Background: Ovarian cancer is usually diagnosed late and is one of leading causes of genital malignan-cies-related mortality in women. Thromboembolic events are a frequent complication of neoplastic disease, occurring four-fold more often in patients with this disease and six-fold more often in persons undergoing chemotherapy. The aim of paper was to answer the following questions: What is the impact of first-line chemotherapy on D-dimer level during treatment? Is there any correlation between D-dimer level and FIGO stage of ovarian cancer? Is there a correlation between D-dimer level during chemotherapy and completeness of tumour resection? Is there a correlation between D-dimer level and incidence of thromboembolic complications during chemotherapy of ovarian cancer? Material and method: Study population initially consisted of 20 women with ovarian cancer treated at the Department of Oncologic Gynaecology of the K. Marcinkowski Medical University in Poznan. Each patient underwent cytoreductive surgery with subsequent paclitaxel-plati-num chemotherapy. Peripheral blood has been obtained four times from each patient in order to assess D-dimer level, CA 125 tumour marker and blood count. Results: In persons with ovarian cancer, preoperative D-dimer level several-fold exceeded normal range. Application of chemotherapy significantly (p<0.05) reduced D-dimer level in women with ovarian cancer. Conclusions: Assessment of risk of thromboembolic complications in women with ovarian cancer during chemotherapy based on D-dimer level is not possible.
Rak jajnika jest nowotworem późno diagnozowanym i jedną z głównych przyczyn zgonów z powodu nowotworów złośliwych narządu rodnego kobiet. Incydenty zakrzepowo-zatorowe są częstym powikłaniem choroby nowotworowej, występują cztery razy częściej u pacjentów z tą chorobą i sześć razy częściej u osób leczonych chemioterapią. Cel badania: Celem badania była próba odpowiedzi na pytania: Jak chemioterapia pierwszego rzutu wpływa na stężenie D-dimerów podczas leczenia? Czy istnieje związek pomiędzy: stężeniem D-dimerów a stopniem zaawansowania raka jajnika według FIGO; stężeniem D-dimerów podczas chemioterapii a radykalnością zabiegu operacyjnego; stężeniem D-dimerów a występowaniem zaburzeń zakrzepowo-zatorowych podczas chemioterapii w raku jajnika? Materiały i metody: Grupę badaną początkowo stanowiło 20 kobiet z rozpoznanym rakiem jajnika hospitalizowanych na Oddziale Ginekologii Onkologicznej Uniwersytetu Medycznego im. K. Marcinkowskiego w Poznaniu. Każda pacjentka została poddana operacji cytoredukcyjnej z następową chemioterapią według schematu paklitaksel i pochodna platyny. Każdej pacjentce czterokrotnie pobrano krew obwodową na oznaczenie stężenia D-dimerów, markera CA 125 oraz morfologii. Wyniki: Stężenie D-dimerów u osób z rakiem jajnika przed zabiegiem operacyjnym kilkakrotnie przewyższało wartości prawidłowe. Stosowanie chemioterapii znamiennie statystycznie (p<0,05) wpływa na obniżenie stężenia D-dimerów u kobiet z rakiem jajnika. Wnioski: Ocena ryzyka wystąpienia zaburzeń zakrzepowo-zatorowych u kobiet z rakiem jajnika podczas chemioterapii na podstawie stężenia D-dimerów nie jest możliwa.

Nowadays, increasing attention has been focused on relation between increased D-dimer levels and cancer among patients without detectable thrombosis. The aim of the study was to measure plasma D-dimer levels in portal and peripheral blood in pancreatic cancer patients with absence of venous thromboembolism. Material and methods. Fifteen consecutive patients hospitalized in the Department of General and Transplant Surgery of Medical University in Łódź, from January to March 2012 who underwent surgery due to a pancreatic cancer were enrolled. At laparotomy, portal and peripheral blood were sampled concurrently. D-dimer and fibrinogen levels were measured. Moreover, to investigate overall coagulation function prothrombin time (PT), prothrombin index (PI), international normalized ratio (INR), thrombin time (TT), activated partial thromboplastin time (APTT), TT and APTT index were evaluated. Results. Peripheral plasma D-dimmer levels above normal range were found in 10/15 patients (66,67%), whereas D-dimer above normal values were confirmed in all portal blood samples. Mean D-dimer values were higher in portal than in peripheral blood (3279.37 vs 824.64, by 297%, p=0,025). These discrepancies were accompanied by normal limits of portal and peripheral levels of fibrinogen and comparable coagulation function indexes. Conclusion. Our preliminary study showed the close relation between activation of hemostasis, reflected by elevated D-dimers in portal blood and presence of pancreatic cancer. These data suggest that measurement of portal blood D-dimer levels may be a potentially useful technique for screening the pancreatic cancer.

