Opis:
In this paper, we describe how several soft computing tools can be used to assist in high throughput screening of potential drug candidates. Individual small molecules (ligands) are assessed for their potential to bind to specific proteins (receptors). Committees of multilayer networks are used to classify protein-ligand complexes as good binders or bad binders, based on selected chemical descriptors. The novel aspects of this paper include the use of statistical analyses on the weights of single layer networks to select the appropriate descriptors, the use of Monte Carlo cross-validation to provide confidence measures of network performance (and also to identify problems in the data), the addition of new chemical descriptors to improve network accuracy, and the use of Self Organizing Maps to analyze the performance of the trained network and identify anomalies. We demonstrate the procedures on a large practical data set, and use them to discover a promising characteristic of the data. We also perform virtual screenings with the trained networks on a number of benchmark sets and analyze the results.

This paper discusses various aspects that should be considered when defining and executing extraction process-related information from the source data (ERP system) to an event log. This includes trace and event selection, as well as important project decisions that should be made beforehand. The basic idea of a proposed approach is demonstrated by performing a case study which is related to a standard sales order processing, that is, business process from sales quotation through sales order and delivery to invoicing. For this processes we showed the characteristics of the event logs, and the models we can discover.

The paper shows how a certain kind of underlying representations (or deep forms) of words can be constructed in a straightforward manner through aligning the surface forms of the morphs of the word forms. The inventory of morphophonemes follows directly from this alignment. Furthermore, the two-level rules which govern the different realisations of such morphophonemes follow fairly directly from the previous steps. The alignment and rules are based upon an approximate general metric among phonemes, e.g., articulatory features, that determines which alternations are likely or possible. This enables us to summarise contexts for the different realisations.

Opis:
The widespread particularist account of the onset of molecular biology that identifies it with the discovery of the DNA structure in 1953 has been recently contested. The paper contributes to this debate by focusing on a more recent discovery of small noncoding RNAs (microRNAs). First, it outlines a particularist account of the microRNAs discovery and the origins of the particularist predilection of the modern scientometric studies of science dynamics. Next, it discusses its limitations and proposes an alternative, modified processualist account of the discovery. In the final part, the paper applies this approach to unravel network dynamics of the research on the first two microRNAs that were discovered, namely lin-4 and let-7.

Opis:
The field of health care is an important element of the economic and social life of every country in the world combining demographic, economic and epidemiological, ethical and social challenges. Spending on the development of new therapies has been increasing over the past two decades, and the amount of drugs approved by regulatory agencies has remained stable. Literature does not provide adequate knowledge about the reasons of the productivity drop that impacts the competitive advantage of the companies taking part in the project’s race to the market (Schuhmacher et al., 2022), and it therefore seems crucial to analyse the factors determining high productivity of the pharmaceutical industry to adjust further actions ensuring the highest quality of health care systems, focusing on the wellbeing of the patient and the development of increasingly safer medicines. To address this need the author performed systematic literature review followed by structured interviews with 14 experts working globally in the field of drug development to determine productivity factors in drug discovery research and development projects, with the goal of answering questions related to which factors play a key role in the productivity of scientific organisations and the relationship between the factors, providing an insight into which parts of drug discovery ecosystem can increase a chance to address highly unmet medical needs of patients waiting for novel, safe and effective forms of treatment. As a result of the research 22 key productivity factors were defined and clustered into 4 categories: scientific, managerial, business, environmental and relations between the factors were discussed.

Artykuł jest oparty na korespondencji między Udo Wid’em i Karin Hofer a Łukaszem Guzkiem i Anką Lesniak. Dotyczy przygotowań do spotkania i konferencji w Dreźnie (Ostrale). Tak więc nowa dyscyplina Art(istic) research wykorzystuje metody artystyczne do tworzenia wiedzy o świecie, podczas gdy przyszłe ArtStudies prawdopodobnie będą wykorzystywać metody epistemiczne do tworzenia wiedzy o sztuce. Oba mogą się uzupełniać.
The article is based on the correspondence between Udo Wid and Karin Hofer and Łukasz Guzek and Anka Lesniak. Concerns preparations for meetings and conferences in Dresden (Ostrale). This is an example of exchanging views and creating theories in a closed group (echo chamber). It is also an example of how the production of knowledge in the humanities works in practice and what role artistic research can play. So the new discipline of Art (istic) Research uses artistic methods to produce knowledge about the world, while future ArtStudies will probably use epistemic methods to produce knowledge about art. Both perhaps will complement each other.

Luck is one of the critical factors determining outcome of many activities we are involved in. In chemistry, and specifically in drug discovery a strike of luck can determine a successful study outcome leading to an approval of potentially blockbuster drug consequently offering a new treatment option for patients in need. On the other hand, lack of luck can often result in termination of activities, wasted resources and years of studies and often have negative impact on a fate of companies and its employees. In a similar fashion, hard work is critical for one’s success. It is not meant in a literal fashion, but rather from the perspective of “going the extra mile” in scientific research and development. It is meant as a proper approach to analysis of study outcome and understanding the reasons “why”, rather than accepting simply yes/no outcomes. Below article discusses two instances where certain level of luck and extra effort invested in understanding of widely available results led to development of two novel therapeutics in oncology field. Described stories deal with development of Abraxane, novel formulation of paclitaxel commercialized by Abraxis Bioscience as well as enzalutamid (Xtandi®) discovered at UCLA and developed and commercialized by Medivation and Astellas Pharma.
