXII, the other isolated proteins interacted with all three contact factors. The determined dissociation constants for the identified protein complexes were of 10-7 M order, and the association rate constants were in a range of 104-105 M-1s-1. The identified fungal pathogen-host protein interactions are potential targets for novel anticandidal therapeutic approaches.

Three monoclonal antibodies specific to the central cell-binding and the C- and N-terminal domains of fibronectin (FN) were used to test antigenic epitope accessibility on human plasma and cerebrospinal fibronectins. In the plasma group, the mean N-terminal FN domain immunoreactivity was about one fourth that of the cell-binding and C-terminal domains, whereas in cerebrospinal fluid they were nearly equal. In the presence of 0.5-6 M urea N-terminal domain immunoreactivity in the plasma increased 3-6-fold, but it decreased 0.7-3-fold in the cerebrospinal fluid. Analysis of fibronectin domain immunoreactivities of the cell-binding and N-terminal domains by a panel of specific monoclonal antibodies may reveal N-terminal fibronectin domain accessibility for reaction with biological partner ligand(s) and/or processes in which FN could be implicated. Such determinations may have important clinical implications.

Introduction: Inflammatory myofibroblastic tumor (IMT) is a rare condition that can mimic potentially more dangerous states such as malignant tumors. The tumor itself can also show local malignancy as well as malignant transformation. The paranasal sinus IMT is quite a rare case in the literature. The manifestation of the disease can include face swelling, nasal obstruction, epistaxis, vision acuity worsening, numbness of the face, pain. Etiology of this type of lesion still remains uncertain but there are a few assumptions on the issue: viral and genetic, as well as posttraumatic and postinflammatory. We report a case of an adult woman with IMT detected in the right maxillary sinus after endoscopic sinus surgery. Wstęp: Zapalny guz miofibroblastyczny (IMT) jest rzadkim stanem, który może naśladować potencjalnie bardziej niebezpieczne choroby, takie jak nowotwory złośliwe. Sam guz może również wykazywać miejscową złośliwość, a także złośliwą transformację. IMT zatok przynosowych jest dość rzadko opisywany w literaturze. Objawem choroby może być obrzęk twarzy, niedrożność nosa, krwawienie z nosa, pogorszenie ostrości wzroku, drętwienie twarzy, ból. Etiologia tego typu zmian nadal pozostaje niejasna, ale rozważa się następujące przyczyny powstawania guza: wirusową, genetyczną, a także pourazową i pozapalną. Niniejszy opis przypadku dotyczy dorosłej kobiety z IMT wykrytym w prawej zatoce szczękowej po endoskopowej operacji zatok. Opis:
The primary physical and chemical parameters that define the hypopolarized plasma cell membrane of malignant (cancer) cells compared to non-malignant cells reflect universal characteristics. The median value for the resting membrane potential is the constant for the Nernst equation without reference to discrepancies in ion concentrations and is identical to Boltzmann energies at 37 °C. The threshold energy defining space-time converges with access to entropic processes that are reflected in the morphology of cancer cells and tumors. Slowing of growth in cancer cell lines but not normal cells following exposure to weak (~1 to 10 μT) patterned magnetic fields occurs when the energy induced within the cell corresponds to the energy equivalent of the hypopolarized membrane potential. The optimal temporal parameters for the efficacy of these fields can be derived from Hubble‟s parameter and the transform function for “noise” or “random” patterns within the system. Quantitative solutions and experimental data indicate that the cancer cell may be dominated by entropic process that can be attenuated or blocked by temporally-structured applied magnetic fields whose intensity matches the increment of energy associated with this threshold.

The attractiveness of synthetic polymers for cell colonization can be affected by physical and chemical modification of the polymer surface. In this study, high density polyethylene (HDPE, m.w. 0.952g/cm3) and low density polyethylene (LDPE, m.w. 0.922g/cm3) were modified by an Ar plasma discharge using Balzers SCD 050 device (exposure time 10, 50, 150 and 400 seconds, discharge power 1.7W). The material was then seeded with rat aortic smooth muscle cells (RASMC; passages 8 to 9, 17 000 cells/cm3) and incubated in a DMEM medium with 10% of fetal calf serum. On day 1 after seeding, the number of initially adhered cells was significantly higher on all modified HDPE and LDPE samples. On day 2, this difference persisted in HDPE, whereas in LDPE only the values on the samples modified by 150 and 400 seconds were significantly higher. On the 5th and 7th day, there were no significant differences in cell number among all LDPE samples. However, on the HDPE foils, significant differences were still apparent on the samples modified for 400 seconds. The cell spreading areas measured on day 1 after seeding were significantly larger on all modified LDPE samples, and, on day 2, on the HDPE samples exposed for 150s. The increased cell colonization was probably due to the formation of oxygen-containing chemical functional groups in the polymer. These results suggest that the responsiveness of the cell to the changes in physiochemical surface properties was more pronounced in HDPE than in LDPE. On both types of polyethylene, the most appropriate exposure time for the enhancement of cell adhesion and growth seemed to be 150 and 400 seconds.

Opis:
Low density polyethylene (LDPE) was modified by an Ar plasma discharge and then grafted with glycine (Gly), bovine serum albumin (BSA) or polyethylene glykol (PEG). Some plasma-treated samples and samples grafted with BSA were exposed to a suspension of colloidal carbon particles (C, BSA+C). Pristine LDPE and tissue culture polystyrene dishes (PSC) were used as control samples. The materials were seeded with rat aortic smooth muscle cells and incubated in a medium DMEM with 10% of fetal bovine serum. On day 1 after seeding, the cells on LDPE modified with plasma only, Gly, BSA and BSA+C adhered in similar numbers as on PSC, while the values on non-modified and PEG-modified samples were significantly lower. On day 5, the highest cell numbers were found again on LDPE with Gly, BSA and BSA+C. On day 7, the highest number of cells was found on LDPE modified only with plasma. The latter cells also dis-played the largest cell spreading area. The increased cell colonization was probably due to the formation of oxygen-containing chemical functional groups after plasma irradiation, and also due to positive effects of grafted Gly, BSA and BSA in combination with colloidal C particles.
