Previously we have shown that aldolase (ALD; EC 4.1.2.13) is present in cardiomyocyte nuclei. Now, we focused our attention on ALD localization in smooth muscle cells. Immunocytochemical methods were used to study the subcellular localization of ALD. Aldolase was localized in the cytoplasm as well as in the nuclei. Within the nuclei ALD was located in the heterochromatin region. Native polyacrylamide gel electrophoresis followed by aldolase activity staining in gel was used to study the ALD isoenzyme pattern in porcine smooth muscle cells. Two ALD isoenzymes, A and C, were found in these cells but in the nuclei only the muscle isoenzyme was detected. To support the nuclear localization of ALD, measurement of aldolase activity in the smooth muscle cell nuclei isolated from porcine stomach was performed. The ALD activity in the isolated nuclei was detectable only after preincubation of the nuclear fraction with Triton X-100 and high concentration of KCl.

Artykuł opisuje historię badań oraz grupy zajmujące się tymi zagadnieniami przez dziesięciolecia. Ponadto, prezentuje wieloletni wkład naukowców z Instytutu Nenckiego w środowisko naukowe w Polsce i za granicą.
In 2018 the Nencki Institute of Experimental Biology in Warsaw, Poland, celebrates the 100th Aniversary of its setting-up and this article is a part of the special issue devoted to the century of a research conducted in the Institute related to the cytoskeleton, cell motility and muscle contraction. The article describes the history of the research, and groups working on the motile and contractile processes through the decades. Also, it presents a long-standing contribution of the Nencki Institute researchers to the scientific community in Poland and abroad.

In electron microscopic studies on structure of the capsule surrounding Trichinella spiralis larva the techniques have been employed which permit demonstration of proteoglycans and collagen fibers at various stages of their formation, i.e. fixation in tannic acid and staining reaction with ruthenium red. The muscle cell has been demonstrated to produce both collagen fibers and glycoproteins plus proteoglycans. The products formed inner part of the capsule, separated from the outer part by the basement membrane. The outer part of the capsule contained collagen fibers and matrix, synthesized by the interstitial tissue cells, by fibroblasts and vascular endothelium cells in particular.

Materials undergoing deformation based on various mechanisms are frequently applied to manufacturing of artificial muscle actuators. In this paper we introduce a methodological step towards more precise determination of investigation object related to such actuators. One defines a deformable cell and a deformable tissue in order to indicate which functions of such objects should be integrated for further applications. One accentuates role of nanoscale models of mechanics of materials as important on way towards design of the deformable cells.

The aim of this study was to evaluate the effect of low-energy laser irradiation, coenzyme Q10 and vitamin E supplementation on the apoptosis of macrophages and muscle precursor cells during skeletal muscle regeneration after bupivacaine-induced injury. The experiment was conducted on 75 gilts, divided into 5 experimental groups: I – control, II – low-energy laser irradiation, III – coenzyme Q10, IV – coenzyme Q10 and vitamin E, V – vitamin E. Muscle necrosis was induced by injection of 0.5% bupivacaine hydrochloride. The animals were euthanized on subsequent days after injury. Samples were formalin fixed and processed routinely for histopathology. Apoptosis was detected using the TUNEL method. The obtained results indicate that low-energy laser irradiation has a beneficial effect on macrophages and muscle precursor cell activity during muscle post-injury regeneration and protects these cells against apoptosis. Vitamin E has a slightly lower protective effect, limited mainly to the macrophages. Coenzyme Q10 co-supplemented with vitamin E increases the activity of macrophages and muscle precursor cells, myotube and young muscle formation. Importantly, muscle precursor cells seem to be more sensitive to apoptosis than macrophages in the environment of regenerating damaged muscle.

Opis:
Heme oxygenase-1 (HO-1), an inducible enzyme degrading heme to biliverdin, iron and carbon monoxide, is involved in regulation of inflammation and angiogenesis. Tin protoporphyrin (SnPPIX) and zinc protoporphyrin (ZnPPIX) are commonly used as competitive inhibitors of HO-1. We aimed to compare the effects of SnPPIX and ZnPPIX on the production of vascular endothelial growth factor (VEGF), activity of inducible nitric oxide synthase (iNOS) and cell viability. All experiments were performed on rat vascular smooth muscle cells and murine RAW264.7 macrophages treated with 3-10 μM protoporphyrins. Some cells were additionally stimulated with IL-1β or with lipopolysaccharide. After a 24 h incubation period SnPPIX and ZnPPIX significantly reduced the generation of VEGF in vascular smooth muscle cells and RAW264.7, both in resting and stimulated cells. The inhibitory potentials of both protoporphyrins on VEGF synthesis were very similar. In contrast, analysis of iNOS activity revealed that results obtained with different HO-1 inhibitors are discrepant. Generation of nitric oxide by iNOS was significantly increased by SnPPIX but strongly decreased by ZnPPIX. Similar differences were observed when cell viability was compared. SnPPIX improved the cell survival rate, whereas the same doses of ZnPPIX exerted some cytotoxic effects. In summary, SnPPIX and ZnPPIX can be used as HO-1 inhibitors in some experimental models. However, these compounds produce also HO-independent effects, which can make the interpretation of experiments very uncertain. Thus the involvement of the HO-1 pathway should be always confirmed by more specific methods.

The attractiveness of synthetic polymers for cell colonization can be affected by physical and chemical modification of the polymer surface. In this study, high density polyethylene (HDPE, m.w. 0.952g/cm3) and low density polyethylene (LDPE, m.w. 0.922g/cm3) were modified by an Ar plasma discharge using Balzers SCD 050 device (exposure time 10, 50, 150 and 400 seconds, discharge power 1.7W). The material was then seeded with rat aortic smooth muscle cells (RASMC; passages 8 to 9, 17 000 cells/cm3) and incubated in a DMEM medium with 10% of fetal calf serum. On day 1 after seeding, the number of initially adhered cells was significantly higher on all modified HDPE and LDPE samples. On day 2, this difference persisted in HDPE, whereas in LDPE only the values on the samples modified by 150 and 400 seconds were significantly higher. On the 5th and 7th day, there were no significant differences in cell number among all LDPE samples. However, on the HDPE foils, significant differences were still apparent on the samples modified for 400 seconds. The cell spreading areas measured on day 1 after seeding were significantly larger on all modified LDPE samples, and, on day 2, on the HDPE samples exposed for 150s. The increased cell colonization was probably due to the formation of oxygen-containing chemical functional groups in the polymer. These results suggest that the responsiveness of the cell to the changes in physiochemical surface properties was more pronounced in HDPE than in LDPE. On both types of polyethylene, the most appropriate exposure time for the enhancement of cell adhesion and growth seemed to be 150 and 400 seconds.
