Gut-derived adenocarcinoma Caco-2 cells were treated with sodium butyrate (NaB) at physiologically relevant concentrations. We characterized its effects on proliferation, differentiation, apoptosis, adhesion to the solid support and interleukin-8 secretion. Differentiation was determined by brush border alkaline phosphatase activity. Apoptosis was assessed by acridine orange and Hoechst stains. Differentiation and apoptosis were analyzed in both adherent and floating cell populations. The transformed Caco-2 cells did not retain their malignant phenotype in the presence of NaB. They appeared to undergo a change in the phenotype induced by NaB, as indicated by reduced proliferation, enhanced differentiation, stimulation of apoptosis leading to decreased viability of cells, and stimulation of interleukin-8 secretion. Considering all the above facts and data, we postulate that Caco-2 cells cultured in NaB supplemented medium could regain the phenotypic characteristics of the phenotype of the parent cell from which originated the Caco-2 line.

In contrast to mRNAs, ribosomal RNAs are generally not considered to be polyadenylated. Only a few recent reports describe non-abundant polyadenylated rRNA-related transcripts that have been detected and characterized in yeast and in human cells. Here we depict the phenomenon of 26S rRNA polyadenylation and degradation that was observed in shoots of Nicotiana tabaccum plants grown in the presence of cadmium. Fragments corresponding to 26S rRNA were identified using suppression subtractive hybridization during screening for genes induced in tobacco plants upon a three-week exposure to 15 µM cadmium chloride. Extracts prepared from the above-ground tissues of cadmium-treated tobacco plants were supposed to contain exclusively polyadenylated mRNAs. Surprisingly, numerous polyadenylated fragments matching parts of 26S rRNA were identified and their presence was confirmed by Northern blot and cDNA amplification techniques. To our knowledge this is the first report on rRNA polyadenylation in plants.

Opis:
Hydrogen peroxide (H2O2) is produced predominantly in plant cells during photosynthesis and photorespiration, and to a lesser extent, in respiration processes. It is the most stable of the so-called reactive oxygen species (ROS), and therefore plays a crucial role as a signalling molecule in various physiological processes. Intra- and intercellular levels of H2O2 increase during environmental stresses. Hydrogen peroxide interacts with thiol-containing proteins and activates different signalling pathways as well as transcription factors, which in turn regulate gene expression and cell-cycle processes. Genetic systems controlling cellular redox homeostasis and H2O2 signalling are discussed. In addition to photosynthetic and respiratory metabolism, the extracellular matrix (ECM) plays an important role in the generation of H2O2, which regulates plant growth, development, acclimatory and defence responses. During various environmental stresses the highest levels of H2O2 are observed in the leaf veins. Most of our knowledge about H2O2 in plants has been obtained from obligate C3 plants. The potential role of H2O2 in the photosynthetic mode of carbon assimilation, such as C4 metabolism and CAM (Crassulacean acid metabolism) is discussed. We speculate that early in the evolution of oxygenic photosynthesis on Earth, H2O2 could have been involved in the evolution of modern photosystem II.

W ramach pracy otrzymano włókninę zbudowaną z włókien o różnych średnicach i wielkości, której mikrostruktura posiada biomimetyczny charakter, tzn. składa się z włókien o średnicach zbliżonych do średnic włókien występujących w tkankach. Badania biologiczne wykazały, że włókna o niskich średnicach są gorzej tolerowane przez tkanki.
A three dimensional fibrous material, made from fibres differing in diameters and porosity, has been designed and prepared. These materials will constitute a 3D scaffold containing fibrous components mimicking the structure of natural tissue. The biological studies indicate that the fibres with bigger diameter allow for more intense and quick regeneration of surrounding tissue.

Opis:
The multixenobiotic resistance (closely related to multidrug resistance) system controls transport across the plasma membrane as a defense against toxic molecules. Multixenobiotic resistance system consists of an efflux pump, ABCB1 (also named P-glycoprotein, P-gp), and/or a molecule of the ABCC family (also named multiple resistance associated protein, MRP). ABCB1 is able to increase efflux of many low-molecular foreign molecules. Measuring system induction may be used as a biomarker of cell/organism exposure to foreign substances. Various established cell lines were tested for constitutive and induced multixenobiotic resistance proteins by Western blotting immunodetection. The pumping function was indirectly assayed with Rhodamine B by visualization of cell fluorescence in the presence of verapamil. Changes in ABC proteins were measured by flow cytometry after exposition to various perfluorinated carboxylic acids. MCF7 and HeLa cells were found to contain the highest constitutive level of both ABCB1 and ABCC1. HEK293 exhibited much less ABCB1 and no activity of pumping out Rhodamine B. The pumping activity was found to be related to the amount of the cell-type specific 170 kDa ABCB1 protein. An 8-day exposure to 10-4 M perfluorononanoic acid resulted in about 2-2.5-fold increase of ABCB1 level. That was confirmed also for short times by flow cytometry of cells exposed to perfluorinated acids and its natural congeners. Both ABCB1- and ABCC1-related fluorescence increased along with the carbon chain in acids from C6 up to C9 and decreased for C10. Measuring of multixenobiotic resistance changes in vitro induced by chemicals may be a convenient test for screening for their potential toxicity.

