The rosaceous branch borer, Ospheranteria coerulescens, is an important pest of rosaceous trees. This insect feeds on the twigs and branches of living trees and causes their death. The characterization of the insect phenoloxidase (PO) is of interest when doing comparative investigations, and so that we may be able to understand its biochemical properties. When designing new methods of insect control such as the use of PO inhibitors, an understanding of the biochemical properties is fundamental. In this study, PO from hemolymph of the rosaceous branch borer was purified using ammonium sulfate precipitation, gel-filtration, and ion-exchange chromatography. The biochemical properties were characterized using l-dihydroxyphenylalanine (L-DOPA) as the specific substrate. The apparent molecular weights of the three isoforms of PO were determined by SDS-PAGE to be 85.23, 79.45, and 66.06 kDa. Optimal pH for PO activity was pH 8, and the optimal temperature was 45°C. Phenoloxidase lost less than 50% of its relative activity after a 60 min incubation at the optimal temperature. The effects of ions and chemical materials such as K+, Ba2+, Zn2+ and EDTA on PO showed that PO activity was strongly inhibited by Zn2+. The Michaelis constant (Km) and maximum velocity (Vmax) were 88.61 mM and 0.14 μmol/min, respectively. The inhibitory effects of kojic acid, 4-hexylresorsinol, and quercetin on PO were determined, and the IC50s (inhibitory concentration) were estimated as 23.31 for kojic acid, 35.75 for 4-hexylresorcinol, and 60.8 μM for quercetin. The inhibitory potency of kojic acid was 1.54 times higher than that of 4-hexylresorcinol and 2.58 times higher than that of quercetin. Phenoloxidase was effectively inhibited by 4-hexylresorcinol, and the inhibition type was competitive. The inhibition types of PO by kojic acid and quercetin were found to be mixed.

Opis:
The fifth instar larvae of multivoltine crossbreed race (PM x CSR2) of silkworm, Bombyx mori (L) were exposed to the magnetic energy of various strengths (1000, 2000, 3000 and 4000 Gauss magnetic field). The magnetization of fifth instar larvae was carried out on the first four days, for half an hour for each day before first feeding. Bioassay of total proteins was carried out on the fifth day of fifth instars. The attempt reveals influence of magnetization of Bombyx mori larvae on the total protein content level in the silk glands, fat bodies and haemolymph. The total protein content was increased with increase in the strength of magnetic field from 1000 to 4000 Gauss magnetic field. The larvae magnetized with 4000 Gauss magnetic field were found with sustained or decreased in total protein contents. Silk gland total proteins were increased from 5.901 to 17.481 percent. Total proteins of fat bodies were increased from 18 to 46.517 percent. And the total proteins of haemolymph were increased from 16.606 to 33.588 percent. Magnetization may have had influence on the increase in the levels of amino acids followed by accelerated rate of protein synthesis in the fifth instar larvae of silkworm, Bombyx mori (L) (Race: PM x CSR2). Magnetic energy should be utilized as efficiently as possible for the progression of growth of larval instars of silkworm, Bombyx mori (L).

Opis:
Total protein and activities of: alpha-amylase, alkaline phosphatase, alanine aminotransferase and aspartate aminotransferase in haemolymph of the freshwater snail Lymnaea stagnalis, naturally infected with digenean larvae were investigated. There were no any changes in these parameters in the snails infected with Cercaria tenuispina that occupies mainly haemocoel sinuses. Significant increase of activities of all examined enzymes in haemolymph of the snails infected with larvae belonging to the Furcocercariae group was ascertained. These changes were proportional to the degree of injury of the digestive gland, examined with the use of the thymol turbidity test. Total protein level was significantly higher only in haemolymph of snails with higher values of this test.

We have proved highly significant differences in glucose concentration and activity of alpha-amylase in haemolymph of two growth classes of Planorbarius corneus with width of shell smaller and equal or greater than 20 mm, respectively. Both these parameters were higher and were characterized by greater range of individual variability in snails with smaller width of shell. Any differences in levels of total protein and haemoglobin in haemolymph of these animals, in comparison with adult individuals, were not observed. Effect of infection of the pulmonary cavity of investigated snails with Chaetogaster limnaei limnaei on the examined parameters was not ascertained.
