Metallic foams are currently being looked at as a new material for automobiles. The main goal of the use of metallic foams in vehicles is to increase sound dampening, reduce the weight of the automobile, and increase energy absorption in case of crashes, or in military applications, to combat the concussive force of IEDs. The metallic foams that are being looked at currently, are aluminum and its alloys due to their low density (0.4-0.9 g/cm3). In addition these foams have a high stiffness, are fire resistant, do not give off toxic fumes, are fully recyclable, have high energy absorbance, have low thermal conductivity, have low magnetic permeability, and are efficient at sound dampening, especially in comparison to light weight hollow parts. In addition, partial addition of metallic foams in hollow parts of the car will decrease weakness points usually associated with car crashes and noisy vibrations. These foams are cheap to cast by using powder metallurgy (as compared to casting of other hollow parts). The aim of the research was to describe and to assess the main mechanisms that appear in the foam structure during the compression. The development process of the finite element model of the closed cell foam microstructure is presented in the paper. The model geometry was based on the real structure research, which was carried out with the use of computed tomography. The model was built with the use of a unique computer code created to transform the scan point cloud into FE raster model based on solid 8-node elements. The experimental and numerical compression test results were compared and showed good compatibility. The stress distributions were studied to describe the main mechanisms in the structure.

In a series of studies it was shown that 1,4-dihydropyridine derivatives (1,4-DHP) show antimutagenic and anticlastogenic properties and accelerate repair of oxidant and ionising radiation generated DNA damage. Here, effects of one of 1,4-DHP compounds (sodium 3,5-bis-ethoxycarbonyl-2,6-dimethyl-1,4-dihydropyridine-4-carboxylate denoted as DHP) in X-irradiated L5178Y cells (murine lymphoma sublines, LY-R and LY-S) are reported. DHP treatment 1 h before, during and after X-irradiation gave a radioprotective effect in double strand break (DSB) repair competent LY-R cells: there was an increase in post-irradiation proliferation and cell viability as well as a slight acceleration of break rejoining as measured by the neutral comet assay. In the radiosensitive LY-S cells with impaired non-homologous end-joining system, the radioprotective effect was seen as enhanced growth and viability. There was, however, no effect on the DSB repair rate. Notably, there was no dependence of the biological effects on DHP concentration in the range of concentrations studied (1 nM - 100 mM), suggesting an all-or-none effect, as in cellular signaling induction observed in radioadaptation or bystander effect. We assume that DHP acts by decreasing fixation of radiation inflicted DNA damage, among others, by increasing the rate of DNA repair and enhancing the efficiency of checkpoint control. Direct confirmation of this assumption is necessary.

We compared the effects of bleomycin (BLM) and ionizing radiation on two sublines of murine lymphoma L5178Y (LY): LY-R, radiation resistant and LY-S, radiation sensitive. This radiosensitivity difference is related to the ability to rejoin DNA double strand breaks. LY-S cells were about two times more sensitive to BLM than LY-R, similarly as in the case of sensitivity to X rays. Since there was no difference in the P-glycoprotein-related drug transport system between the sublines, it could be expected that the enhanced sensitivity of LY-S cells to BLM was caused by the DNA repair defect. Growth disturbances in BLM treated cell populations were proportional to the lethal effect and their duration was observed until elimination of dead cells (3-6 days after 50 ěM BLM, 1 h at 37oC). There was no slow growth phase accompanied by normal viability, as previously described for X-irradiated LY-S cells. Initial DNA damage, estimated with the single cell gel electrophoresis method was linearly related to BLM dose in LY-S cells; in LY-R cells - in the low dose range (up to 10 ěM) - there was more damage than in LY-S cells, however, at higher doses the dose - effect curves became identical. The doseeffect relationship for ă rays was linear and identical in both cell sublines. DNA damage distribution in BLM treated cells was much less uniform as compared to that in irradiated cells and indicated the presence of cells with severely damaged DNA, a feature typical for BLM action in vitro.

DNA fragmentation by single strand breaks (SSBs) or double strand breaks (DSBs) is major concern in genotoxicity research. DNA damages can be easily known through comet assay or single cell gel electrophoresis (SCGE). Since decades, scoring through software for DNA damages in images have been developed by researchers. These softwares depend upon manual scoring on individual comet in a particular interface. The evaluation under software may have biasness and error during scoring by each researcher and few softwares are unable to access easily because many of these are commercial products. However, CellProfiler (CP) image analysis software (Version 2.1.0) is free, easy operation, faster and automated screening by computer itself. An attempt was made to detect DNA damages mainly comet scoring through CP software as whole comet, comet head and comet tail from image of previously studied single cell gel electrophoresis (SCGE) in the peripheral erythrocytes of fish exposed to benzene as experimental image. The results particularly on length and area of whole comet, head and tail were obtained after automated analysis in the CP software. The image processing study was done of the objects present in fluorescence microscopy image to know maximum DNA damage at each cell level for the fish erythrocytes. It was concluded that the present study of image based screening for DNA damages as details of comet and its head and tail evaluation by shape and area in the fish erythrocytes can be a suitable tool for genotoxicity prediction along with risk assessment at DNA level. The shape descriptor as Zernike moments order 0 to 9 can also be suitable parameters to know accuracy of the shape of comet and its head and tail in the image. Finally, high-throughput automated screening of comet test can help in disease diagnosis and repair mechanisms as well as environmental monitoring of genotoxin(s) within short period of time.