Interactions between materials and cells were examined through determination of cells adhesion and secretion of cytokines involved in the development of immune response.

Opis:
Previously, a stable cell suspension of cucumber tolerant to 100 µM CdCl2 was obtained (Gzyl & Gwóźdź, 2005, Plant Cell Tissue Organ Cult 80: 59-67). In this study, the relationship between the activity of antioxidant enzymes and cadmium tolerance of cucumber cells was analyzed. A cadmium-sensitive and the cadmium-tolerant cell lines were exposed to 100 µM and 200 µM CdCl2 and the activities of superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APOX) and guaiacol peroxidase (POX) were determined. In the sensitive cell line, a decrease of total activity of SOD and POX was observed, whereas the activity of CAT and APOX significantly increased in metal-supplemented medium. By contrast, in the tolerant cells, the total activity of antioxidant enzymes decreased (SOD, CAT) or was maintained at approximately the same level (APOX, POX). Moreover, a different pattern of isoenzyme activity was observed in the tolerant and sensitive cells. These results suggest that an enhanced activity of antioxidant enzymes is not directly involved in the increased tolerance to cadmium of the selected cucumber cell line.

Opis:
The influence of surface (nano)roughness of soft elastomeric materials containing nanocrystalline TiO2 on tissue cell behaviour after implantation tests was investigated. Addition of small amount (0.2wt.%) of TiO2into polymer matrix changed surface roughness giving material of well developed lamellar morphology and slightly diminished contact angle (wettability). This lamellar (fibrilar) morphology had strong influence on cell response after implantation into soft tissue indi-cating the absence of eosynophiles in tissue around implant.

Cardiovascular disease is recognized as an important clinical problem in radiotherapy and radiation protection. However, only few radiobiological models relevant for assessment of cardiotoxic effects of ionizing radiation are available. Here we describe the isolation of mouse primary cardiac endothelial cells, a possible target for cardiotoxic effects of radiation. Cells isolated from hearts of juvenile mice were cultured and irradiated in vitro. In addition, cells isolated from hearts of locally irradiated adult animals (up to 6 days after irradiation) were tested. A dose-dependent formation of histone γH2A.X foci was observed after in vitro irradiation of cultured cells. However, such cells were resistant to radiation-induced apoptosis. Increased levels of actin stress fibres were observed in the cytoplasm of cardiac endothelial cells irradiated in vitro or isolated from irradiated animals. A high dose of 16 Gy did not increase permeability to Dextran in monolayers formed by endothelial cells. Up-regulated expression of Vcam1, Sele and Hsp70i genes was detected after irradiation in vitro and in cells isolated few days after irradiation in vivo. The increased level of actin stress fibres and enhanced expression of stress-response genes in irradiated endothelial cells are potentially involved in cardiotoxic effects of ionizing radiation.

Opis:
Basophils are one of major effector cells participating in the immune response despite the fact that they constitute a small population of peripheral blood leukocytes. They are activated by cross-linking of an antigen with IgE through FcεRI and then they release a wide range of mediators, implicated in the pathogenesis of allergic diseases. Basophils are essential cells in IgG-mediated systemic anaphylaxis and currently are considered as antigen-presenting cells. This review summarizes recent studies and reports on basophils, their origin and role in the immune response, and their participation in diseases. Widening of our knowledge about basophils can help us to understand better the mechanisms of the complex processes in which those cells are involved.
Bazofile to jedne z najistotniejszych komórek uczestniczących w odpowiedzi immunologicznej, mimo iż stanowią niewielką populację we krwi obwodowej. Ulegają aktywacji w krzyżowej reakcji wiązania antygenu przez przeciwciało IgE do receptora FcεRI. Uwalniają szerokie spektrum mediatorów, co przekłada się na implikacje w patogenezie chorób alergicznych. Odgrywają także znaczącą rolę w uogólnionych reakcjach anafilaktycznych z udziałem przeciwciał IgG, a także jako komórki prezentujące antygen. Ta praca jest podsumowaniem aktualnej wiedzy o bazofilach, ich pochodzeniu i roli w reakcjach alergicznych oraz znaczeniu w jednostkach chorobowych. Poszerzanie wiedzy z tego zakresu jest niezbędne do lepszego zrozumieniu mechanizmów skomplikowanych procesów, w których bazofile uczestniczą.
