Opis:
Using the Kirkwood-Buff theory of solutions, the preferential solvation of the N-methylacet-amide (NMA), N-methylformamide, (NMF), and N,N-dimethylformamide, (DMF), molecule has been investigated in the binary {amide+methanol} mixtures at 313.15 K. Moreover, for the {amide+methanol} mixtures, where amide = NMF, DMF, and NMA, the molecular dynamics calculations at xamide=0.518 were performed. From the obtained molecule-molecule radial distribution functions, (rdf), and atom-atom rdf, it was possible to estimate the local mole fractions around the amide molecule, the orientation effects of molecules within the solvation shell, and a possibility of the formation of complexes. The general picture obtained from analysis of the molecular dynamics results is consistent with the deductions derived from thermodynamic data.

Opis:
W niniejszej pracy przedstawiono metodę cerometrycznego oznaczania amidów kwasów: akrylowego, malonowego, winowego i sulfanilowego. Opracowano również sposób zidentyfikowania kwasu mrówkowego w wodnej mieszaninie z kwasem nadchlorowym, nadchloranem cerowym i cerawym. Zaproponowana metoda jest stosunkowo dokładna, a jednocześnie prosta w stosowaniu i nie wymaga skomplikowanej aparatury.
Ceric perchlorate, introduced to chemical analysis by Smith and Getz [1], is increasingly applied to the titration of both inorganic and organic compounds [2-7]. Its high oxidizing potential (1.7 V) and the high stability of seric perchlorate solution [1] have made it possible to develop methods of titrating amides.

Opis:
Molecular models derived from results of quantum-chemical calculations present an important category of didactic instruments in chemistry education in upper secondary school and, particularly, at university. These models can be used especially as tools for supporting the students’ understanding by visual learning, which can adequately address complexity of many chemical topics, incorporate appropriate didactic principles, as well as utilize the benefits brought up by the actual information technology. The proposed molecular models are non-trivial examples of didactic application of computational chemistry techniques in illustration of electron interactions in amidic group, namely the interaction of the free electron pair on the nitrogen atom with the carbonyl group and also the interaction of atoms in the amide group with other surrounding atoms in the molecule. By these molecular models it is possible to explain acid-base properties of amides applying knowledge of electron density distribution in the molecules and the resulting electrostatic potential. Presentation of the structure and properties of the amides within education is important also for the reason that amidic functions are involved in many important natural substances (e.g. proteins, peptides, nucleic acids or alkaloids), synthetic macromolecular substances (e.g. Silon) or pharmaceutical preparations (e.g. paracetamol). Molecular models then serve to support better understanding of the structure of these substances and, in relation to it, their properties.

Reactions involving carbonyl groups are one of the most important transformations in organic chemistry. The nucleophilic properties of carbonyl compounds, when they are in the form of enolate ions, offer many possibilities for creating new carbon-carbon bonds in reactions with electrophiles. In the case of cyclic ketones, enolate formation can be stereocontrolled by deprotonation with the use of chiral lithium amides. Stereoselective formation of the chiral lithium enolate determines the stereochemistry of the product of the subsequent reaction with the electrophile. The induction of chirality in the reaction of enolate ions with electrophiles can also be achieved by using metals other than lithium, i.e. magnesium. When other alkali metals are used, an organometallic catalyst containing a chiral ligand must be present in the reaction. The presence of particular structural elements allow distinguishing the chiral lithium amides between eight major classes. Due to the high reactivity of lithium enolates, they are often converted into the silyl enol ether. This is done in two ways: internal quench (in situ reaction with TMSCl) or external quench. Due to aggregation of the chiral lithium amides and, thus, a decrease in asymmetric induction, the addition of LiCl is necessary for reactions run in external quench conditions. Although there are known examples of the use of chiral lithium amides in a catalytic amount in the deprotonation of epoxides, there is only one example of using less than stoichiometric amounts of chiral lithium amides in the deprotonation of ketones. There are many reports in the literature on the use of chiral lithium amides in total syntheses. The chiral lithium amides were used to form chiral enol silyl ether intermediates, e.g. in synthesis of chlortetaine or (+)-ibogamine. They are also used to form chiral lithium enolates which reacts directly with electrophiles, e.g. in synthesis of lasonolide A.

Opis:
Six hybrid N-ureidoethylamides of octapeptides in which an N-terminal cyclic structure related to enkephalin was elongated by a C-terminal fragment of deltorphin were synthesized on MBHA resin. The synthetic procedure involved deprotection of Boc groups with HCl/dioxane and cleavage of the peptide resin with 45 % TFA in DCM. d-Lys and d-Orn were incorporated in position 2, and Lys, Orn, Dab, or Dap in position 5. The side chains of the dibasic amino function were protected with the Fmoc group. This protection was removed by treatment with 55 % piperidine in DMF, and cyclization was achieved by treatment with bis-(4-nitrophenyl)carbonate. Using various combinations of dibasic amino acids, peptides containing a 17-, 18-, 19- or 20-membered ring structure were obtained. The peptides were tested in the guinea-pig ileum (GPI) and mouse vas deferens (MVD) assays. Diverse opioid activities were observed, depending on the size of the ring. Extension of the enkephalin sequence at the C-terminus by a deltorphin fragment resulted in a change of receptor selectivity in favor of the δ receptor. The conformational propensities of selected peptides were determined using the EDMC method in conjunction with data derived from NMR experiments carried out in water. This approach allowed proper examination of the dynamical behavior of these small peptides. The results were compared with those obtained earlier with corresponding N-(ureidoethyl)pentapeptide amides.

Opis:
A series of substituted amides of isocyclic and heterocyclic acids have been synthesized. All obtained compounds were evaluated qualitatively for their anticonvulsant activity in the maximal electroshock seizure test (MES test) and psychomotor seizure test (6 Hz test) in mice after intraperitoneal administration. The neurological toxicity was determined in the rotorod neurotoxicity test (Tox test). Two of these compounds appeared most promising: 3-oxo-1-cyklopentanecarboxylic acid 2-fluorobenzylamide (5), 1-cyclopentenecarboxylic acid 2-(trifluoromethyl)benzylamide (8) and were tested quantitatively in MES test in mice after oral administration (o.p.) and in 6 Hz test in mice after intraperitoneal administration(i.p.). Compound (5) showed MES ED50 =82.65 mg/kg and compound (8) showed 6 Hz ED50=84.13 mg/kg.

Potassium and Lithium N-chloramide-p-azobenzenosulphonate and sodium N-chloramide benzenosulphonate (synthesized at the Department of Organic Chemistry, Łódź University, under the direction of Prof. Dr. A. Chrząszczewska) were used to treat trichomonadosis of the vagina, after the elimination of extravaginal invasion foci. At the preceding Symposium the preliminary results were reported which point to strong trichomonacidal properties of these compounds in vitro. Totally, 143 women were treated by the administration of 0.05% solutions of the compounds and of globules in cocoa butter containg 0.05 g of the substance. For 15 days the treatment consisted in vaginal cleansings with fresh compound solutions (in boiled water) and application of l globule overnight; 123 (84.6%) patients were regarded as cured in whom the 3-month follow-up examination showed no T. vaginalis. The differences in the percentage of cures following the use of potassium and lithium N-chloramide-p-azobenzenosulphonate (80%) and sodium N-chloramide benzenosulphonate (p > 95%) were not statistically significant.
