Opis:
We report the studies on the mechanism of oxidation of dopamine (DA) to 3’,4’-dihydroxyphenylacetaldehyde (DOPAL) by enzyme tyramine oxidase (EC 1.4.3.6) using kinetic isotope effects (KIE) and solvent isotope effects (SIE) methods. For kinetic studies, the selectively deuterium labelled isotopologues of dopamine, i.e., [(1R)-2H]-, and [(1S)- -2H]-DA were used. The numerical values of KIE’s for the (1R)- and (1S)-isotopologues of dopamine in the enzymatic oxidation of DA to DOPAL were determined using the non competitive spectrophotometric method. Also, the SIE’s for this reaction carried out in heavy water were obtained spectrophotometrically. Some mechanistic details of enzymatic oxidation of DA to DOPAL were discussed.

Opis:
Deuterium kinetic isotope effects (KIE’s) in the reaction of oxidation of dopamine (DA) catalyzed by the enzyme horseradish peroxidase (HRP, EC 1.11.1.7) was determined using a non-competitive spectrophotometric method. For kinetic studies, the deuterium ring labelled isotopomer [2’,5’,6’-2H3]-dopamine was synthesized using acid catalyzed isotopic exchange between authentic dopamine and heavy water. Deuterium solvent isotope effects (SIE’s) for dopamine and norepinephrine were determined separately for the enzymatic oxidation carried out in the presence or absence of hydrogen peroxide. Some mechanistic details of enzymatic oxidation of dopamine and norepinephrine to corresponding catecholchromes catalyzed by HRP were discussed.

Opis:
GPCRs are a vast family of seven-domain transmembrane proteins. This family includes dopamine receptors (D1, D2, D3, D4, and D5), which mediate the variety of dopamine-controlled physiological functions in the brain and periphery. Ligands of dopamine receptors are used for managing several neuropsychiatric disorders, including bipolar disorder, schizophrenia, anxiety, and Parkinson’s disease. Recent studies have revealed that dopamine receptors could be part of multiple signaling cascades, rather than of a single signaling pathway. For these targets, a variety of experimental and computational drug design techniques are utilized. In this work, dopamine receptors D2, D3, and D4 were investigated using molecular dynamic method as well as computational ab initio Fragment Molecular Orbital method (FMO), which can reveal atomistic details about ligand binding. The results provided useful insights into the significances of amino acid residues in ligand binding sites. Moreover, similarities and differences between active-sites of three studied types of receptors were examined.

Opis:
Addiction to drugs is one of the significant problems in many countries. Opium is a kind of drug that its use goes back to many years ago. Dopamine and serotonin are neurotransmitters involved in the process of addiction. As a result, this study aimed to investigate the blood levels of serotonin, dopamine and physical fitness factors in active and inactive men addicted to opium during their rehabilitation period. This study is a descriptive and Causal after the occurrence. A total of 34 men addicted to opium referring to rehabilitation center had been admitted voluntarily as samples were available in the study. According to a questionnaire answered by them, 16 of them did exercise during their rehabilitation period (active group) and 18 of them did not do any physical activity during this period (inactive group). 5 cc of peripheral blood samples were obtained from both groups and were tested their aerobic capacity, muscular endurance, flexibility, body composition, blood pressure and heart rate. Statistical analysis showed that blood levels of serotonin and dopamine in active group were significantly more than inactive group (p<0.05). In addition, the active group compared with the inactive group had low body fat percent, more muscular endurance, more flexibility, and low resting heart rate (p<0.05). According to the findings, we can conclude that low physical activity but as regular walking can affect blood levels of serotonin and dopamine, and health-related fitness factors of addicted men and as a non-drug treatment is helpful.

Opis:
Leukocyte adhesion to the vascular endothelium contributes to many immunological and inflammatory disorders. These processes have been shown to be mediated by bradykinin receptor type 2 (B2R) and dopamine receptor type 2 (D2R). In a previous study, we reported the formation of a B2R-D2R heterodimer, possibly altering cellular functions. Hence, in the present study, we examined the effect of co-activation of endothelial cells with B2R and D2R agonists on the interaction of these cells with neutrophils. Bradykinin, the main B2R agonist, significantly increased cell adhesion, and this effect was reversed when the endothelial cells were additionally co-treated with a selective D2R agonist, sumanirole. These results were dependent on the incubation time, showing an opposite tendency after prolonged stimulation. Significant changes in the expression of adhesion proteins, such as E-selectin and intercellular adhesion molecule 1 in endothelial cells were observed. Additionally, the cells preincubated with tumor necrosis factor-α showed decreased cell adhesion and IL-8 release after long incubation with both agonists. The modulation of cell adhesion by D2R and B2R seem to be mediated via STAT3 phosphorylation. In summary, this study demonstrated a protective role of D2R in neutrophil-endothelial cell adhesion induced by bradykinin, especially in cytokine-stimulated endothelial cells.

Sleep and wakefulness are regulated by a number of neurotransmitters and neuromodulators of the central nervous system. This paper presents the facts supporting the role of the dopaminergic mesocorticolimbic and mesostriatal systems in the regulation of sleep and wakefulness. The effects of dopamine, its agonists and dopamine transporter on wakefulness, sleep, sleep disorders and the role of dopamine in the induction of REM sleep and dreams are discussed. Mesolimbic pathways are included in a suggested model of generation of hippocampal theta rhythm, which is one of the criterial features of REM sleep.
