Opis:
Multidrug resistance-associated protein (MRP1) causes cellular drug resistance in several cancer cell lines. In this paper we show that antisense oligonucleotides decrease MRP1 expression in human leukaemia cells. We investigated biological activity of a series of 12 linear phosphorothioate oligonucleotides, complementary to several regions of MRP1 mRNA. The oligonucleotides were administered to leukaemia HL60/ADR cells overexpressing MRP1 protein. Then, the level of MRP1 mRNA was determined by means of semiquantitative RT-PCR and the protein level by reaction with specific monoclonal antibodies. Some of the investigated antisense oligonucleotides decrease the expression level of the MRP1 protein by 46% and its mRNA level by 76%.

The world market constantly undergoes changes that make the companies improve the standards of their services. New technologies shorten product life cycle and put the companies, which do not apply them, at the risk of losing the market. The main tool used to develop more and more flexible approach to the production control systems, which help to gain competitive advantage, is the use of the integrated management information systems. The article attempts to present the development and evolution of a company management support systems.
W opracowaniu ukazano rozwój i ewolucję systemów wspomagania zarządzania przedsiębiorstwem. Omówiono informatyczne systemy zarządzania, takie jak: systemy klasy IC (Inventory Control), MRP I (Material Requirements Planning), MRP II (Manufacturing Resource Planning) oraz ERP (Enterprise Resource Planning).

Opis:
W artykule przyjęto tezę, zgodnie z którą wprowadzenie komputerowo wspomaganych metod zarządzania przyczynia się do poprawy skuteczności i konkurencyjności firmy. W retrospektywny i prospektywny sposób przedstawiono rozwój metod zarządzania produkcją klasy MRP/ERP, podano ich zastosowanie, wady i zalety oraz klasyfikację. Na końcu opracowania zaprezentowano współczesny (najnowszy) hybrydowy system POLCA. Koncepcja kontroli przepływu materiałów (POLCA) zawiera elementy systemu ciągnionego (pull), charakterystycznego dla JIT, oraz systemu wypychania (push) cechującego tradycyjne systemy wytwarzania. Nadto wskazano na wymierne korzyści, jakie stosowanie tej metody może przynieść przedsiębiorstwu. Scharakteryzowano również zintegrowany system informatyczny zarządzania, a w głównej mierze system komputerowy wspierający planowanie i sterowanie przedsiębiorstwem produkcyjnym.
According to the thesis presented in the article, implementing computer assisted management methods contributes to an improvement, efficiency and competitiveness of a company. Methods of the development of manufacturing management have been presented in a retrospective and prospective way also their applications, strengths and weaknesses as well as classification. At the end of the article the newest hybrid system POLCA was presented. This conception contains the elements of the Pull system, which is characteristic for JIT, and the Push system with is characteristic for traditional system of production. Furthermore, measurable advantages were indicated that can be achieved by a company, which uses the method. The article describes an integrated computer system of management and in great measure it describes a computer aided planning and control of a production enterprise.

The article presents an integrated technological system of management retrospectively, in particular the computer system supporting planning and operating a production enterprise. Furthermore, it provides, among other things, identification, application, advantages and disadvantages of MRP / ERP class systems. The major part of the article is dedicated to a description of the most advanced integrated system of management which is ERP II system. The Polish market of information systems supporting production management was analysed as well.

The article deals with the logistic strategies enabling an enterprise to create a strategy flexible in terms of business and marketing and stable in terms of manufacturing. In order to create a strategy model the following principles can be applied: shorter periods of capacity planning combined with flexible planning, SYNCRO – MRP principle, the application of forecasting in operative planning, creation with partners one of the cooperation form as supply chain, demand chain, lean supply chain, agile supply chain, leagile supply chain, and using the DBR, APS and SCP systems.
W artykule zaprezentowano rozwiązania logistyczne umożliwiające przedsiębiorstwom tworzenie elastycznych strategii biznesowych i marketingowych oraz stabilnych w środowiskach produkcyjnych. Do utworzenia modelu strategii mogą być stosowane następujące zasady: krótsze okresy planowania zdolności w połączeniu z elastycznym planowaniem, zasada Syncro-MRP, stosowanie prognozowania w planowaniu operacyjnym, utworzenie z partnerami jednej z form współpracy w łańcuchu dostaw, łańcuchu popytu, odchudzonym łańcuchu dostaw, zwinnym łańcuchu dostaw oraz wykorzystanie systemów klsy DBR, APS i SCP.

