Opis:
Experiments had been carried out on gametophytes and very young fronds of sporophytes with application of Murashige and Skoog (1962) medium. The paper described the results of 15 years in vitro experiments on 16 species of tree ferns belonging to various genera: Blechnum, Cibotium, Cyathea and Dicksonia. Genus Cyathea was represented by: C. australis (R.Br.) Domin., C. capensis (L.f.) Sm., C. cooperi (F.Muell.) Domin, C. brownii Domin, C. dealbata (G.Forest) Sw., C. dregei Kunze, C. leichhardtiana (F.Muell.) Copel., C. robertsiana (F.Muell.) Domin., C. schanschin Mart., C. smithii Hook.f. and Cyathea sp. In case of genus Dicksonia only two species were introduced into our experiments: D. fibrosa Colenso and D. sellowiana Hook.. Taxa Blechnum was presented by B. brasiliense Desv. and Cibotium by C. glaucum (Sm.) Hook. and Arn. and C. schiedei Schltdl. and Cham.. The studied species presented various responses on culture conditions depending on the level of stage of development. Time required for spores germination differed between species and took from only a few to 16 weeks. Prothalium formations showed various types of growth presented by marginal meristems. For all investigated species long term gametophyte in vitro cultures was established. Mature gametophyte possessed functional antheridia and archegonia. Spontaneous fertilization helped to establish the culture of young sporophytes. For all species the ex vitro culture in greenhouse collection was established. Manipulation of sucrose content in the medium stimulated the multiplication of gametophytes, but its lack induced formation of gemmae. Apospory was observed when culture of very young fronds was extended for 6 months and new generation of gametophytes was developed. Finally, sporophytes of 12 species were obtained and they have been growing in our greenhouse.

As continuation of investigations in to the mechanism of the role of the H1 histone, which is a crucial protein component chromosomes of all eukaryotes, transgenic tobacco plants with different levels of the H1 histone variants were examined. Tobacco has six sequential variants of the H1 histone: two major ones (H1A and H1B), constituting ca. 90% of all H1, and four minor ones (H1C, H1D, H1E and H1F), occurring in very small quantities. The following groups of plants were examined: K - control group with a full set of histone variants; -AB -with the A and B variants removed; -ABCD - with the A, B, C and D variants removed; and -CD - with the C and D variants removed. The analysis of microsporogenesis in those plants, based on preparations squeezed in acetoorcein, revealed the asynchronous course of meiosis in -AB and -ABCD plants, occurrence of chromosomal aberration, and, consequently, the formation of sterile pollen grains (accordingly: 84,4% and 81,4%). In -CD plants, the percentage of aberration and sterile pollen grains was similar to the control material. Electron microscope observations of microsporogenesis showed ultrastructural changes. In -AB and -ABCD plants, a major portion of the pollen grains were degraded. The smallest number of degraded pollen grains, in comparison with the control, was found in the -CD group.

Megasporogenesis and female gametophyte development were investigated in ovules of the everbearing strawberry Fragaria × ananassa Duch. cv. Selva. Observations of thin sections revealed that ovule development starts from the formation of a nucellus and coincides in time with the beginning of receptacle overgrowth. The most characteristic feature during nucellus differentiation is the formation of a multicellular archesporium, beginning from at least two cells. Analysis of female gametophyte development indicated that in addition to the meiotic mode, female gametophytes develop by an apomeiotic mode of Antennaria type. Asynchronous development of female gametophytes of different origin occurs. The mature, eight-nucleate, seven-celled female gametophyte of meiotic origin is cylindrical and slightly curved. It occupies the central part of the nucellus. The egg apparatus, consisting of an egg cell and two synergids, is formed in the micropylar part of the female gametophyte; the opposite chalazal pole is occupied by antipodal cells. Besides the ovule in which only one seven-celled female gametophyte finally develops, ovules with a different number of cells were observed to initiate female gametophyte development. Some ovules contain a nucellus with a tetrad of linearly arranged megaspores surrounded by enlarged cells, each of which has the potential to develop into an apomictic female gametophyte. After degeneration of some post-meiotic cells or developmentally advanced female gametophytes, some of the chalazal cells initiated female gametophyte development.

