In search for new markers of insulin-dependent diabetes (IDDM) susceptibility we studied the CATT tetranucleotide repeat in intron 1 of tyrosine hydroxylase (TH) gene on chromosome lip, the CA repeat at T-cell receptor a chain (TCRA) locus on chromosome 14q and two CA repeats at D10S211 and D10S213 loci in the chromosome 10 region containing glutamic acid decarboxylase (GAD2) gene. Alleles at these microsatellite loci were identified in a population of diabetic children and unrelated healthy controls originating from Wielkopolska, a midwestern region of Poland. We found significant association of certain alleles at TH, TCRA and D10S211 loci with diabetes in the population under study. On the contrary, none of the alleles at D10S213 locus was associated with the disease. Our findings indicate that typing of microsatellite markers may represent useful additional tool for identifying individuals at high risk of developing IDDM. Regarding loci on chromosome 10 our data and data published by other authors may suggest the extistence of two separate regions of association with IDDM susceptibility on this chromosome.

Opis:
Alloantibodies detecting antigenic determinants called BgC1 and AmiF1 were obtained in cattle. The analysis of determinants' inheritance was done on 1754 offsprings after 72 sires and 1740 dams. The molecule-carriers of both traits were characterized using serological and physico-chemical methods. It was found that the identified determinants are markers of beta-globulin and alpha-microglobulin proteins, and of autosomal, dominant genes BgC1 and AmiF1 from independent loci, which control them.

Until recently marker chromosomes have presented a difficult diagnostic problem for cytogeneticists as well as for clinicians. Introduction of FISH to cytogenetic analysis has enabled identification of their origin giving possibility to outline specific phenotypic effects of defined marker chromosomes. Nine marker chromosomes were analysed with FISH using centromeric probes, chromosome- specific libraries and unique DNA sequences probes for PWS/AS critical region. The origin from acrocentric chromosomes was established in 6 cases. One marker was a product of maternal 11;22 translocation and two others were pericentromeric regions of chromosome 2 and 4. Among 6 markers, derived from acrocentric chromosomes, 2 consisted of pericentromeric part of chromosome 15, one was identified as mar (21) and in 3 other cases the origin could not be differentiated between chromosomes 13 and 21 or 14 and 22. Clinical consequences of marker chromosomes including the risk for chromosomal nondisjunction and trisomy 21 as well as the risk for uniparental disomy (UPD) are discussed.

The genetic similarity of three species: Septoria tritici, Stagonospora nodorum and Stagonospora avenae f. sp. triticea - important pathogens in many cereal production areas worldwide was assessed by random amplified polymorphic DNA (RAPD) assay. In preliminary research DNA of 14, 9, and 7 monopyenidios- pore isolates of S. nodorum, S. tritici, and S. a. tritícea, respectively, were amplified by PCR with four primers. Afterwards the research was focused on three mono- pyenidiospore isolates from each species studied. The isolates of each species selected for the study varied in pathogenicity and were diverse geographically. PCR with the set of 14 selected primers resulted in 99 different bands, ranged from 180 to 2500 base pairs in length. Most primers in PCR (especially RAD11, RAD31, RAD32, RAD33) revealed uniform bands for isolates of S. a. tritícea, that allow to identify this species among the others. The cluster analysis using Unweighed Pair-Group Method with Averaging (UPGMA) revealed interspecies disagreement among the isolates ranging from 32 to 53%. The intraspecies disagreement ranges were 17-20%, 38-43%, 42-44% for S. avenae f. sp. triticea, S. nodorum and S. tritici, respectively. Cluster analysis classified isolates into three homogeneous clusters. Each cluster grouped isolates of one species according to their current taxonomie ranks based on spore size, colony morphology and host ranges. In addition, two of the clusters represented by isolates of S. nodorum and S. a. tritícea were distinctly separated at a lower linkage distance from the third one comprising isolates of S. tritici. A slight inconsistency found in grouping some isolates indicates that such groupings should be done with caution. The present study indicates that the PCR- RAPD assay is of a potential use in taxonomy of Stagonospora spp. and Septoria tritici as well as in molecular identification of casual disease agents.

The results of previous research on antigenic (allotypic) specificities of immunoglobulins, alpha-globulins, beta-globulins and lipoproteins in cattle are reviewed. The suitability of heteroantibodies and alloantibodies for identification of this type of markers was analysed. New names/symbols of antigenic markers of proteins in cattle, identified at the Department of Immuno- and Cytogenetics of the National Research Institute of Animal Production (Balice/Kraków, Poland), were introduced.

Hybridization of a multi-locus DNA probe, R 18.1, to genomic DNA from poultry showed a highly polymorphic fingerprint pattern. The detected DNA fingerprints are individually specific and differ between Green Legged Patridgenous (GLP) and Rhode Island Red (RIR) stock of chickens. The average numbers of detected bands in RIR were 18.68 and in GLP - 15.33, but the average band sharing levels were 0.619 and 0.431, respectively. The level of polymorphism may be connected possibly with a higher level of inbreeding in the examined stock of chicken.

In this study we have analysed haplotypes of microsatellite markers of the CFTR gene: IVS8CA, IVS17BTA, IVS17BCA in 17 CF chromosomes of Polish origin and in 19 chromosomes of German origin bearing CF mutations other than ΔF508. In the Polish population, the G542X mutation is connected with haplotypes 16/17-28/32/38-13; in the German population, a more diverse haplotype association has been detected (23-33-13 and 16-32-13). The 1717-1G->A mutation is associated with the 15/16-7-13 haplotype in the Polish population, like the G551D mutation in Germany. The only analysed case of N1303K of Polish origin is connected with the 23-30-13 haplotype, like in the German population. One N1303K chromosome of an entirely different haplotype (16-29-17) turned out to be of Greek origin. These data suggest an ancient, Palaeolithic or Neolithic origin of these mutations in the territory of current Northern Europe.