Opis:
The goal of research is to perform a cluster analisys of the Bulgarian meat industry from the point of view of the logistics development. In order to study the state and development of the meat industry logistics in Bulgarian meat processing organizations, a wide range of statistical and analytical methods are used (comparison, group average, graphical method, cluster analysis and others). The date processing in done by a computer program for statistical date processing - SPSS (Statistical Package for Social Sciences). The object of the industry is the meat processing companies in Bulgariq. According to the MFA Agro statictics for November 2010, the meat processing industry includes 65 factories for cutting red meat, 30 white meat slaughterhouses, 283 meat processing factories and the minced meat factories - meat preparations and mechanically separated meat, making a total of 378 companies in the industry. The result of the study show that logistics plays a vital role for the meat processing organizations, because of the growing demand for quality, price and speed of delivery of products. Successful management of logistics allows time saving through faster movement of information, details and products from one process to another.

Opis:
Multidrug resistance (MDR) of cancer cells poses a serious obstacle to successful chemotherapy. The overexpression of multispecific ATP-binding cassette transporters appears to be the main mechanism of MDR. A search for MDR-reversing agents able to sensitize resistant cells to chemotherapy is ongoing in the hope of their possible clinical use. Studies of MDR modulators, although they have not produced clinically beneficial effects yet, may greatly enrich our knowledge about MDR transporters, their specificity and mechanism of action, especially substrate and/or inhibitor recognition. In the present review, interactions of three groups of modulators: phenothiazines, flavonoids and stilbenes with both P-glycoprotein and MRP1 are discussed. Each group of compounds is likely to interact with the MDR transporters by a different mechanism. Phenothiazines probably interact with drug binding sites, but they also could indirectly affect the transporter's activity by perturbing lipid bilayers. Flavonoids mainly interact with ABC proteins within their nucleotide-binding domains, though the more hydrophobic flavonoids may bind to regions within transmembrane domains. The possible mechanism of MDR reversal by stilbenes may result from their direct interaction with the transporter (possibly within substrate recognition sites) but some indirect effects such as stilbene-induced changes in gene expression pattern and in apoptotic pathways should also be considered. Literature data as well as some of our recent results are discussed. Special emphasis is put on cases when the interactions of a given compound with both P-glycoprotein and MRP1 have been studied simultaneously.

It presents the pre-implementation analysis with the consideration for the specific character of the production company. The paper also presents information related to the possibilities of the management of the project of the practical implementation of the computer system and the example of the practical implementation of the system in a Metals Processing Center KOMECH.

Przedmiotem pracy jest opis narzędzia wspomagającego proces planowania potrzeb materiałowych (MRP) w przedsiębiorstwie produkcyjnym. Model został stworzony w oparciu o aplikację Microsoft Excel. Szczególną uwagę poświęcono analizie przydatności narzędzia w przypadku zmian struktur produktów oraz wystąpienia ograniczeń dostaw na rynku.
The aim of this article was to present material requirements planning system (MRP) as a useful tool that can be used for the planning of production irrespectively of the capacity of a company, changing structures of manufactured bands and cases to impose restrictions on the service providers. The main system objective is to ensure adequate quantities of raw materials and materials necessary to achieve the planned production and delivery to the customer, to maintain the lowest inventory levels and to support the planning policies relating to business scheduling of deliveries and assembly operations. Due to that, the author developed and described an application that enables the efficient and accurate calculation of material requirements for various raw materials and components depending on the demand for the final product. There were analyzed and presented the examples of scheduling orders for a lot number of products, for the procurement of limited quantities and for production processes with a variable structure of the product. In description of the methodology of calculation of MRP system, the author presented simple examples (maximum of three structural levels). However, it is shown in this article that this tool can be efficiently and effectively used for the calculation and planning a lot more complex cases, in which it will be necessary to use computer equipment.