Rosaceae fruit trees are characterized by gametophytic self-incompatibility, with their production typically requiring artificial pollination or pollination tree is required in production. Both of these solutions cause reductions in production efficiency, and self-incompatibility has become a major issue in agricultural biology, and as such, has been extensively studied. In this review, we discuss the relationship between S-RNase content in the style and self-incompatibility, and the role of the SLF gene in stamen-determining factor. Considering mutations in self-compatibility-related genes and self-compatibility in polyploid fruit trees, we discuss the potential mechanisms of self-incompatibility. Based on a preliminary study of the role of pollen tube Ca2+ gradients in self-incompatibility in Pyrus, we propose a new mechanistic model of self-incompatibility taking into account the effect of Ca2+. We also discuss the potential for hormone regulation to be used to control selfincompatibility in Rosaceae fruit trees.

In F₂, F₃ and F₄ generations of one hybrid line of Lupinus varius × L. digitatus, segregation into infertile and fertile plants in the ratio 3:1 was observed. Cytoembryological analyses showed that sterility was caused by irregularities in megasporocyte formation, in megasporogenesis and megagametogenesis. The following abnormalities lead to female sterility: no megasporogenic cells separated in nucelluses; in other nucelluses with megaspore mother cells, these cells underwent vacuolation and died before reduction division. In the megaspore mother cells, in which probably meiosis occurs, the megasporocyte division is irregular; a restitution nucleus is frequently formed after reduction division and such megasporocyte develops into an embryo sac. In 8-nucleate embryo sacs a change in the nucellus polarization was observed, while in rarely encountered embryo sacs the embryo cells underwent vacuolation and then died. Female sterility in the studied segregants is determined genetically, whereas sterilization of reproduction cells is a developing process throughout the period of sporocyte and female gametophyte formation.

Colobanthus quitensis (Kunth) Bartl, produced two types very small bisexual flowers. In the Antarctic natural conditions chasmogamic and cleistogamic flowers most often form five stamina with short filaments. Two microsporangia with a three-layer wall form in the anther. Microspore mother cells, which develop into microspores after meiosis, form inside the microsporangium. Microsporocytes of Colobanthus quitensis are surrounded with a thick callose layer, the special wall. After meiosis, the callose wall is dissolved and microspores are released from the tetrad. The production of proorbicules, orbicules and peritapetal membrane, and the construction of a complex sporoderm with numerous apertural sites were observed. When microspore and pollen protoplasts underwent necrosis, probably as a result of temperature and osmotic stress, sporoderm layers formed around microspores, and the cell tapetum did not disintegrate. However, woody wall layers did not accumulate in endothecium cells.
Colobanthus quitensis, jedyna dwuliścienna roślina kwiatowa rosnąca w ekstremalnie niesprzyjających warunkach antarktycznej strefy geobotanicznej, wykształca zróżnicowane, obupłciowe kwiaty; chasmogamiczne i kleistogamiczne. Kleistogamia jest tu indukowana niską temperaturą dużą wilgotnością powietrza i silnym wiatrem. W mikrosporangiach Colobanthus quitensis różnicowała się mała liczba mikrospor, co jest cechą typową dla gatunków z kleistogamią. Mikrosporocyty tworzyły bardzo grube kalozowe ściany specjalne. Nie było jednak kalozy w ścianie oddzielającej komórkę generatywną od wegetatywnej. W rozwoju pyłku Colobanthus quitensis zaobserwowano męską jednostkę rozrodczą ze zróżnicowanymi komórkami plemnikowymi. Mniejsza komórka zawierała przede wszystkim mitochondria, a większa głównie piastydy. Dojrzały pyłek wysypywał się z mikrosporangium lub kiełkował na miejscu. Obserwowano trójkomórkowe ziarna pyłku z łagiewkami wewnątrz otwartych pylników.