Opis:
Intermodular interactions play an important role in many processes at the molecular level. In the contemporary science, there is a growing interest concerning the characteristics of such interactions. Therefore, the computational chemistry can provide answers to many questions, which could not be answered using experimental methods. The Symmetry-Adapted Perturbation Theory (SAPT) method was applied to characterize the energy partitioning in dimers, trimers and microsolvation models. The investigated complexes belong to various classes of compounds, e.g. • dimers of: NH3 ˑˑˑHX, HF-pyridine, cycloalkanes, hypohalous acids; • trimers of: NH3 ˑˑˑNH3ˑˑˑHF or NH3ˑˑˑHFˑˑˑHF; • microsolvation models (biotin - water molecules). The current study summarizes recent years of our research devoted to the intermolecular interactions.

Opis:
In this work we present cloning and overexpression of lactococcal CcpA protein in Escherichia coli Xl1blue strain as a fusion with 6×His tag. A high yield of the CcpA protein was obtained when the cells were cultured in liquid medium LB with 100 Μg/ml ampicillin at 37°C and subsequently for 4 h after induction by IPTG. The procedure let us obtain 5 mg of homogenous CcpA protein. Glutaraldehyde crosslinking analysis indicated the formation of dimer or tetramer forms of the CcpA protein.

Opis:
Ultrafast excited-state relaxation process has been studied with zinc porphyrin dimers and circular trimer. Following 80 fs excitation at Soret band (420 nm) or Q band (580 nm) of zinc porphyrin, the fluorescence decay curves exhibit ultrafast decays with lifetimes of 80 fs in o-dimer, 450 fs in trimer and 540 fs in m-dimer. The timeresolved fluorescence spectra show that the fast decay process correspond to disappearance of monomer-like emission followed by red-shifted and broaden spectra. These ultrafast processes are assigned as due to excitation transfer among monomers and delocalization of excitation yielding excitonic states.

The primary structure and function of nucleoside diphosphate kinase (NDK), a substrate non-specific enzyme involved in the maintenance of nucleotide pools is also implicated to play pivotal roles in many other cellular processes. NDK is conserved from bacteria to human and forms a homotetramer or hexamer to exhibit its biological activity. However, the nature of the functional oligomeric form of the enzyme differs among different organisms. The functional form of NDKs from many bacterial systems, including that of the human pathogen, Mycobacterium tuberculosis (MtuNDK), is a hexamer, although some bacterial NDKs are tetrameric in nature. The present study addresses the oligomeric property of MsmNDK and how a dimer, the basic subunit of a functional hexamer, is stabilized by hydrogen bonds and hydrophobic interactions. Homology modeling was generated using the three-dimensional structure of MtuNDK as a template; the residues interacting at the monomer-monomer interface of MsmNDK were mapped. Using recombinant enzymes of wild type, catalytically inactive mutant, and monomer-monomer interactive mutants of MsmNDK, the stability of the dimer was verified under heat, SDS, low pH, and methanol. The predicted residues (Gln17, Ser24 and Glu27) were engaged in dimer formation, however the mutated proteins retained the ATPase and GTPase activity even after introducing single (MsmNDK- Q17A, MsmNDK-E27A, and MsmNDK-E27Q) and double (MsmNDK-E27A/Q17A) mutation. However, the monomer-monomer interaction could be abolished using methanol, indicating the stabilization of the monomer-monomer interaction by hydrophobic interaction.

The EF-hand Ca^(2+)-binding protein calmyrin is expressed in many tissues and can interact with multiple effector proteins, probably as a sensor transferring Ca^(2+) signals. As oligomerization may represent one of Ca^(2+)-signal transduction mechanisms, we characterised recombinant calmyrin forms using non-reducing SDS/PAGE, analytical ultracentrifugation and gel filtration. We also aimed at identification of biologically active calmyrin forms. Non-reducing SDS/PAGE showed that in vitro apo- and Ca^(2+)-bound calmyrin oligomerizes forming stable intermolecular disulfide bridges. Ultracentrifugation indicated that at a 220 µM initial protein concentration apo-calmyrin existed in an equilibrium of a 21.9 kDa monomer and a 43.8 kDa dimer (trimeric or tetrameric species were not detected). The dimerization constant was calculated as Ka = 1.78 × 103 M^(-1) at 6oC. Gel filtration of apo- and Ca^(2+)-bound calmyrin at a 100 µM protein concentration confirmed an equilibrium of a monomer and a covalent dimer state. Importantly, both monomer and dimer underwent significant conformational changes in response to binding of Ca^(2+). However, when calmyrin forms were analyzed under non-reducing conditions in cell extracts by Western blotting, only monomeric calmyrin was detected in human platelets and lymphocytes, and in rat brain. Moreover, in contrast to recombinant calmyrin, crosslinking did not preserve any dimeric species of calmyrin regardless of Ca^(2+) concentrations. In summary, our data indicate that although calmyrin forms stable covalent dimers in vitro, it most probably functions as a monomer in vivo.