Promieniowanie ultrafioletowe (UV) jest fizycznym czynnikiem mutagennym i karcynogennym. Do powierzchni Ziemi dociera ok. 95% promieniowania ultrafioletowego A (UVA) (320–400 nm) i ok. 5% UVB (280–320 nm). Naturalnym czynnikiem ochronnym skóry przed UV jest melanina. Do raków skóry, których rozwój jest związany z długotrwałym narażeniem na promieniowanie UV, zalicza się niebarwnikowe raki skóry (non-melanoma skin cancers – NMSC) – raka podstawnokomórkowego (basal cell carcinoma – BCC) i raka kolczystokomórkowego (squamous cell carcinoma – SCC) oraz czerniaka złośliwego skóry (cutaneous malignant melanoma – CMM). Wysokie ryzyko rozwoju BCC związane jest z ostrą i wielokrotną ekspozycją na UV, powodującą oparzenia słoneczne. Badania molekularne BCC wykazały zaburzenia na szlaku regulacji sygnalizacji komórkowej sonic hedgehog (SHH), które były związane z mutacjami genu supresorowego PTCH1 (protein patched homolog 1). Ryzyko rozwoju BCC jest również związane z polimorfizmem genu receptora melanokortyny-1 (MC1R). Obserwowane w komórkach BCC mutacje genu P53 klasyfikowano jako wtórne zmiany genetyczne. W komórkach SCC mutacje indukowane UV najczęściej dotyczyły genu P53. W rozwoju SCC istotne znaczenie ma zwiększona ekspresja genu enzymu cyklooksygenazy-2 (COX-2). Innym czynnikiem patogenetycznym SCC jest nasilenie syntezy cytokin prozapalnych (czynnika martwicy nowotworu α (tumor necrosis factor α – TNF-α), interleukiny-1 α (IL-1α), IL-1β i IL-6). Obecnie w patogenezie CMM powszechnie uznawana jest rola UVB. W komórkach CMM notowano mutacje w genach: p16 INK4A (cyclin-dependent kinase inhibitor 2A INK4A), genie kodującym cyklinozależną kinazę 4 (cyclin-dependent kinase 4 – CDK4), Ras, PTEN (phosphatase and tensin homolog deleted on chromosome 10) i BRAF (proto-oncogene B-Raf). Mutacje genu BRAF stwierdzano w ok. 50% przypadków CMM. Mutacje genu P53 nie są charakterystyczne dla komórek CMM. Med. Pr. 2016;67(2):255–266
Ultraviolet radiation is a physical mutagenic and cancerogenic factor. About 95% of ultraviolet A (UVA) (320–400 nm) and 5% of UVB (280–320 nm) reach the Earth’s surface. Melanin is a natural skin protective factor against UV radiation. Skin cancers associated with long-term exposure to UV radiation are: basal cell carcinoma (BCC), squamous cell carcinoma (SCC) and cutaneous malignant melanoma (CMM). The high risk of BCC development is related to acute and repeated exposure to UV causing sunburn. Molecular studies of BBC demonstrated disorders in sonic hedgehog (SHH) cell signaling regulation pathway, associated with the suppressor protein patched homolog 1 gene (PTCH1) mutations. The risk of the BCC development is related to the polymorphism of melanokortin-1 receptor gene (MC1R). Tumor P53 gene mutations observed in BCC cells has been classified as secondary genetic changes. In SCC cells UV-induced mutations were mostly related to P53 gene. Increased expression of cyclooxigenase- 2 gene (COX-2) plays a significant role in the development of SCC. Other pathogenetic factors include intensification of the synthesis of pro-inflammatory cytokines (tumor necrosis factor α (TNF-α), interleukin-1 α (IL-1α), IL-1β and IL-6). Currently, the role of UVB has been recognized in the pathogenesis of CMM. In CMM cells the following gene mutations were noted: cyclindependent kinase inhibitor 2A INK4A (p16 INK4A), cyclin-dependent kinase 4 (CDK4), Ras, phosphatase and tensin homolog deleted on chromosome 10 (PTEN) and proto-oncogene B-Raf (BRAF). The BRAF gene mutations were observed in ~50% of CMM cases. Mutations of P53 gene are not characteristic of CMM cells. Med Pr 2016;67(2):255–266

Labile iron pool (LIP) constitutes a crossroad of metabolic pathways of iron-containing compounds and is midway between the cellular need for iron, its uptake and storage. In this study we investigated oxidative DNA damage in relation to the labile iron pool in a pair of mouse lymphoma L5178Y (LY) sublines (LY-R and LY-S) differing in sensitivity to hydrogen peroxide. The LY-R cells, which are hydrogen peroxide-sensitive, contain 3 times more labile iron than the hydrogen peroxide-resistant LY-S cells. Using the comet assay, we compared total DNA breakage in the studied cell lines treated with hydrogen peroxide (25 μM for 30 min at 4°C). More DNA damage was found in LY-R cells than in LY-S cells. We also compared the levels of DNA lesions sensitive to specific DNA repair enzymes in both cell lines treated with H2O2. The levels of endonuclease III-sensitive sites and Fapy-DNA glycosylase-sensitive sites were found to be higher in LY-R cells than in LY-S cells. Our data suggest that the sensitivity of LY-R cells to H2O2 is partially caused by the higher yield of oxidative DNA damage, as compared to that in LY-S cells. The critical factor appears to be the availability of transition metal ions that take part in the OH radical-generating Fenton reaction (very likely in the form of LIP).

Poly(ADP-ribose) polymerase (PARP-1) is an abundant nuclear protein with a high affinity for single- and double-strand DNA breaks. Its binding to strand breaks promotes catalysis of the covalent modification of nuclear proteins with poly(ADP-ribose) synthesised from NAD+. PARP-1-knockout cells are extremely sensitive to alkylating agents, suggesting the involvement of PARP-1 in base excision repair; however, its role remains unclear. We investigated the dependence of base excision repair pathways on PARP-1 and NAD+ using whole cell extracts derived from normal and PARP-1 deficient mouse cells and DNA substrates containing abasic sites. In normal extracts the rate of repair was highly dependent on NAD+. We found that in the absence of NAD+ repair was slowed down 4-6-fold after incision of the abasic site. We also established that in extracts from PARP-1 deficient mouse cells, repair of both regular and reduced abasic sites was increased with respect to normal extracts and was NAD+-independent, suggesting that in both short- and long-patch BER PARP-1 slows down, rather than stimulates, the repair reaction. Our data support the proposal that PARP-1 does not play a major role in catalysis of DNA damage processing via either base excision repair pathway.

Opis:
Effectiveness of green tea (compared to two single polyphenols) in removing ozone derived reactive oxygen species acting on dioleoylphosphatidylcholine monolayers was determined. Lipid oxidation was followed by changes in mechanical properties of the layer spread on the aqueous subphase containing various amounts of ozone in the presence and absence of polyphenolics. It was shown that the tea extract (containing 8.5 x 10-4 mg/cm3 polyphenols) is capable of inactivating 0.4 ppm ozone. The DPPH radical scavenging test set polyphenols in the order of increasing activity, consistent with their protective effect in relation to lipid oxidation, showing the highest efficacy of EGCG.

Phthalate are found in the environmental samples due to their wide use in the industry as plasticizers. Di-n-butyl phthalate (DBP) is mainly used in nitrocellulose and polyvinyl acetate products as well as in personal-care products. This study was performed to investigate the influence of exposure to DBP on the quantity and quality (motility, morphology) and DNA damage (induction of micronuclei and DNA strand breaks) of male mice gametes. The estimation of DBP residues was also done. Eight weeks exposure to DBP (500 mg/kg bw and 2000 mg/kg bw) did not significantly affect testes and epididymes weights as well as sperm count. DBP clearly diminished sperm motility, enhanced frequency of abnormal sperm heads and not significantly increased DNA strand breaks in germ cells as well as frequency of micronuclei in spermatids. There were no bioacumulation of DBP in mice. Results suggest that DBP may affect the male mice germ cells.
Ftalany są powszechnie wykorzystywane w przemyśle jako plastyfikatory, dlatego też często występują w próbach środowiskowych. Ftalan di-n-butylu (DBP) jest składnikiem produktów nitrocelulozowych i poliwinylowych, jak również produktów do higieny osobistej. Celem pracy było zbadanie wpływu podawania DBP na ilość i jakość gamet męskich myszy (ruchliwość, morfologia) oraz na występowanie uszkodzeń DNA (indukcja mikrojąder oraz pęknięć nici DNA). Zbadano także stężenie pozostałości DBP w gametach. Ośmiotygodniowe narażanie na DBP (500 mg/kg mc i 2000 mg/kg mc) nie powodowało istotnych zmian w ciężarze jąder i najądrzy ani w liczebności plemników. Narażenie na DBP, zwłaszcza w dawce 2000 mg/kg mc wpływało wyraźnie na zmniejszenie ruchliwości plemników i zwiększenie odsetka plemników o nieprawidłowej budowie morfologicznej. Powodowało też nieznaczne zwiększenie pęknięć nici DNA w gametach oraz zwiększenie częstości występowania mikrojąder w spermatydach. Nie wykazano bioakumulacji DBP w gonadach samców myszy. Uzyskane wyniki sugerują, że DBP może oddziaływać niekorzystnie na męskie komórki rozrodcze myszy.
